    Analysis of flow cytometric aneuploid DNA histograms: validation of an automatic procedure against ad hoc experimental data

    In this paper we present an improved version of a method for the automatic analysis of flow cytometric DNA histograms from samples containing a mixture of two cell populations. The procedure is tested against two sets of ad hoc experimental data, obtained by mixing cultures of cell lines in different known proportions. The potentialities of the method are enlightened and discussed with regard to its capability of recovering the population percentages, the DNA index and the G0/G1, S, G2+M phase fractions of each population. On the basis of the obtained results, the procedure appears to be a promising tool in the flow cytometric data analysis and, in particular, in problems of diagnosis and prognosis of tumor diseases

    Data and performance of an active-set truncated Newton method with non-monotone line search for bound-constrained optimization

    In this data article, we report data and experiments related to the research article entitled “A Two-Stage Active-Set Algorithm for Bound-Constrained Optimization”, by Cristofari et al. (2017). The method proposed in Cristofari et al. (2017), tackles optimization problems with bound constraints by properly combining an active-set estimate with a truncated Newton strategy. Here, we report the detailed numerical experience performed over a commonly used test set, namely CUTEst (Gould et al., 2015). First, the algorithm ASA-BCP proposed in Cristofari et al. (2017) is compared with the related method NMBC (De Santis et al., 2012). Then, a comparison with the renowned methods ALGENCAN (Birgin and Martínez et al., 2002) and LANCELOT B (Gould et al., 2003) is reported

    Presence of multiple bacterial markers in clinical samples might be useful for presumptive diagnosis of infection in cirrhotic patients with culture-negative reports

    Bacterial infections in cirrhotic patients with ascites are associated with a severe prognosis and an increased risk of death. The microbiological standard tests for the diagnosis of suspected infection, based on culture test of blood and ascitic fluid, are, in many cases (30-40 %), negative, even when patients show symptoms of infection. A multiple culture-independent protocol was applied and evaluated as a diagnostic and prognostic tool for the detection of bacterial infection in cirrhotic patients. Sixty-four culture-negative samples obtained from 34 cirrhotic patients, with PMN < 250 cells/ÎŒl of ascitic fluid, were screened for the presence of bacterial DNA, endotoxin, peptidoglycan/ÎČ-glucan and microscopically visible bacterial cells. Correlations between the presence of multiple markers and various clinical and laboratory parameters were evaluated. Bacterial DNA was detected in 23 samples collected from 16 patients; a large part of these samples also showed the presence of other bacterial markers, which was associated with a worsening of liver functionality, a higher incidence of infections during the follow-up and a higher mortality rate in our cohort of cirrhotic patients. We believe that the detection of additional bacterial markers in bacterial DNA-positive clinical samples makes the bacterial presence and its clinical significance more realistic and might be useful as early markers of an ongoing bacterial infection and in establishing a clinical prognosis

    Hybridization of multi-objective deterministic particle swarm with derivative-free local searches

    The paper presents a multi-objective derivative-free and deterministic global/local hybrid algorithm for the efficient and effective solution of simulation-based design optimization (SBDO) problems. The objective is to show how the hybridization of two multi-objective derivative-free global and local algorithms achieves better performance than the separate use of the two algorithms in solving specific SBDO problems for hull-form design. The proposed method belongs to the class of memetic algorithms, where the global exploration capability of multi-objective deterministic particle swarm optimization is enriched by exploiting the local search accuracy of a derivative-free multi-objective line-search method. To the authors best knowledge, studies are still limited on memetic, multi-objective, deterministic, derivative-free, and evolutionary algorithms for an effective and efficient solution of SBDO for hull-form design. The proposed formulation manages global and local searches based on the hypervolume metric. The hybridization scheme uses two parameters to control the local search activation and the number of function calls used by the local algorithm. The most promising values of these parameters were identified using forty analytical tests representative of the SBDO problem of interest. The resulting hybrid algorithm was finally applied to two SBDO problems for hull-form design. For both analytical tests and SBDO problems, the hybrid method achieves better performance than its global and local counterparts

    A multi-objective DIRECT algorithm for ship hull optimization

    The paper is concerned with black-box nonlinear constrained multi-objective optimization problems. Our interest is the definition of a multi-objective deterministic partition-based algorithm. The main target of the proposed algorithm is the solution of a real ship hull optimization problem. To this purpose and in pursuit of an efficient method, we develop an hybrid algorithm by coupling a multi-objective DIRECT-type algorithm with an efficient derivative-free local algorithm. The results obtained on a set of “hard” nonlinear constrained multi-objective test problems show viability of the proposed approach. Results on a hull-form optimization of a high-speed catamaran (sailing in head waves in the North Pacific Ocean) are also presented. In order to consider a real ocean environment, stochastic sea state and speed are taken into account. The problem is formulated as a multi-objective optimization aimed at (i) the reduction of the expected value of the mean total resistance in irregular head waves, at variable speed and (ii) the increase of the ship operability, with respect to a set of motion-related constraints. We show that the hybrid method performs well also on this industrial problem

    High‐tech training for birds of prey

    Raptors are some of the most at‐risk groups of birds in the world and saving these top predators is essential for maintaining the health of many ecosystems. After hospitalization, raptors are often released when muscular recovery is still unfitting when they are unable to hunt efficiently and are at risk of dying from starvation within a few days. On the other hand, if a convalescent bird is trained with the only use of classic falconry techniques, it is likely to remain dependent on the caretaker/falconer even long after the release, so unable to hunt independently. To overcome these problems, a new training method was conceived, which could improve raptors’ muscular strength while limiting habituation to humans. This has been possible due to the combination of classic falconry techniques and modern technologies, such as the introduction of specific workouts with drones. Three falconry raptors and one wild Eurasian hobby were trained through high‐tech falconry to develop the ability to catch, grasp, and airlift their prey at a different speed, altitude, and resistance. The main findings of this study were: (i) The rapid increase of the raptors’ speed; (ii) the muscular growth and endurance, and (iii) successful reintroduction of a wild bird

    From perceived autonomy support to intentional behaviour: Testing an integrated model in three healthy-eating behaviours

    A motivational model integrating self-determination theory, the theory of planned behaviour, and the health action process approach was tested in three samples in three behavioural contexts: fruit and vegetable, breakfast, and snack consumption. Perceived support for autonomous (self-determined) forms of motivation from parents and autonomous motivation from self-determination theory were hypothesised to predict intention and behaviour indirectly via the mediation of attitude and perceived behavioural control from the theory of planned behaviour. It was also expected that planning strategies would mediate the effect of intention on behaviour. Relations in the proposed models were expected to be similar across the behaviours. A two-wave prospective design was adopted. Three samples of high-school students (total N = 1041; 59.60% female; M age = 17.13 years ± 1.57) completed measures of perceived autonomy support, autonomous motivation, theory of planned behaviour constructs, planning strategies and behaviour for each of the three behavioural contexts. Three months later, 816 participants (62,24% female; M age: 17.13 years, SD = 1.58) of the initial sample self-reported their behaviour referred to the previous three months. Structural equation models provided support for the key hypothesised effects of the proposed model for the three health-related behaviours. Two direct effects were significantly different across the three behaviours: the effect of perceived autonomy support on perceived behavioural control and the effect of attitude on intention. In addition, planning strategies mediated the effect of intention on behaviour in fruit and vegetable sample only. Findings extend knowledge of the processes by which psychological antecedents from the theories affect energy-balance related behaviours

    POINT CLOUD EXPLOITATION FOR STRUCTURAL MODELING AND ANALYSIS: A RELIABLE WORKFLOW

    none4noThe digitization and geometric knowledge of the historical built heritage is currently based on point cloud, that rarely or only partially is used as digital twin for structural analysis. The present work deals with historical artefacts survey, with particular reference to masonry structures, aimed to their structural analysis and assessment. In detail, the study proposes a methodology capable of employing semi-directly the original data obtained from the 3D digital survey for the generation of a Finite Element Model (FEM), used for structural analysis of masonry buildings. The methodology described presents a reliable workflow with twofold purpose: the improvement of the transformation process of the point cloud in solid and subsequently obtain a high-quality and detailed model for structural analyses. Through the application of the methodology to a case study, the method consistency was assessed, regarding the smoothness of the whole procedure and the dynamic characterization of the Finite Element Model. The main improvement in respect with similar or our previous workflows is obtained by the introduction of the retopology in data processing, allowing the transformation of the raw data into a solid model with optimal balancing between Level of Detail (LOD) and computational weight. Another significant aspect of the optimized process is undoubtedly the possibility of faithfully respecting the semantics of the structure, leading to the discretization of the model into different parts depending on the materials.     RESCUE MANAGEMENT AND ASSESSMENT OF STRUCTURAL DAMAGE BY UAV IN POST-SEISMIC EMERGENCY

    Abstract. The increasing frequency of emergencies urges the need for a detailed and thorough knowledge of the landscape. The first hours after a disaster are not only chaotic and problematic, but also decisive to successfully save lives and reduce damage to the building stock. One of the most important factors in any emergency response is to get an adequate awareness of the real situation, what is only possible after a thorough analysis of all the available information obtained through the Italian protocol Topography Applied to Rescue. To this purpose geomatic tools are perfectly suited to create, manage and dynamically enrich an organized archive of data to have a quick and functional access to information useful for several types of analysis, helping to develop solutions to manage the emergency and improving the success of rescue operations. Moreover, during an emergency like an earthquake, the conventional inspection to assess the damage status of buildings requires special tools and a lot of time. Therefore, given the large number of buildings requiring safety measures and rehabilitation, efficient use of limited resources such as time and equipment, as well as the safety of the involved personnel are important aspects. The applications shown in the paper are intended to underline how the above-mentioned objective, in particular the rehabilitation interventions of the built heritage, can be achieved through the use of data acquired from UAV platform integrated with geographic data stored in GIS platforms
