Depletion of the SR-related protein TbRRM1 leads to cell cycle arrest and apoptosis-like death in Trypanosoma brucei
Arginine-Serine (RS) domain-containing proteins are RNA binding proteins with multiple functions in RNA metabolism. In mammalian cells this group of proteins is also implicated in regulation and coordination of cell cycle and apoptosis. In trypanosomes, an early branching group within the eukaryotic lineage, this group of proteins is represented by 3 members, two of them are SR proteins and have been recently shown to be involved in rRNA processing as well as in pre-mRNA splicing and stability. Here we report our findings on the 3rd member, the SR-related protein TbRRM1. In the present study, we showed that TbRRM1 ablation by RNA-interference in T. brucei procyclic cells leads to cell-cycle block, abnormal cell elongation compatible with the nozzle phenotype and cell death by an apoptosis-like mechanism. Our results expand the role of the trypanosomal RS-domain containing proteins in key cellular processes such as cell cycle and apoptosis-like death, roles also carried out by the mammalian SR proteins, and thus suggesting a conserved function in this phylogenetically conserved protein family.Fil: Levy, Gabriela Vanesa. Exploiting electron storage in TiO2 nanoparticles for dark reduction of As(v) by accumulated electrons
UV irradiation of an ethanolic sol of TiO2 nanoparticles produces a trapped Ti(III) centre and a noticeable Burstein shift. Surface Morphology Evolution of Chemical Vapor-Deposited Tungsten Films on Si(100)
Chemical vapor deposition (CVD) is used to prepare research-grade heterostructures and to produce the majority of industrially important thin films.[1] In particular, CVD tungsten films are used for many technological applications.[2,3] In CVD an external source maintains a fixed concentration of reactant molecules at a distance above the film surface.[4] Then, gas diffusion drives the molecules through the diffusion layer[2] towards the film surface. At the film interface a reaction must occur before new material is incorporated into the solid. Kinetic studies show that two growth regimes are usually present in CVD. At a low deposition temperature (low rate, regime I) the kinetics is controlled by the surface reaction, whereas at a high temperature (high rate, regime II), mass transport of reactants to, or reaction products from, the surface is the rate-controlling step.Instituto de Investigaciones FisicoquĂmicas TeĂłricas y Aplicada
Effects of immunosuppressive drugs on COVID-19 severity in patients with autoimmune hepatitis
Background: We investigated associations between baseline use of immunosuppressive drugs and severity of Coronavirus Disease 2019 (COVID-19) in autoimmune hepatitis (AIH). Patients and methods: Data of AIH patients with laboratory confirmed COVID-19 were retrospectively collected from 15 countries. The outcomes of AIH patients who were on immunosuppression at the time of COVID-19 were compared to patients who were not on AIH medication. The clinical courses of COVID-19 were classified as (i)-no hospitalization, (ii)-hospitalization without oxygen supplementation, (iii)-hospitalization with oxygen supplementation by nasal cannula or mask, (iv)-intensive care unit (ICU) admission with non-invasive mechanical ventilation, (v)-ICU admission with invasive mechanical ventilation or (vi)-death and analysed using ordinal logistic regression. Results: We included 254 AIH patients (79.5%, female) with a median age of 50 (range, 17-85) years. At the onset of COVID-19, 234 patients (92.1%) were on treatment with glucocorticoids (n = 156), thiopurines (n = 151), mycophenolate mofetil (n = 22) or tacrolimus (n = 16), alone or in combinations. Overall, 94 (37%) patients were hospitalized and 18 (7.1%) patients died. Use of systemic glucocorticoids (adjusted odds ratio [aOR] 4.73, 95% CI 1.12-25.89) and thiopurines (aOR 4.78, 95% CI 1.33-23.50) for AIH was associated with worse COVID-19 severity, after adjusting for age-sex, comorbidities and presence of cirrhosis. Baseline treatment with mycophenolate mofetil (aOR 3.56, 95% CI 0.76-20.56) and tacrolimus (aOR 4.09, 95% CI 0.69-27.00) were also associated with more severe COVID-19 courses in a smaller subset of treated patients. Conclusion: Baseline treatment with systemic glucocorticoids or thiopurines prior to the onset of COVID-19 was significantly associated with COVID-19 severity in patients with AIH.Fil: Efe, Cumali. A Large-Scale Genetic Analysis Reveals a Strong Contribution of the HLA Class II Region to Giant Cell Arteritis Susceptibility
We conducted a large-scale genetic analysis on giant cell arteritis (GCA), a polygenic immune-mediated vasculitis. A case-control cohort, comprising 1,651 case subjects with GCA and 15,306 unrelated control subjects from six different countries of European ancestry, was genotyped by the Immunochip array. We also imputed HLA data with a previously validated imputation method to perform a more comprehensive analysis of this genomic region. The strongest association signals were observed in the HLA region, with rs477515 representing the highest peak (p = 4.05 Ă 10â40, OR = 1.73). A multivariate model including class II amino acids of HLA-DRÎČ1 and HLA-DQα1 and one class I amino acid of HLA-B explained most of the HLA association with GCA, consistent with previously reported associations of classical HLA alleles like HLA-DRB1â04. An omnibus test on polymorphic amino acid positions highlighted DRÎČ1 13 (p = 4.08 Ă 10â43) and HLA-DQα1 47 (p = 4.02 Ă 10â46), 56, and 76 (both p = 1.84 Ă 10â45) as relevant positions for disease susceptibility. Outside the HLA region, the most significant loci included PTPN22 (rs2476601, p = 1.73 Ă 10â6, OR = 1.38), LRRC32 (rs10160518, p = 4.39 Ă 10â6, OR = 1.20), and REL (rs115674477, p = 1.10 Ă 10â5, OR = 1.63). Our study provides evidence of a strong contribution of HLA class I and II molecules to susceptibility to GCA. In the non-HLA region, we confirmed a key role for the functional PTPN22 rs2476601 variant and proposed other putative risk loci for GCA involved in Th1, Th17, and Treg cell function

En esta tesis se analiza el rol de la credibilidad en la dinĂĄmica inflacionaria y su relevancia
para la estabilidad económica. A través de un enfoque teórico basado en el aprendizaje
adaptativo de los agentes econĂłmicos, se examinan casos de estudio para cuantificar el
impacto de la credibilidad en la inflaciĂłn. Los resultados revelan que una mayor credibilidad
en el gobierno, conduce a una menor inflaciĂłn. A su vez, se concluye que la credibilidad no
lo es todo, sino que importa también la velocidad del ajuste fiscal. Un ajuste mås råpido
debilita el rol de la credibilidad en la dinĂĄmica inflacionaria
