Long-term tolerability and efficacy after initial PegIFN-alpha addition to dasatinib in CML-CP : Five-year follow-up of the NordCML007 study
Objectives Treatment-free remission (TFR) has emerged as a treatment goal in chronic myeloid leukemia in the chronic phase (CML-CP). Attempts to increase proportion of patients achieving TFR include combination of tyrosine kinase inhibitors (TKI) and other drugs. Interferon-alpha in addition to TKI has shown promising efficacy but with dose-dependent toxicity and discontinuations. NordCML007 was initiated to study the efficacy and safety of low dose pegylated IFN-alpha (PegIFN-alpha) in combination with dasatinib (DAS) in CML-CP. Methods Forty patients with newly diagnosed CML-CP were given DAS upfront. After month 3 (M3) 15 mu g/wk of PegIFN-alpha was added and increased to 25 mu g/wk from M7 until M15. DAS treatment was continued and adverse events and BCR-ABL1 qRT-PCR values were reported yearly after M24. Results from M1 to M18 have previously been published, and here we present long-term data. Results After 5 years of follow-up, there were no suspected unexpected serious adverse reactions, no increase in serosal effusions, no disease progressions and no CML-related deaths. Rates of MR3.0 (MMR), MR4.0 and MR4.5 were 84.6%, 64.1% and 51.3% respectively at M60, and 95% of patients reached MMR at some point during the study. Conclusion Initial addition of PegIFN-alpha to DAS shows good long-term efficacy without increased toxicity.Peer reviewe
The 3rd DBCLS BioHackathon: improving life science data integration with Semantic Web technologies.
BACKGROUND: BioHackathon 2010 was the third in a series of meetings hosted by the Database Center for Life Sciences (DBCLS) in Tokyo, Japan. The overall goal of the BioHackathon series is to improve the quality and accessibility of life science research data on the Web by bringing together representatives from public databases, analytical tool providers, and cyber-infrastructure researchers to jointly tackle important challenges in the area of in silico biological research. RESULTS: The theme of BioHackathon 2010 was the 'Semantic Web', and all attendees gathered with the shared goal of producing Semantic Web data from their respective resources, and/or consuming or interacting those data using their tools and interfaces. We discussed on topics including guidelines for designing semantic data and interoperability of resources. We consequently developed tools and clients for analysis and visualization. CONCLUSION: We provide a meeting report from BioHackathon 2010, in which we describe the discussions, decisions, and breakthroughs made as we moved towards compliance with Semantic Web technologies - from source provider, through middleware, to the end-consumer.RIGHTS : This article is licensed under the BioMed Central licence at http://www.biomedcentral.com/about/license which is similar to the 'Creative Commons Attribution Licence'. Personal factors associated with health-related quality of life in persons with morbid obesity on treatment waiting lists in Norway
Purpose To explore relationships of socio-demographic
variables, health behaviours, environmental characteristics
and personal factors, with physical and mental health
variables in persons with morbid obesity, and to compare
their health-related quality of life (HRQoL) scores with
scores from the general population.
Methods A cross-sectional correlation study design was
used. Data were collected by self-reported questionnaire
from adult patients within the first 2 days of commencement
of a mandatory educational course. Of 185 course
attendees, 142 (76.8%) volunteered to participate in the
study. Valid responses on all items were recorded for 128
participants. HRQoL was measured with the Short Form
12v2 from which physical (PCS) and mental component
summary (MCS) scores were computed. Other standardized
instruments measured regular physical activity, social
support, self-esteem, sense of coherence, self-efficacy and
coping style.
Results Respondents scored lower on all the HRQoL subdomains
compared with norms. Linear regression analyses
showed that personal factors that included self-esteem,
self-efficacy, sense of coherence and coping style
explained 3.6% of the variance in PCS scores and 41.6% in
MCS scores.
Conclusion Personal factors such as self-esteem, sense of
coherence and a high approaching coping style are strongly
related to mental health in obese persons
High-accuracy chemical structure restoration from molecular descriptors and its application to chemical design
