    Resumo: Uma das preocupações quanto ao uso agrícola de lodo de esgoto é a lixiviação de ânions no perfil do solo, principalmente o nitrato, que podem atingir o lençol freático e contaminar as coleções hídricas após aplicações sucessivas do resíduo. O objetivo foi avaliar a concentração de nitrato, nitrito e sulfato no perfil do solo de 1 a 5 metros de profundidade após seis aplicações anuais de doses superestimadas de lodo de esgoto, seguidas de dez anos de cultivo sem adição do resíduo em poço escavado em uma área experimental. Observaram-se o acúmulo de sulfato até 2 metros de profundidade e as maiores concentrações de nitrato a 4 e 5 metros de profundidade, indicando maior movimentação deste ânion em relação aos demais. Abstract: One concern about using sewage sludge in agriculture is the leaching of anions through the soil profile, particularly nitrate, which could contaminate groundwater after successive residue applications. IN this study, anions concentrations were evaluated through the soil profile (1 to 5 m of depth) after six annual applications of superdoses of sewage sludge followed by 10 years of cultivation without adding the residue using a well excavated at an experimental area. Sulfate accumulation was observed until 2m of depth and mayor nitrate concentrations at 4 and 5m of depth, indicating more intense leaching of this anion

    Rapid diagnosis of coccidioidomycosis by nested PCR assay of sputum

    Coccidioidomycosis is a deep infection caused by two dimorphic fungi, Coccidioides immitis and Coccidioides posadasii. Diagnosis of the disease requires culture of suspicious clinical samples on mycological media. However, as these species are virulent pathogens, handling of their cultures is a high-risk activity, and is limited to Biosafety Level 3 laboratories.     Mineralização do nitrogênio de lodo de esgoto após aplicação no solo.

    Resumo: Lodos de esgoto podem ser reciclados em solo agrícola para fornecimento de nutrientes às plantas, principalmente nitrogênio (N), cuja concentração média nesse tipo de resíduo é de 2 a 4%. O presente trabalho teve como objetivo estudar a mineralização do nitrogênio aplicado no solo via doses de lodo de esgoto. O lodo de esgoto foi aplicado na área experimental da Embrapa Meio Ambiente entre os anos de 1999 e 2010, sendo a última aplicação em 2012. Foram testadas doses de lodo para fornecimento de 120, 240, 480 e 960 kg ha-1 de N, além de controle (0N). Tubos de PVC foram inseridos no solo (0 - 20 cm) e posteriormente retirados em duas épocas para quantificação do N inorgânico. No tempo inicial de avaliação os teores de amônio foram superiores aos de nitrato e após 45 dias estes se aproximaram. O amônio aumentou linearmente com a dose de lodo, enquanto o nitrato variou no tempo, com incremento de 407 mg kg-1 de N-NO3- em 45 dias. A taxa de mineralização do N do lodo nesse período foi igual a 92% do total de N aplicado, ou seja, houve disponibilidade excessiva de N em comparação com a demanda pelo milho

    The Influence of Dentin Age and the Presence of Cracks in Removal of the Root Filling Material

    Introduction: This study evaluated the removal of the filling material during endodontic retreatment considering the presence of cracks and the dentin age. Methods and Materials: A total of 20 freshly extracted single-rooted teeth were categorized into the following two groups according to the age of the patients: Group Young (Y; aged 18-30 years) and Group Old (O; aged ≥60 years). Each tooth specimen was scanned by microcomputed tomography (micro-CT) subsequently after endodontic retreatment with the Reciproc instruments (REC). The images were analyzed for differences in the volume of dentin cracks and the presence of the filling material in the middle and apical thirds of the teeth among the groups, according to the dentin age. Results: The micro-CT images showed that after retreatment, there were more cracks in the old root dentin than those in the young root dentin, although the difference was not statistically significant (P>0.05). The greatest reduction in the filling material was achieved when the old root dentin with cracks was retreated when compared with that of the young root dentin with cracks, but the difference was not statistically significant (P>0.05). Conclusion: The dentinal age and the presence of cracks were not found to be relevant factors for the removal of the filling material.Keywords: Dentin; Microcomputed Tomography; Retreatmen

    Low-level Laser Therapy to the Mouse Femur Enhances the Fungicidal Response of Neutrophils against Paracoccidioides brasiliensis

    Neutrophils (PMN) play a central role in host defense against the neglected fungal infection paracoccidioidomycosis (PCM), which is caused by the dimorphic fungus Paracoccidioides brasiliensis (Pb). PCM is of major importance, especially in Latin America, and its treatment relies on the use of antifungal drugs. However, the course of treatment is lengthy, leading to side effects and even development of fungal resistance. the goal of the study was to use low-level laser therapy (LLLT) to stimulate PMN to fight Pb in vivo. Swiss mice with subcutaneous air pouches were inoculated with a virulent strain of Pb or fungal cell wall components (Zymosan), and then received LLLT (780 nm; 50 mW; 12.5 J/cm2; 30 seconds per point, giving a total energy of 0.5 J per point) on alternate days at two points on each hind leg. the aim was to reach the bone marrow in the femur with light. Non-irradiated animals were used as controls. the number and viability of the PMN that migrated to the inoculation site was assessed, as well as their ability to synthesize proteins, produce reactive oxygen species (ROS) and their fungicidal activity. the highly pure PMN populations obtained after 10 days of infection were also subsequently cultured in the presence of Pb for trials of protein production, evaluation of mitochondrial activity, ROS production and quantification of viable fungi growth. PMN from mice that received LLLT were more active metabolically, had higher fungicidal activity against Pb in vivo and also in vitro. the kinetics of neutrophil protein production also correlated with a more activated state.
