Spectral X-ray CT for fast NDT using discrete tomography
We present progress in fast, high-resolution imaging, material classification, and fault detection using
hyperspectral X-ray measurements. Classical X-ray CT approaches rely on data from many projection
angles, resulting in long acquisition and reconstruction times. Additionally, conventional CT cannot
distinguish between materials with similar densities. However, in additive manufacturing, the majority of
materials used are known a priori. This knowledge allows to vastly reduce the data collected and increase
the accuracy of fault detection. In this context, we propose an imaging method for non-destructive testing
of materials based on the combination of spectral X-ray CT and discrete tomography. We explore the
use of spectral X-ray attenuation models and measurements to recover the characteristic functions of
materials in heterogeneous media with piece-wise uniform composition. We show by means of numerical
simulation that using spectral measurements from a small number of angles, our approach can alleviate
the typical deterioration of spatial resolution and the appearance of streaking artifacts.Mechanical Engineerin
Structural insights into the autoregulation and cooperativity of the human transcription factor Ets-2
Ets-2, like its closely related homologue Ets-1, is a member of the Ets family of DNA binding transcription factors. Both proteins are subject to multiple levels of regulation of their DNA binding and transactivation properties. One such regulatory mechanism is the presence of an autoinhibitory module, which in Ets-1 allosterically inhibits the DNA binding activity. This inhibition can be relieved by interaction with protein partners or cooperative binding to closely separated Ets binding sites in a palindromic arrangement. In this study we describe the 2.5 Å resolution crystal structure of a DNA complex of the Ets-2 Ets domain. The Ets domain crystallized with two distinct species in the asymmetric unit, which closely resemble the autoinhibited and DNA bound forms of Ets-1. This discovery prompted us to re-evaluate the current model for the autoinhibitory mechanism and the structural basis for cooperative DNA binding. In contrast to Ets-1, in which the autoinhibition is caused by a combination of allosteric and steric mechanisms, we were unable to find clear evidence for the allosteric mechanism in Ets-2. We also demonstrated two possibly distinct types of cooperative binding to substrates with Ets binding motifs separated by four and six base pairs and suggest possible molecular mechanisms for this behavior
Laser-generated ultrasound with optical fibres using functionalised carbon nanotube composite coatings
Optical ultrasound transducers were created by coating optical fibres with a composite of carbon nanotubes (CNTs) and polydimethylsiloxane (PDMS). Dissolution of CNTs in PDMS to create the composite was facilitated by functionalisation with oleylamine. Composite surfaces were applied to optical fibres using dip coating. Under pulsed laser excitation, ultrasound pressures of 3.6 MPa and 4.5 MPa at the coated end faces were achieved with optical fibre core diameters of 105 and 200 μm, respectively. The results indicate that CNT-PDMS composite coatings on optical fibres could be viable alternatives to electrical ultrasound transducers in miniature ultrasound imaging probes

Regularity issues in the problem of fluid structure interaction
We investigate the evolution of rigid bodies in a viscous incompressible
fluid. The flow is governed by the 2D Navier-Stokes equations, set in a bounded
domain with Dirichlet boundary conditions. The boundaries of the solids and the
domain have H\"older regularity , . First, we
show the existence and uniqueness of strong solutions up to collision. A key
ingredient is a BMO bound on the velocity gradient, which substitutes to the
standard estimate for smoother domains. Then, we study the asymptotic
behaviour of one body falling over a flat surface. We show that
collision is possible in finite time if and only if
Reverberation Mapping of the Kepler-Field AGN KA1858+4850
KA1858+4850 is a narrow-line Seyfert 1 galaxy at redshift 0.078 and is among
the brightest active galaxies monitored by the Kepler mission. We have carried
out a reverberation mapping campaign designed to measure the broad-line region
size and estimate the mass of the black hole in this galaxy. We obtained 74
epochs of spectroscopic data using the Kast Spectrograph at the Lick 3-m
telescope from February to November of 2012, and obtained complementary V-band
images from five other ground-based telescopes. We measured the H-beta light
curve lag with respect to the V-band continuum light curve using both
cross-correlation techniques (CCF) and continuum light curve variability
modeling with the JAVELIN method, and found rest-frame lags of lag_CCF = 13.53
(+2.03, -2.32) days and lag_JAVELIN = 13.15 (+1.08, -1.00) days. The H-beta
root-mean-square line profile has a width of sigma_line = 770 +/- 49 km/s.
Combining these two results and assuming a virial scale factor of f = 5.13, we
obtained a virial estimate of M_BH = 8.06 (+1.59, -1.72) x 10^6 M_sun for the
mass of the central black hole and an Eddington ratio of L/L_Edd ~ 0.2. We also
obtained consistent but slightly shorter emission-line lags with respect to the
Kepler light curve. Thanks to the Kepler mission, the light curve of
KA1858+4850 has among the highest cadences and signal-to-noise ratios ever
measured for an active galactic nucleus; thus, our black hole mass measurement
will serve as a reference point for relations between black hole mass and
continuum variability characteristics in active galactic nuclei
Stage-specific proteomes from onchocerca ochengi, sister species of the human river blindness parasite, uncover adaptations to a nodular lifestyle
Despite 40 years of control efforts, onchocerciasis (river blindness) remains one of the most important neglected tropical diseases, with 17 million people affected. The etiological agent, Onchocerca volvulus, is a filarial nematode with a complex lifecycle involving several distinct stages in the definitive host and blackfly vector. The challenges of obtaining sufficient material have prevented high-throughput studies and the development of novel strategies for disease control and diagnosis. Here, we utilize the closest relative of O. volvulus, the bovine parasite Onchocerca ochengi, to compare stage-specific proteomes and host-parasite interactions within the secretome. We identified a total of 4260 unique O. ochengi proteins from adult males and females, infective larvae, intrauterine microfilariae, and fluid from intradermal nodules. In addition, 135 proteins were detected from the obligate Wolbachia symbiont. Observed protein families that were enriched in all whole body extracts relative to the complete search database included immunoglobulin-domain proteins, whereas redox and detoxification enzymes and proteins involved in intracellular transport displayed stage-specific overrepresentation. Unexpectedly, the larval stages exhibited enrichment for several mitochondrial-related protein families, including members of peptidase family M16 and proteins which mediate mitochondrial fission and fusion. Quantification of proteins across the lifecycle using the Hi-3 approach supported these qualitative analyses. In nodule fluid, we identified 94 O. ochengi secreted proteins, including homologs of transforming growth factor-β and a second member of a novel 6-ShK toxin domain family, which was originally described from a model filarial nematode (Litomosoides sigmodontis). Strikingly, the 498 bovine proteins identified in nodule fluid were strongly dominated by antimicrobial proteins, especially cathelicidins. This first high-throughput analysis of an Onchocerca spp. proteome across the lifecycle highlights its profound complexity and emphasizes the extremely close relationship between O. ochengi and O. volvulus The insights presented here provide new candidates for vaccine development, drug targeting and diagnostic biomarkers
