Novel strategies enhancing endodontic disinfection: antibacterial biodegradable calcium hydroxide nanoparticles in an ex vivo model.
Due to the high failure rates associated to endodontic disinfection, this study aimed to investigate the antibacterial properties of poly(lactic-co-glycolic acid) (PLGA) nanoparticles (NPs) loaded with Ca(OH)2 for endodontic disinfection procedures. Ca(OH)2 NPs production and physicochemical characterization were carried out as well as multiple antibacterial tests using three bacterial strains and an ex vivo model of endodontic infection with extracted human teeth. Agar diffusion test and broth dilution determined the inhibition growth zones (n=5) and the minimal inhibitory concentration (MIC, n=5), respectively. Cell viability was assessed using Live/Dead staining with confocal microscopy (n=5). Data was analysed using ANOVA followed by post-hoc analysis. After 24 hours of incubation, Ca(OH)₂ NPs demonstrated a MIC of 10 µg/mL for Porphyromonas gingivalis (p<0.001) and Enterococcus faecalis and 5 µg/mL for Fusobacterium nucleatum (p<0.001). Although the agar diffusion test did not exhibit any inhibition for Ca(OH)2 nor for Ca(OH)₂ NPs area probably due to the buffering effect of the agar medium. However, the antibacterial capacity was confirmed in an ex vivo model, where instrumentalized teeth were infected with Enterococcus Faecalis and treated after 28 days of culture. A significant reduction in bacterial metabolic activity was confirmed for Ca(OH)2 NPs (40% reduction with a single dose) and confirmed by Live/Dead staining. In conclusion, Ca(OH)₂-loaded PLGA NPs present promising antibacterial efficacy for endodontic disinfection procedures
Efficacy of the unified protocol for the treatment of emotional disorders in the Spanish public mental health system using a group format: study protocol for a multicenter, randomized, non-inferiority controlled trial
Background:
Emotional disorders, which include both anxiety and depressive disorders, are the most prevalent
psychological disorders according to recent epidemiological studies. Consequently, public costs associated with their
treatment have become a matter of concern for public health systems, which face long waiting lists. Because of their
high prevalence in the population, finding an effective treatment for emotional disorders has become a key goal of
today
’
s clinical psychology. The Unified Protocol for the Transdiagnostic Treatment of Emotional Disorders might serve
the aforementioned purpose, as it can be applied to a variety of disorders simultaneously and it can be
easily performed in a group format.
Methods:
The study is a multicenter, randomized, non-inferiority controlled clinical trial. Participants will be 220
individuals with emotional disorders, who are randomized to either a treatment as usual (individual cognitive
behavioral therapy) or to a Unified Protocol condition in group format. Depression, anxiety, and diagnostic criteria are
the primary outcome measures. Secondary measures include the assessment of positive and negative affect, anxiety
control, personality traits, overall adjustment, and quality of life. An analysis of treatment satisfaction is also conducted.
Assessment points include baseline, post-treatment, and three follow-ups at 3, 6, and 12 months. To control for missing
data and possible biases, intention-to-treat and per-protocol analyses will be performed.
Discussion:
This is the first randomized, controlled clinical trial to test the effectiveness of a transdiagnostic
intervention in a group format for the treatment of emotional disorders in public settings in Spain. Results obtained
from this study may have important clinical, social, and economic implications for public mental health settings in
Spain.

active and recruitment is ongoing. Recruitment is expected to finish by January 202
Association between Neutrophil-to-Lymphocyte Ratio with Abdominal Obesity and Healthy Eating Index in a Representative Older Spanish Population
Poor diet quality and obesity, especially abdominal obesity, have been associated with systemic inflammation. The neutrophil-to-lymphocyte Ratio (NLR) is an available and inexpensive inflammation biomarker. The aim of the present study was to determine the association of dietary patterns and obesity with an inflammatory state. A group of 1747 Spanish noninstitutionalized older adults individuals were included, and a food-frequency questionnaire was applied. The Global Food Score (GFS) and Healthy Eating Index for Spanish population (SHEI) were calculated. Weight, height and waist (WC) and hip circumferences were measured, and BMI, waist-to-height ratio (WHtR), and waist-to-hip ratio (WHR) determined. In addition, body-fat percentage was measured by bioimpedance. NLR was calculated (NLR ≥ p80: 2.6; 2.8 and 2.4 as inflammatory status in the entire population, men and women, respectively). The men with inflammatory status presented significative higher values of WC, WHtR, WHR, and body-fat percentage (101.82 ± 10.34 cm, 0.61 ± 0.06, 0.98 ± 0.06, and 31.68 ± 5.94%, respectively) than those with better inflammatory status (100.18 ± 10.22 cm, 0.59 ± 0.06, 0.97 ± 0.07, and 30.31 ± 6.16%, respectively). Those males with worse inflammatory state had lower scores for protein foods (OR = 0.898 (0.812–0.993); p = 0.037). The women with NLR ≥ 2.4 had higher WHtR and WHR (0.62 ± 0.09 and 0.91 ± 0.09) than those with NLR < 2.4 (0.60 ± 0.08 and 0.90 ± 0.08). In multiple linear regression analysis, NLR was positively related with WHtR and negatively related with SHEI score (β = 0.224 ± 0.094; R 2 = 0.060; p < 0.05 and β = −0.218 ± 0.101; R 2 = 0.061; p < 0.05), adjusting by sex, age, marital status, education level, smoking, hours of sleeping and inflammatory diseases. In women, the higher the SHEI and GFS scores were and the better meeting the aims of cereal and vegetable servings, the less the odds of inflammatory status (OR = 0.970 (0.948–0.992); p = 0.008; OR = 0.963 (0.932–0.995); p = 0.024; OR = 0.818 (0.688–0.974); p = 0.024 and OR = 0.829 (0.730–0.942); p = 0.004, respectively). WHtR and quality of diet is related to the inflammation status in older adults regardless to the sex
Chemical Abundances and Ionizing Clusters of HII Regions in the LINER Galaxy NGC 4258
We present long-slit observations in the optical and near infrared of eight
HII regions in the spiral galaxy NGC 4258. Six of the observed regions are
located in the SE inner spiral arms and the other two are isolated in the
northern outer arms. A detailed analysis of the physical conditions of the gas
has been performed. For two of the regions, an electron temperature has been
derived from the [SIII] 6312 A line. For the rest, an empirical calibration
based on the red and near infrared sulphur lines has been used. The oxygen
abundances derived by both methods are found to be significantly lower (by a
factor of two) than previously derived by using empirical calibrations based on
the optical oxygen lines. In the brightest region, 74C, the observation of a
prominent feature due to Wolf-Rayet stars provides an excellent constraint over
some properties of the ionizing clusters. In the light of the current
evolutionary synthesis models, no consistent solution is found to explain at
the same time both the WR feature characteristics and the emission line
spectrum of this region. In principle, the presence of WR stars could lead to
large temperature fluctuations and also to a hardening of the ionizing
radiation. None of these effects are found in region 74C for which the electron
temperatures found from the [SIII] 6312A and the Paschen discontinuity at 8200A
are equal within the errors and the effective temperature of the ionizing
radiation is estimated at around 35,300K. Both more observations of confirmed
high metallicity regions and a finer metallicity grid for the evolutionary
synthesis models are needed in order to understand the ionizing populations of
HII regions.Comment: 16 pages, 16 figures. To appear in MNRA
