    Crystal Nucleation by Laser-Induced Cavitation\ud

    High-speed and high-resolution photography have been used to investigate the relationship between creation, expansion, and collapse of a vapor cavity induced by a 6 ns laser pulse and the subsequent nucleation of crystals. A thin layer of supersaturated aqueous solutions of (NH4)2SO4 and KMnO4 was confined between two glass plates with a separation of 50 and 100 μm. The expansion and collapse of the laser-induced vapor bubble occurred over a total time scale of 200 μs, while the first identifiable crystal appears one second after the laser pulse. Crystals were observed to form on a ring with a diameter of 70 μm centered in the focal point of the laser. The ring is preceded by an optical disturbance observed through the cavity around 30–50 μs after the laser pulse and vapor cavity formation. This ring-shaped optical disturbance originates from changes in refractive index induced by crystal nuclei formation. The formation of the nuclei most probably coincides with the formation of the bubble, when the rate of evaporation and the supersaturation are at their maxima.     Infection of Chlorella NC64A cells by PBCV-1 produces a rapid depolarization of the host probably by incorporation of a viral-encoded K+ channel (Kcv) into the host membrane. To examine the effect of an elevated conductance, we monitored the virus-stimulated efflux of K+ from the Chlorella cells. The results indicate that all 8 Chlorella viruses tested evoked a host specific K+ efflux with a concomitant decrease in the intracellular K+. This K+ efflux is partially reduced by blockers of the Kcv channel. Qualitatively these results support the hypothesis that depolarization and K+ efflux are at least partially mediated by Kcv. The virus-triggered K+ efflux occurs in the same time frame as host cell wall degradation and ejection of viral DNA. Therefore, it is reasonable to postulate that loss of K+ and associated water fluxes from the host lower the pressure barrier to aid ejection of DNA from the virus particles into the host

    Um entlang der deutsch-tschechischen Grenze im Falle von Störfällen grenznaher Industriebetriebe auf tschechischer Seite umweltmesstechnische Untersuchungen durchführen zu können, recherchierte und bewertete das Labor für Physik und Umweltmesstechnik der Hochschule Düsseldorf die technischen Möglichkeiten der unbemannten Immissionsmessung. Im Rahmen der Auswertung wurden umfangreiche Hinweise zur Auswahl eines adäquaten Flug- und Messsystems gegeben. Der Fokus lag darauf, den Gefahrenabwehrkräften vor Ort Messtechnik empfehlen zu können, mit der diese dann an repräsentativen Orten in Grenznähe möglichst schnell und ausreichend aussagekräftig überprüfen können, ob es zu einer Schadstoffeinwirkung kommen kann bzw. bereits gekommen ist.     Challenges in producing policy-relevant global scenarios of biodiversity and ecosystem services

    Scenario-based modelling is a powerful tool to describe relationships between plausible trajectories of drivers, possible policy interventions, and impacts on biodiversity and ecosystem services. Model inter-comparisons are key in quantifying uncertainties and identifying avenues for model improvement but have been missing among the global biodiversity and ecosystem services modelling communities. The biodiversity and ecosystem services scenario-based inter-model comparison (BES-SIM) aims to fill this gap. We used global land-use and climate projections to simulate possible future impacts on terrestrial biodiversity and ecosystem services using a variety of models and a range of harmonized metrics. The goal of this paper is to reflect on the steps taken in BES-SIM, identify remaining methodological challenges, and suggest pathways for improvement. We identified five major groups of challenges; the need to: 1) better account for the role of nature in future human development storylines; 2) improve the representation of drivers in the scenarios by increasing the resolution (temporal, spatial and thematic) of land-use as key driver of biodiversity change and including additional relevant drivers; 3) explicitly integrate species- and trait-level biodiversity in ecosystem services models; 4) expand the coverage of the multiple dimensions of biodiversity and ecosystem services; and finally, 5) incorporate time-series or one-off historical data in the calibration and validation of biodiversity and ecosystem services models. Addressing these challenges would allow the development of more integrated global projections of biodiversity and ecosystem services, thereby improving their policy relevance in supporting the interlinked international conservation and sustainable development agendas

    Comparing future patterns of energy system change in 2°C scenarios with historically observed rates of change

    This paper systematically compares modeled rates of change provided by global integrated assessment models aiming for the 2 °C objective to historically observed rates of change. Such a comparison can provide insights into the difficulty of achieving such stringent climate stabilization scenarios. The analysis focuses specifically on the rates of change for technology expansion and diffusion, emissions and energy supply investments. The associated indicators vary in terms of system focus (technology-specific or energy system wide), temporal scale (timescale or lifetime), spatial scale (regional or global) and normalization (accounting for entire system growth or not). Although none of the indicators provide conclusive insights as to the achievability of scenarios, this study finds that indicators that look into absolute change remain within the range of historical growth frontiers for the next decade, but increase to unprecedented levels before mid-century. Indicators that take into account or normalize for overall system growth find future change to be broadly within historical ranges. This is particularly the case for monetary-based normalization metrics like GDP compared to energy-based normalization metrics like primary energy. By applying a diverse set of indicators alternative, complementary insights into how scenarios compare with historical observations are acquired but they do not provide further insights on the possibility of achieving rates of change that are beyond current day practice

    Fine Mapping of the 1p36 Deletion Syndrome Identifies Mutation of PRDM16 as a Cause of Cardiomyopathy

    Deletion 1p36 syndrome is recognized as the most common terminal deletion syndrome. Here, we describe the loss of a gene within the deletion that is responsible for the cardiomyopathy associated with monosomy 1p36, and we confirm its role in nonsyndromic left ventricular noncompaction cardiomyopathy (LVNC) and dilated cardiomyopathy (DCM). With our own data and publically available data from array comparative genomic hybridization (aCGH), we identified a minimal deletion for the cardiomyopathy associated with 1p36del syndrome that included only the terminal 14 exons of the transcription factor PRDM16 (PR domain containing 16), a gene that had previously been shown to direct brown fat determination and differentiation. Resequencing of PRDM16 in a cohort of 75 nonsyndromic individuals with LVNC detected three mutations, including one truncation mutant, one frameshift null mutation, and a single missense mutant. In addition, in a series of cardiac biopsies from 131 individuals with DCM, we found 5 individuals with 4 previously unreported nonsynonymous variants in the coding region of PRDM16. None of the PRDM16 mutations identified were observed in more than 6,400 controls. PRDM16 has not previously been associated with cardiac disease but is localized in the nuclei of cardiomyocytes throughout murine and human development and in the adult heart. Modeling of PRDM16 haploinsufficiency and a human truncation mutant in zebrafish resulted in both contractile dysfunction and partial uncoupling of cardiomyocytes and also revealed evidence of impaired cardiomyocyte proliferative capacity. In conclusion, mutation of PRDM16 causes the cardiomyopathy in 1p36 deletion syndrome as well as a proportion of nonsyndromic LVNC and DCM

    Nitrogen infusion R&D at DESY a case study on cavity cut-outs

    A first series of nitrogen infusion runs of 1.3 GHz single-cell cavities at DESY resulted in an unexpected and severe deterioration observed during the vertical cold test. To investigate the origin of the deterioration, one of the cavities underwent extensive radio-frequency measurements and a temperature- and magnetic field-mapping was performed in collaboration with the Helmholtz-Zentrum Berlin. After combining all results, regions of interests were identified and cut-out of the cavity. Subsequent surface analysis techniques (EBSD, PALS, PIXE, SEM/EDX, SIMS, XPS) were applied in order to identify the microscopic origin of the deterioration and especially the differences between hot and cold spots as well as quench spots. An excess of niobium carbides, reducing the thermal conductivity, was identified as the probable cause for the deterioration, and the size- and density-distributions were observed to be crucial for the resulting performance reduction.
