    Research Note: Effect of Light Quality on Fruit Growth, Composition and the Sensory Impact of the Wines

    The stage at which grapes are harvested has an influence on the aromatic and phenolic composition of the berries and the resulting wines. The aim of this study was to evaluate wines harvested sequentially as outlined in the berry sugar accumulation model. Two vintages and treatments in which the light qualityand quantity were altered at the fruit zone were compared. In 2010/2011, the grapes were harvested at two ripening stages after the sugar loading plateau was reached, namely the “fresh fruit” stage (20-25 days afterwards) and “pre-mature” stage (at approximately 35 days). In the 2011/2012 season, grapes were harvested 45 days after the sugar loading plateau was reached (the “mature fruit” stage). Vegetative aromas were synonymous with the “fresh fruit” stage in 2010/2011, while the 2011/2012 wines from the “mature fruit” harvest date were characterized by raisin, prune and spicy aromas. In both seasons, the control treatments were rated more intense in ‘satin in the mouth’ in and after expectoration. Wines in which the UV-B radiation was excluded during berry growth were rated the highest in the mouthfeel attribute ‘coarseness’ in both treatment seasons. Wines were analyzed chemically for phenolic content using HPLC, and sensorial using descriptive analysis with a trained panel. In the leaf removal treatments,higher acidity content enhanced the perception of astringency in the wine. Overall, the data showed that grape composition was altered by varying light quality, within a season, but seasonalvariation overrode treatment effects. Flavonol concentration in 2011/2012 wine was higher in the exposed leaf removal treatment compared to the other treatments. High light intensities in 2011/2012 season increased anthocyanin concentration in the wine.. This study emphasizes the importance of the quality and quantity of light on the composition and quality of wines, and presents new findings regarding sensory attributes associated with harvesting at different ripening stages.

    Pathogenesis-related proteins in grapevines induced by salicylic acid and Botrytis cinerea

    The grapevine pathogenesis-related proteins (PRs) were studied in order to determine the spatial localization and the temporal production of these inducible proteins. We used leaves of plants obtained from woody cuttings grown in greenhouse. Elicitations were done either with salicylic acid or with strains of Botrytis cinerea. Several PRs extractable at pH 2.8 were found to accumulate in grapevine leaves after salicylic acid treatment or Botrytis infection (SDS-PAGE, coomassie blue). Elicitation with salicylic acid has induced one new protein at about 32 kDa. Botrytis infection has resulted in the accumulation of four major acid-soluble proteins with apparent molecular weights of 27, 32, 34 and 38 kDa. Immunodetections using antisera raised against the tobacco PR-2 family have shown several bands, particularly two bands at 34 and 36 kDa revealed by the anti-2a and present both with salicylic acid and Botrytis

    Spatiotemporal changes in the accumulation of sugar and potassium within individual 'Sauvignon Blanc' (Vitis vinifera L.) berries

    It has been speculated that there may be a link between the transport of sugar and potassium into grape berries during ripening as they exhibit similar accumulation patterns. It is unclear if this proposed link is apparent in individual grape berries and in the grape berry compartments. Single grape berries were therefore analysed for sugar and potassium content and concentration within the skin, seeds and the pulp from pre-véraison until harvest. Sugar and potassium had similar accumulation patterns and positive relationships were confirmed between the sugar and potassium content within individual berries and compartments. The sugar content in the grape berry, however, increased 5-fold during ripening whereas the potassium content only doubled. Both sugar and potassium increased with berry size, suggesting a ternary relationship with berry water. The high variability in sugar and potassium contents between berries however affirms plasticity in their accumulation within individual berries

    Transcription of a β-1,3-glucanase gene in grape berries in a late developmental period, or earlier after wounding treatments

    The hydrolytic enzymes β-1,3-glucanases (EC3.2.1.39) an known to be involved in plant defense reactions against pathogens and in developmental events. We have obtained two partial cDNA clones, corresponding to Vitis vinifera β-1,3-glucanase gene sequences (VvGlu1/7 and VvGlu26). The expression of VvGlu1/7 was studied in grape berries at different developmental stages and in wounded berries with or without salicylic acid elicitation. No constitutive expression was observed in young berries, whereas the induction of VvGlu1/7 transcription was detected during fruit ripening. By contrast, synthesis of mRNA coding for this isoform was generated in the first stage of rapid berry growth in response to wounding treatments with and without salicylic acid

    The notion of distinct ‘public’ and ‘private’ spheres underpins much normative and practical engagement with political misconduct. What is less clear is whether citizens draw distinctions between misdemeanours in the ‘public’ and ‘private’ spheres, and whether they judge these in systematically different ways. This paper explores attitudes to political misconduct in France. French citizens are often said to be particularly relaxed about politicians’ private affairs, but there has been little empirical evidence for this proposition. Drawing on original survey data, this paper demonstrates clearly that French citizens draw a sharp distinction between politicians’ public and private transgressions, and are more tolerant of the latter

    The effects of applied water at various fractions of measured evapotranspiration on reproductive growth and water productivity of Thompson Seedless grapevines

    The reproductive growth and water productivity (WPb) of Thompson Seedless grapevines were measured as a function of applied water amounts at various fractions of measured grapevine ETc for a total of eight irrigation treatments. Shoots were harvested numerous times during the growing season to calculate water productivity. Berry weight was maximized at the 0.6–0.8 applied water treatments across years. As applied water amounts increased soluble solids decreased. Berry weight measured at veraison and harvest was a linear function of the mean midday leaf water potential measured between anthesis and veraison and anthesis and harvest, respectively. As applied water amounts increased up to the 0.6–0.8 irrigation treatments there was a significant linear increase in yield. Yields at greater applied water amounts either leveled off or decreased. The reduction in yield on either side of the yearly maximum was due to fewer numbers of clusters per vine. Maximum yield occurred at an ETc ranging from 550 to 700 mm. Yield per unit applied water and WPb increased as applied water decreased. The results from this study demonstrated that Thompson Seedless grapevines can be deficit irrigated, increasing water use efficiency while maximizing yields

    ViralORFeome: an integrated database to generate a versatile collection of viral ORFs

    Large collections of protein-encoding open reading frames (ORFs) established in a versatile recombination-based cloning system have been instrumental to study protein functions in high-throughput assays. Such ‘ORFeome’ resources have been developed for several organisms but in virology, plasmid collections covering a significant fraction of the virosphere are still needed. In this perspective, we present ViralORFeome 1.0 (http://www.viralorfeome.com), an open-access database and management system that provides an integrated set of bioinformatic tools to clone viral ORFs in the Gateway® system. ViralORFeome provides a convenient interface to navigate through virus genome sequences, to design ORF-specific cloning primers, to validate the sequence of generated constructs and to browse established collections of virus ORFs. Most importantly, ViralORFeome has been designed to manage all possible variants or mutants of a given ORF so that the cloning procedure can be applied to any emerging virus strain. A subset of plasmid constructs generated with ViralORFeome platform has been tested with success for heterologous protein expression in different expression systems at proteome scale. ViralORFeome should provide our community with a framework to establish a large collection of virus ORF clones, an instrumental resource to determine functions, activities and binding partners of viral proteins

    Physicochemical characterization of must and wine Moscato Giallo from grapevines grown under plastic overhead cover

    O objetivo deste trabalho foi avaliar a influência da cobertura de plástico sobre as características físico-químicas do mosto e do vinho da cultivar Moscato Giallo. Na safra 2006, um experimento em delineamento completamente casualizado foi realizado em vinhedo com cobertura de plástico impermeável, e sem cobertura como controle. De cada vinhedo, três microvinificações (20 L) foram elaboradas. Foram realizadas avaliações físico-químicas quanto ao: mosto – oBrix, açúcares redutores, densidade, acidez total, ácido tartárico, ácido málico e pH; e vinho – densidade, graduação alcoólica, acidez total, acidez volátil, pH, extrato seco, açúcares redutores, cinzas, I 420, compostos voláteis e minerais. O mosto das videiras cobertas apresentou maior rendimento, porém, menor concentração de açúcares pelo fato de a maturação das uvas ter-se atrasado. Como conseqüência, os vinhos do cultivo protegido tiveram menor graduação alcoólica, embora tenham sido beneficiados pela sanidade das uvas, com a redução de acetato de etila e acidez volátil. O microclima da cobertura também restringiu a concentração de alguns minerais no vinho, principalmente P e K. A cobertura beneficiou a qualidade enológica, porém requer atraso na data de colheita, para as uvas atingirem adequada maturação fisiológica e tecnológica.The objective of this work was to evaluate the plastic overhead cover (POC) effect on must and wine composition of the cultivar Moscato Giallo. In 2006 vintage, it was performed a completely randomly designed experiment, in a vineyard covered with an impermeable plastic cloth, and without covering as control. From each vineyard, three microvinification (20 L) were elaborated. Physico-chemical analysis were performed for: musts – oBrix, reducer sugar contents, density, total acidity, tartaric acid, malic acid and pH; and wines – density, alcohol degree, acidity, volatile acidity, pH, dry extract, reducer sugar contents, ashes, I 420, volatile compounds and minerals. The must of the covered vineyard presented higher yield, but lower sugar content, due to delay on ripening. As consequence, the POC wines showed lower alcohol degree, but with the benefit of a lower ethyl-acetate and volatile acidity levels. The POC microclimate also caused a decrease of some minerals (mainly P and K) in the wines. POC improved the enological quality, however, this demands delay in the harvest date to achieve the adequate physiological and technological maturation of grapes
