    Auroral vector electric field and particle comparisons. 1: Pre-midnight convection topology

    Polar 3 was launched in northern Norway on January 27, 1974. Traversing nearly 3 deg latitude, the rocket crossed over a stable IBC II auroral arc in the positive bay region and continued north to a convection boundary which was identified as the Harang discontinuity. Measurement of the complete electric field vector, of energetic electrons and of the auroral N+2 and OI emissions were used to study the convection topology in the pre-magnetic-midnight region. A strong anticorrelation was observed between the electric field and the precipitating energetic electrons. The inverted V nature of the electron precipitations at the convection boundary, compared with the lack of such structure over the arc which was within the positive bay region, leads to the conclusion that auroral arcs are likely to be associated with inverted V type precipitation only at or poleward of convection boundaries and their eddy structures

    Robotic weld groove scanning for large tubular T-joints using a line laser sensor

    This paper presents a novel procedure for robotic scanning of weld grooves in large tubular T-joints. The procedure is designed to record the discrete weld groove scans using a commercially available line laser scanner which is attached to the robot end-effector. The advantage of the proposed algorithm is that it does not require any prior knowledge of the joint interface geometry, while only two initial scanning positions have to be specified. The position and orientation of the following scan are calculated using the data from two previous weld groove scans, so once initiated, the scanning process is fully autonomous. The procedure is a two-step algorithm consisting of the prediction and correction substeps, where the position and orientation of the sensor for the following scan are predicted and corrected. Such a procedure does not require frequent weld groove scanning for navigation along the groove. The performance of the proposed procedure is studied experimentally using an industrial-size T-joint specimen.     Impact of Rubin Observatory LSST Template Acquisition Strategies on Early Science from the Transients and Variable Stars Science Collaboration: Non-time-critical Science Cases

    Vera C. Rubin Observatory Legacy Survey of Space and Time, LSST, will revolutionize modern astronomy by producing an extremely deep (coadded depth ~27 mag) depth-limited survey of the entire southern sky (LSST Science Collaboration et al. 2009). The 8.4 m large-aperture, wide-field telescope, which is based in Cerro Pachón, will image the entire Southern sky every three nights in multiple bands (SDSS-u, g, r, i, z, y) and produce a fire-hose of data, 20 Tb each night, concluding in a 60 petabyte data set as the legacy of the 10 yr survey. Extracting meaningful light curves from variable objects requires difference imaging to both identify variability and calibrate light curve data products. Templates, co-added groups of visits that act as an image of the "static" sky, are a key component of Difference Imaging Analysis (DIA) and as such are of paramount importance for all science that involves variable objects. As the "non-time-critical" science cases discussed here are mostly periodic, they generally do not depend upon the survey alert stream; however, templates are still crucial for performing science and calibrations during the first year. We provide recommendations for observing strategies for template acquisition starting from commissioning and through Year 1 of the survey

    Photoelectrochemical synthesis of DNA microarrays

    Optical addressing of semiconductor electrodes represents a powerful technology that enables the independent and parallel control of a very large number of electrical phenomena at the solid-electrolyte interface. To date, it has been used in a wide range of applications including electrophoretic manipulation, biomolecule sensing, and stimulating networks of neurons. Here, we have adapted this approach for the parallel addressing of redox reactions, and report the construction of a DNA microarray synthesis platform based on semiconductor photoelectrochemistry (PEC). An amorphous silicon photoconductor is activated by an optical projection system to create virtual electrodes capable of electrochemically generating protons; these PEC-generated protons then cleave the acid-labile dimethoxytrityl protecting groups of DNA phosphoramidite synthesis reagents with the requisite spatial selectivity to generate DNA microarrays. Furthermore, a thin-film porous glass dramatically increases the amount of DNA synthesized per chip by over an order of magnitude versus uncoated glass.     First E region observations of mesoscale neutral wind interaction with auroral arcs

    We report the first observations of E region neutral wind fields and their interaction with auroral arcs at mesoscale spatial resolution during geomagnetically quiet conditions at Mawson, Antarctica. This was achieved by using a scanning Doppler imager, which can observe thermospheric neutral line-of-sight winds and temperatures simultaneously over a wide field of view. In two cases, the background E region wind field was perpendicular to an auroral arc, which when it appeared caused the wind direction within ∌50 km of the arc to rotate parallel along the arc, reverting to the background flow direction when the arc disappeared. This was observed under both westward and eastward plasma convection. The wind rotations occurred within 7–16 min. In one case, as an auroral arc propagated from the horizon toward the local zenith, the background E region wind field became significantly weaker but remained unaffected where the arc had not passed through. We demonstrate through modeling that these effects cannot be explained by height changes in the emission layer. The most likely explanation seems to be the greatly enhanced ion drag associated with the increased plasma density and localized ionospheric electric field associated with auroral arcs. In all cases, the F region neutral wind appeared less affected by the auroral arc, although its presence is clear in the data

    Every year, suicide is among the top 20 leading causes of death globally for all ages. Unfortunately, suicide is difficult to prevent, in large part because the prevalence of risk factors is high among the general population. In this review, clinical and psychological risk factors are examined and methods for suicide prevention are discussed. Prevention strategies found to be effective in suicide prevention include means restriction, responsible media coverage, and general public education, as well identification methods such as screening, gatekeeper training, and primary care physician education. Although the treatment for preventing suicide is difficult, follow-up that includes pharmacotherapy, psychotherapy, or both may be useful. However, prevention methods cannot be restricted to the individual. Community, social, and policy interventions will also be essentia

    Design of a combinatorial DNA microarray for protein-DNA interaction studies

    BACKGROUND: Discovery of precise specificity of transcription factors is an important step on the way to understanding the complex mechanisms of gene regulation in eukaryotes. Recently, double-stranded protein-binding microarrays were developed as a potentially scalable approach to tackle transcription factor binding site identification. RESULTS: Here we present an algorithmic approach to experimental design of a microarray that allows for testing full specificity of a transcription factor binding to all possible DNA binding sites of a given length, with optimally efficient use of the array. This design is universal, works for any factor that binds a sequence motif and is not species-specific. Furthermore, simulation results show that data produced with the designed arrays is easier to analyze and would result in more precise identification of binding sites. CONCLUSION: In this study, we present a design of a double stranded DNA microarray for protein-DNA interaction studies and show that our algorithm allows optimally efficient use of the arrays for this purpose. We believe such a design will prove useful for transcription factor binding site identification and other biological problems
