    Performance of leaf wetness sensor used in winter wheat disease management

    Wetness on crop leaves has particular epidemiological significance because many fungal diseases affect plants only when free moisture is present on leaves. The leaf wetness sensor detects the presence of wetness on a leaf’s surface, enabling researchers and producers to forecast disease and protect plant canopies, and consequently to optimize fungicide application and often reduce environmental load. This research project aimed at better understanding the leaf wetness duration and its influence in winter wheat disease. Measurement of surface wetness duration by three electronic flat-plate sensors (Model 237-Campbell Scientific, Inc) in wheat fields were compared with tactile and visual observations in replicated field experiments at the site of Arlon (Belgium) during the period May-July 2006 and April-July 2007. Performances of the sensor were evaluated against SWEB model outputs and visual observations of disease symptoms. On the field, dew-onset and dry-off of wetness on leaves were observed visually (with a flash light for dew-onset) at 15-minute intervals. Each sensor was placed close the flag leaf. For the three sensors, the two dew-onset and dry-off times measured in both 2006 and 2007 crop seasons gave a leaf wetness duration (LWD) which was on average one hour less than visual observations. In order to establish a relationship between the surface wetness periods and wheat foliar diseases, LWD was compared with the Septoria leaf blotch (SLB) development risk (main winter wheat disease).     Rouille brune du blé, un modèle pour évaluer les risques

De 2000 à 2003, cette maladie apparaissait à la fin de l’épiaison, mais depuis 2003, elle apparaît de plus en plus tôt (GS45 stade gonflement). Cette apparition précoce est probablement liée à des températures printanières supérieures par rapport à la normale 1971-2000. Une analyse des données météorologiques nocturnes et des données d’observation de la maladie sur quatre sites expérimentaux (Everlange, Christnach, Burmerange et Reuler) entre 2000 et 2003 a révélé une forte corrélation positive entre la prédiction de la maladie basée sur le critère d’au moins 12 heures consécutives avec une température comprise entre 8 et 16°C et une humidité supérieure à 60% et la maladie observée sur la F1 (R = 0.93 ; P < 0.05) et sur la F2 (R = 0.87 ; P < 0.05). Les sorties de ce modèle qui a été développé sur base d’une approche stochastique ont été utilisées dans les bulletins d’avertissements diffusés conjointement par le Centre de Recherche Public – Gabriel Lippmann et l’Université de Liège-Campus d’Arlon à partir de 2004. La mise en application de ce modèle a montré un taux de réussite oscillant entre 80 et 85% pour la simulation de la rouille brune au G.-D. de Luxembourg. L’effort se poursuit pour spatialiser les sorties du modèle sur tout le territoire luxembourgeois et faciliter son utilisation par tous les vulgarisateurs agricoles

    Environmental and effluent monitoring at ANSTO sites, 2001.

    Results are presented of environmental surveillance and effluent monitoring conducted in the calendar year 2001 at the two sites owned and operated by ANSTO at the Lucas Heights Science and Technology Centre (LHSTC) and the National Medical Cyclotron (NMC). All low-level liquid and gaseous effluent discharges complied with existing discharge authorisations and relevant environmental regulations. Potential effective doses to the general public from LHSTC-controlled airborne discharges were estimated for 2001 using the PC-Cream atmospheric dispersion and dosimetry code. The potential effective doses to the public in 2001 were estimated to be less than 0.01 mSv/year for all receptor locations on the LHSTC 1.6 km buffer zone boundary or beyond. This is well below the ALARA objective of 0.02 mSv per year for off-site doses that ANSTO has set and much lower than the public dose limit of 1 mSv per year (above natural background and medical doses) and the natural background dose in Australia of 1.5 mSv per year (Webb et al; 1999). It is concluded that there is no impact on the health of the community as a consequence of operations at the Lucas Heights Science and Technology Centre or the National Medical Cyclotron

    Recombinase polymerase amplification assay for rapid detection of lumpy skin disease virus

    Background&nbsp; Lumpy skin disease virus (LSDV) is aCapripoxvirusinfecting cattle and Buffalos. Lumpy skin disease (LSD) leads to significant economic losses due to hide damage, reduction of milk production, mastitis, infertility and mortalities (10 %). Early detection of the virus is crucial to start appropriate outbreak control measures. Veterinarians rely on the presence of the characteristic clinical signs of LSD. Laboratory diagnostics including virus isolation, sequencing and real-time polymerase chain reaction (PCR) are performed at well-equipped laboratories. In this study, a portable, simple, and rapid recombinase polymerase amplification (RPA) assay for the detection of LSDV-genome for the use on farms was developed.&nbsp; Results&nbsp; The LSDV RPA assay was performed at 42 &deg;C and detected down to 179 DNA copies/reaction in a maximum of 15 min. Unspecific amplification was observed with neither LSDV-negative samples (n= 12) nor nucleic acid preparations from orf virus, bovine papular stomatitis virus, cowpoxvirus, Peste des petits ruminants and Blue tongue virus (serotypes 1, 6 and 8). The clinical sensitivity of the LSDV RPA assay matched 100 % (n= 22) to real-time PCR results. In addition, the LSDV RPA assay detected sheep and goat poxviruses.&nbsp; Conclusion&nbsp; The LSDV RPA assay is a rapid and sensitive test that could be implemented in field or at quarantine stations for the identification of LSDV infected case

The Authors would also like to acknowledge funding from The MRC – United Kingdom, Grant G1100021.Pulsed electron-electron double resonance (PELDOR) coupled with site-directed spin labeling is a powerful technique for the elucidation of protein or nucleic acid, macromolecular structure and interactions. The intrinsic high sensitivity of electron paramagnetic resonance enables measurement on small quantities of bio-macromolecules, however short relaxation times impose a limit on the sensitivity and size of distances that can be measured using this technique. The persistence of the electron spin-echo, in the PELDOR experiment, is one of the most crucial limitations to distance measurement. At a temperature of around 50 K one of the predominant factors affecting persistence of an echo, and as such, the sensitivity and measurable distance between spin labels, is the electron spin echo dephasing time (Tm). It has become normal practice to use deuterated solvents to extend Tm and recently it has been demonstrated that deuteration of the underlying protein significantly extends Tm. Here we examine the spatial effect of segmental deuteration of the underlying protein, and also explore the concentration and temperature dependence of highly deuterated systems.Publisher PDFPeer reviewe

    Hydrological impacts of climate change at catchment scale : a case study in the Grand-Duchy of Luxembourg

    As a consequence of an increase of days with westerly atmospheric fluxes, bringing humid air masses from the Atlantic Ocean to Western Europe, important changes in the annual and seasonal distribution of rainfall have been observed over the past 150 years. Annual rainfall totals observed during the second half of the 19th century were less important than those observed during the second half of the 20th century. Moreover, during the past 50 years winter rainfall totals have significantly increased, while summer rainfall totals have been decreasing. Streamflow observations through the second half of the 20th century have shown a significant increase of winter maximum daily streamflow, in reaction to the winter rainfall increase. The modelling of the streamflow under the 19th century climatological conditions suggests that since then, the number of winter flood days has increased, while the occurrence of summer flood days has decreased. Moreover, high floods appear to have been more frequent in the second half of the 20th century

    This report presents the results of environmental and effluent monitoring at the Lucas Heights Science and Technology Centre (LHSTC) and the National Medical Cyclotron (NMC) from January 2002 to June 2003. Potential effective dose rates to the general public from airborne discharges from the LHSTC site were less than 0.01 mSv/year, well below the 1 mSv/year dose rate limit for long term exposure that is recommended by the Australian National Occupational Health and Safety Commission. The effective dose rates to hypothetical individuals potentially exposed to radiation in routine liquid effluent discharges from the LHSTC were recently calculated to be less than 0.001 mSv/year. This is much less than dose rates estimated for members of public potentially exposed to airborne emissions. The levels of tritium detected in groundwater and stormwater at the LHSTC were less than the Australian drinking water guidelines. The airborne and liquid effluent emissions from the NMC were below the ARPANSA-approved notification levels and NSW EPA limits, respectively. ANSTO's routine operations at the LHSTC and the NMC make only a very small addition to the natural background radiation dose experienced by members of the Australian public

    This report presents the results of ANSTO’s environmental and effluent monitoring at the Lucas Heights Science and Technology Centre (LHSTC) and the National Medical Cyclotron (NMC) sites, from July 2006 to June 2007. Estimated effective doses to the members of the public potentially affected by routine airborne emissions from the LHSTC were lower than in previous years due to the closure of the HIFAR research reactor. The maximum potential off-site dose of 0.002 mSv/year was 10% of the As Low As Reasonably Achievable (ALARA) objective of 0.02 mSv/year, and much lower than the public dose limit of 1 mSv/year that is recommended by the Australian Radiation Protection and Nuclear Safety Agency (ARPANSA). The effective doses to the critical group potentially exposed to routine liquid effluent releases from the LHSTC have been realistically assessed at less than 25% of the estimated doses to the critical group for airborne releases. Based on releases of liquid effluent from the LHSTC during 2006-07, a radiological risk assessment was conducted for marine biota in the receiving environment at Potter Point. The possible dose-rates to various species of marine biota were evaluated against international criteria recommended for the protection of biota from radiological hazards. In all cases it was concluded that the radiological risk to marine biota from ANSTO’s effluent releases was negligible. The median tritium concentrations detected in groundwater and surface waters at the LHSTC were typically less than 2% of those set out in the Australian Drinking Water Guidelines. The airborne emissions from the NMC were below the ARPANSA-approved notification levels. Results of environmental monitoring at both ANSTO sites confirm that the facilities continue to be operated well within regulatory limits. ANSTO’s routine operations at the LHSTC and NMC make only a very small addition to the natural background radiation dose of approximately 1.5 mSv/year experienced by members of the Australian public
