In vivo imaging of pyrrole-imidazole polyamides with positron emission tomography
The biodistribution profiles in mice of two pyrrole-imidazole polyamides were determined by PET. Pyrrole-imidazole polyamides are a class of small molecules that can be programmed to bind a broad repertoire of DNA sequences, disrupt transcription factor-DNA interfaces, and modulate gene expression pathways in cell culture experiments. The 18F-radiolabeled polyamides were prepared by oxime ligation between 4-[18F]-fluorobenzaldehyde and a hydroxylamine moiety at the polyamide C terminus. Small animal PET imaging of radiolabeled polyamides administered to mice revealed distinct differences in the biodistribution of a 5-ring β-linked polyamide versus an 8-ring hairpin, which exhibited better overall bioavailability. In vivo imaging of pyrrole-imidazole polyamides by PET is a minimum first step toward the translation of polyamide-based gene regulation from cell culture to small animal studies
Discovery of multiple Lorentzian components in the X-ray timing properties of the Narrow Line Seyfert 1 Ark 564
We present a power spectral analysis of a 100 ksec XMM-Newton observation of
the narrow line Seyfert 1 galaxy Ark~564. When combined with earlier RXTE and
ASCA observations, these data produce a power spectrum covering seven decades
of frequency which is well described by a power law with two very clear breaks.
This shape is unlike the power spectra of almost all other AGN observed so far,
which have only one detected break, and resemble Galactic binary systems in a
soft state. The power spectrum can also be well described by the sum of two
Lorentzian-shaped components, the one at higher frequencies having a hard
spectrum, similar to those seen in Galactic binary systems. Previously we have
demonstrated that the lag of the hard band variations relative to the soft band
in Ark 564 is dependent on variability time-scale, as seen in Galactic binary
sources. Here we show that the time-scale dependence of the lags can be
described well using the same two-Lorentzian model which describes the power
spectrum, assuming that each Lorentzian component has a distinct time lag. Thus
all X-ray timing evidence points strongly to two discrete, localised, regions
as the origin of most of the variability. Similar behaviour is seen in Galactic
X-ray binary systems in most states other than the soft state, i.e. in the
low-hard and intermediate/very high states. Given the very high accretion rate
of Ark 564 the closest analogy is with the very high (intermediate) state
rather than the low-hard state. We therefore strengthen the comparison between
AGN and Galactic binary sources beyond previous studies by extending it to the
previously poorly studied very high accretion rate regime.Comment: 11 pages, 11 figures, accepted for publication in MNRA
B and I-band optical micro-variability observations of the BL Lac objects S5 2007+777 and 3C371
We have observed S5 2007+777 and 3C371 in the B and I bands for 13 and 8
nights, respectively, during various observing runs in 2001, 2002 and 2004. The
observations resulted in almost evenly sampled light curves, 6-9 hours long. We
do not detect any flares within the observed light curves, but we do observe
small amplitude, significant variations, in both bands, on time scales of hours
and days. The average variability amplitude on time scales of minutes/hours is
2.5% and 1-1.5% in the case of S5 2007+777 and 3C371, respectively. The average
amplitudes increase to 5-12% and 4-6%, respectively, on time scales of days. We
find that the B and I band variations are highly correlated, on both short and
long time scales. During the 2004 observations, which resulted in the longest
light curves, we observe two well defined flux-decay and rising trends in the
light curves of both objects. When the flux decays, we observe significant
delays, with the B band flux decaying faster than the flux in the I band. As a
result, we also observe significant, flux related spectral variations as well.
The flux-spectral relation is rather complicated, with loop-like structures
forming during the flux evolution. The presence of spectral variations imply
that the observed variability is not caused by geometric effects. On the other
hand, our results are fully consistent with the hypothesis that the observed
variations are caused by perturbations which affect different regions in the

Correlated X-ray/Ultraviolet/Optical variability in the very low mass AGN NGC 4395
We report the results of a one year Swift X-ray/UV/optical programme
monitoring the dwarf Seyfert nucleus in NGC 4395 in 2008-2009. The UV/optical
flux from the nucleus was found to vary dramatically over the monitoring
period, with a similar pattern of variation in each of the observed UV/optical
bands (spanning 1900 - 5500 {\AA}). In particular, the luminosity of NGC 4395
in the 1900 {\AA} band changed by more than a factor of eight over the
monitoring period. The fractional variability was smaller in the UV/optical
bands than that seen in the X-rays, with the X-ray/optical ratio increasing
with increasing flux. Pseudo-instantaneous flux measurements in the X-ray and
each UV/optical band were well correlated, with cross correlation coefficients
of >0.7, significant at 99.9 per cent confidence. Archival Swift observations
from 2006 sample the intra-day X-ray/optical variability on NGC 4395. These
archival data show a very strong correlation between the X-ray and b bands,
with a cross-correlation coefficient of 0.84 (significant at >99 per cent
confidence). The peak in the cross correlation function is marginally resolved
and asymmetric, suggesting that X-rays lead the b band, but by 1 hour. In
response to recent (August 2011) very high X-ray flux levels from NGC4395 we
triggered Swift ToO observations, which sample the intra-hour X-ray/UV
variability. These observations indicate, albeit with large uncertainties, a
lag of the 1900 {\AA} band behind the X-ray flux of ~400 s. The tight
correlation between the X-ray and UV/optical lightcurves, together with the
constraints we place on lag time-scale are consistent with the UV/optical
variability of NGC 4395 being primarily due to reprocessing of X-ray photons by

Expanding the Repertoire of Natural Product-Inspired Ring Pairs for Molecular Recognition of DNA
A furan amino acid, inspired by the recently discovered proximicin natural products, was incorporated into the scaffold of a DNA-binding hairpin polyamide. While unpaired oligomers of 2,4-disubstituted furan amino acids show poor DNA-binding activity, furan (Fn) carboxamides paired with N-methylpyrrole (Py) and N-methylimidazole (Im) rings demonstrate excellent stabilization of duplex DNA as well as discrimination of noncognate sequences, consistent with function as a Py mimic according to the Py/Im polyamide pairing rules
Managing the complexity of doing it all : an exploratory study on students' experiences when trained stepwise in conducting consultations
Background: At most medical schools the components required to conduct a consultation, medical knowledge, communication, clinical reasoning and physical examination skills, are trained separately. Afterwards, all the knowledge and skills students acquired must be integrated into complete consultations, an art that lies at the heart of the medical profession. Inevitably, students experience conducting consultations as complex and challenging. Literature emphasizes the importance of three didactic course principles: moving from partial tasks to whole task learning, diminishing supervisors' support and gradually increasing students' responsibility. This study explores students' experiences of an integrated consultation course using these three didactic principles to support them in this difficult task.
Methods: Six focus groups were conducted with 20 pre-clerkship and 19 clerkship students in total. Discussions were audiotaped, transcribed and analysed by Nvivo using the constant comparative strategy within a thematic analysis.
Results: Conducting complete consultations motivated students in their learning process as future physician. Initially, students were very much focused on medical problem solving. Completing the whole task of a consultation obligated them to transfer their theoretical medical knowledge into applicable clinical knowledge on the spot. Furthermore, diminishing the support of a supervisor triggered students to reflect on their own actions but contrasted with their increased appreciation of critical feedback. Increasing students' responsibility stimulated their active learning but made some students feel overloaded. These students were anxious to miss patient information or not being able to take the right decisions or to answer patients' questions, which sometimes resulted in evasive coping techniques, such as talking faster to prevent the patient asking questions.
Conclusion: The complex task of conducting complete consultations should be implemented early within medical curricula because students need time to organize their medical knowledge into applicable clinical knowledge. An integrated consultation course should comprise a step-by-step teaching strategy with a variety of supervisors' feedback modi, adapted to students' competence. Finally, students should be guided in formulating achievable standards to prevent them from feeling overloaded in practicing complete consultations with simulated or real patients
