    Survival and activity of an indigenous iron-reducing microbial community from MX80 bentonite in high temperature / low water environments with relevance to a proposed method of nuclear waste disposal

    MX80 bentonite clay has been selected as the buffer and backfill in a proposed method for long-term deep geological storage of nuclear waste. Extensive studies have been carried out on the geomechanical properties of the clay; however, it is not clear what effect microbes, specifically iron-reducing bacteria, will have on its ability to function as an affective barrier. Iron-reducing bacteria can reduce structural or external Fe(III) to Fe(II) and have been previously identified in the indigenous microbial community of MX80 bentonite. Experiments to assess bacterial survival at the high temperature and low water conditions likely to exist in the repository were carried out at different temperatures with the addition of steel to represent a nuclear waste canister. The resulting microbial enrichments were analysed, and mineralogical and geomechnical analysis was carried out on the clay. Microbial sequencing revealed that iron-reducing bacteria, and other indigenous species can survive these conditions in MX80 bentonite in either an active or dormant state. Microbial influenced mineralogical changes may lead to a loss of silica from the clay and reduction of Fe(III) to Fe(II). These changes could alter the ability of the clay to act as an effective barrier in nuclear waste disposal. Furthermore, evidence of reduced steel corrosion when microbes were present suggested that microbial activity may lead to either a protective coating on the steel or depletion of oxygen to slow corrosion. The production of such a layer would benefit nuclear waste disposal by inhibiting corrosion of a metal waste canister

    Vision screening in children:a retrospective study of social and demographic factors with regard to visual outcomes

    BACKGROUND: Amblyopia and its risk factors have been demonstrated to be more common among children from low socioeconomic backgrounds. We sought to investigate this association in a region with orthoptic-delivered screening and whole population coverage, and to also examine the association of the Health Plan Indicator (HPI) with screening outcome. METHODS: Screening examination outcomes, postcodes and HPIs were extracted from the community child health database for every child who underwent preschool vision screening between March 2010 and February 2011 Tayside. We obtained the Scottish Index of Multiple Deprivation score for every child as a measure of area-based deprivation. We assessed the vulnerability/needs of the individual family through the HPI—‘Core’ (children and families receiving universal health visiting service), ‘Additional’ (receiving additional health/social support) and ‘Intensive’ (receiving high levels of support). The outcomes from follow-up examinations for those who failed screening were extracted from the orthoptic department database. RESULTS: 4365 children were screened during the year 2010–2011 of whom 523 (11.9%) failed. The odds of children from the least deprived socioeconomic group passing the visual screening test was 1.4 times higher than those from the most deprived socioeconomic group (OR 1.4, 95% CI 1.07 to 1.89, p=0.01). The odds of a child from a family assigned as ‘Intensive’ failing the preschool visual screening test was three times greater than the odds of a child from a family assigned as ‘Core’ (OR 3.59, 95% CI 1.6 to 7.8, p=0.001). CONCLUSIONS: We found that children from the most deprived backgrounds and those from unstable homes were more likely to fail preschool vision screening

    The siblings of children diagnosed with cancer can get lost in the cancer journey. Through a mother’s eyes, this paper examines the issues faced by three siblings when their teenage sister is diagnosed with and dies of cancer. It focusses on the impact their sister’s illness has on their educational, social, emotional and communication skills. Each sibling not only had their own unique set of needs but this paper argues that the term ‘cancer survivor’ should also be extended to the siblings as they comprise an intrinsic part of the family’s cancer journey. The paper calls on professionals to adopt a holistic approach to reduce the impact that childhood cancer has on the siblings and it highlights the far-reaching consequences of not properly supporting the siblings during this time

    What does it mean when people call a place a shithole? Understanding a discourse of denigration in the United Kingdom and the Republic of Ireland

    This paper investigates what people mean when they engage in the discourse of denigration. Building on existing literature on territorial stigmatisation that either focuses on macro‐scale uses and effects of territorial stigmatisation or micro‐scale ethnographic studies of effects, we develop a novel approach that captures the diverse voices that engage in the discourse of denigration by tracing the use of the word and hashtag “shithole” on the social media platform Twitter in order to examine who is engaged in the stigmatising discourse, the types of place that are stigmatised and the responses to stigmatised places. Using a robust data set, we highlight two key findings. First, the majority of tweets were aimed at places where the tweeter was not from, a form of othering consistent with how territories are stigmatised by those in positions of power such as policymakers, politicians and journalists. Second, we note that an important and gendered minority of tweets can be characterised by a “cry for help” and powerlessness, where the stigma is aimed at their own places. We offer an interpretive lens through which to understand and frame these minoritarian voices by engaging with theories of abjection that allow us to see how minoritarian voices relate to place

    Primordialists and Constructionists: a typology of theories of religion

    This article adopts categories from nationalism theory to classify theories of religion. Primordialist explanations are grounded in evolutionary psychology and emphasize the innate human demand for religion. Primordialists predict that religion does not decline in the modern era but will endure in perpetuity. Constructionist theories argue that religious demand is a human construct. Modernity initially energizes religion, but subsequently undermines it. Unpacking these ideal types is necessary in order to describe actual theorists of religion. Three distinctions within primordialism and constructionism are relevant. Namely those distinguishing: a) materialist from symbolist forms of constructionism; b) theories of origins from those pertaining to the reproduction of religion; and c) within reproduction, between theories of religious persistence and secularization. This typology helps to make sense of theories of religion by classifying them on the basis of their causal mechanisms, chronology and effects. In so doing, it opens up new sightlines for theory and research

    Theoretical studies of the historical development of the accounting discipline: a review and evidence

    Many existing studies of the development of accounting thought have either been atheoretical or have adopted Kuhn's model of scientific growth. The limitations of this 35-year-old model are discussed. Four different general neo-Kuhnian models of scholarly knowledge development are reviewed and compared with reference to an analytical matrix. The models are found to be mutually consistent, with each focusing on a different aspect of development. A composite model is proposed. Based on a hand-crafted database, author co-citation analysis is used to map empirically the entire literature structure of the accounting discipline during two consecutive time periods, 1972–81 and 1982–90. The changing structure of the accounting literature is interpreted using the proposed composite model of scholarly knowledge development

    Deficiency of Pkc1 activity affects glycerol metabolism in Saccharomyces cerevisiae

    In pressProtein kinase C is apparently involved in the control of many cellular systems: the cell wall integrity pathway, the synthesis of ribosomes, the appropriated reallocation of transcription factors under specific stress conditions and also the regulation of N-glycosylation activity. All these observations suggest the existence of additional targets not yet identified. In the context of the control of carbon metabolism, previous data demonstrated that Pkc1 p might play a central role in the control of cellular growth and metabolism in yeast. In particular, it has been suggested that it might be involved in the derepression of genes under glucose-repression by driving an appropriated subcellular localization of transcriptional factors, such as Mig1 p. In this work, we show that pkc1∆ mutant is unable to grow on glycerol because it cannot perform the derepression of GUT1 gene that encodes for glycerol kinase. Additionally, active transport is also partially affected. Using this phenotype, we were able to isolate a new pkc1∆ revertant. We also isolated two transformants identified as the nuclear exportin Msn5 and the histone deacetylase Hos2 extragenic suppressors of this mutation. Based on these results, we postulate that Pkc1 p may be involved in the control of the cellular localization and/or regulation of the activity of nuclear proteins implicated in gene expression.Fundação Universidade Federal de Ouro Preto (FUFOP).     Effects of valproic acid on the cell cycle and apoptosis through acetylation of histone and tubulin in a scirrhous gastric cancer cell line

    <p>Abstract</p> <p>Background</p> <p>Management of peritoneal dissemination is the most critical problem in gastric cancer. This study was performed to investigate the inhibitory effects of valproic acid (VPA) on a highly peritoneal-seeding cell line of human scirrhous gastric cancer, OCUM-2MD3, and to explore the mechanism and the potential of VPA.</p> <p>Methods</p> <p>The effects of VPA on the growth of OCUM-2MD3 cells were assessed by MTT assay. In addition, paclitaxel (PTX) was combined with VPA to evaluate their synergistic effects. HDAC1 and HDAC2 expression were evaluated by western blotting in OCUM-2MD3 cells and other gastric cancer cell lines (TMK-1, MKN-28). The acetylation status of histone H3 and α-tubulin after exposure to VPA were analyzed by western blotting. The activities of cell cycle regulatory proteins and apoptosis-modulating proteins were also examined by western blotting. The effects of VPA <it>in vivo </it>were evaluated in a xenograft model, and apoptotic activity was assessed by TUNEL assay.</p> <p>Results</p> <p>OCUM-2MD3 cells showed high levels of HDAC1 and HDAC2 expression compared with TMK-1 and MKN-28. The concentration of VPA required for significant inhibition of cell viability (<it>P </it>< 0.05) was 5 mM at 24 h and 0.5 mM at 48 h and 72 h. The inhibition of VPA with PTX showed dose-dependent and combinatorial effects. VPA increased acetyl-histone H3, acetyl-α-tubulin, and p21WAF1 levels accompanied by upregulation of p27, caspase 3, and caspase 9, and downregulation of bcl-2, cyclin D1, and survivin. In the xenograft model experiment, the mean tumor volume of the VPA-treated group was significantly reduced by 36.4%, compared with that of the control group at 4 weeks after treatment (<it>P </it>< 0.01). The apoptotic index was significantly higher in the VPA-treated group (42.3% ± 3.5%) than in the control group (7.7% ± 2.5%) (<it>P </it>< 0.001).</p> <p>Conclusions</p> <p>VPA induced dynamic modulation of histone H3 and α-tubulin acetylation in relation with the anticancer effect and the enhancement of PTX in the OCUM-2MD3 cell line. Therefore, VPA in combination with PTX is expected to be a promising therapy for peritoneal dissemination of scirrhous gastric cancer.</p
