    Evaluating pharmacological models of high and low anxiety in sheep

    New tests of animal affect and welfare require validation in subjects experiencing putatively different states. Pharmacological manipulations of affective state are advantageous because they can be administered in a standardised fashion, and the duration of their action can be established and tailored to suit the length of a particular test. To this end, the current study aimed to evaluate a pharmacological model of high and low anxiety in an important agricultural and laboratory species, the sheep. Thirty-five 8-month-old female sheep received either an intramuscular injection of the putatively anxiogenic drug 1-(m-chlorophenyl)piperazine (mCPP; 1 mg/kg; n = 12), an intravenous injection of the putatively anxiolytic drug diazepam (0.1 mg/kg; n = 12), or acted as a control (saline intramuscular injection n = 11). Thirty minutes after the treatments, sheep were individually exposed to a variety of tests assessing their general movement, performance in a ‘runway task’ (moving down a raceway for a food reward), response to startle, and behaviour in isolation. A test to assess feeding motivation was performed 2 days later following administration of the drugs to the same animals in the same manner. The mCPP sheep had poorer performance in the two runway tasks (6.8 and 7.7 × slower respectively than control group; p < 0.001), a greater startle response (1.4 vs. 0.6; p = 0.02), a higher level of movement during isolation (9.1 steps vs. 5.4; p < 0.001), and a lower feeding motivation (1.8 × slower; p < 0.001) than the control group, all of which act as indicators of anxiety. These results show that mCPP is an effective pharmacological model of high anxiety in sheep. Comparatively, the sheep treated with diazepam did not display any differences compared to the control sheep. Thus we suggest that mCPP is an effective treatment to validate future tests aimed at assessing anxiety in sheep, and that future studies should include other subtle indicators of positive affective states, as well as dosage studies, so conclusions on the efficacy of diazepam as a model of low anxiety can be drawn

    The Clinical Usefulness of Measuring Apolipoproteins in Diabetic Patients: A Preliminary Report

    A commercial assay for apolipoproteins A-1 and B as well as total cholesterol, triglyceride, and high-density lipoprotein (HDL) cholesterol was applied to 12-hour fasted serum from 24 insulin-dependent and 19 noninsulin-dependent diabetic persons. Women with noninsulin-dependent diabetes mellitus (NIDDM) had the highest levels of total cholesterol and apolipoprotein B. Apolipoprotein B values fell within the normal range in all patients except the NIDDM females, where four of the ten (40%) samples were elevated. When apolipoprotein B was elevated, total cholesterol was also elevated, over 220 mg/dL. Apolipoprotein A-1 values fell within or above the normal range in all subjects, and a considerable discordance was observed between apolipoprotein A-1 elevations and HDL cholesterol elevations. The divergence between HDL cholesterol and apolipoprotein A-1 supports the altered composition of HDL in the diabetic persons studied. Further study should elucidate the clinical usefulness of apolipoprotein measurements in diabetic patients

    The aim of this research was to get optimum number of maintained young fruit per panicle and good criteria of fruit that must be maintained so it will get a high yield of mango. The factors under investigation were number of young fruit to be maintained per panicle: 2, 4, and 6; the criteria of young fruit to be maintained: the biggest fruit, fruit attached at the beginning of panicle, and fruit attached at panicle at the same distance. The result of research showed that 6 fruits that maintained per panicle gave the highest weight per tree of mango (33,45 kg), but the lowest weight per fruit (466,98 kg). Maintaining the 2 biggest fruits of mango gave the highest weight per fruit (510,53 g)

    Estimating Random Errors Due to Shot Noise in Backscatter Lidar Observations

    In this paper, we discuss the estimation of random errors due to shot noise in backscatter lidar observations that use either photomultiplier tube (PMT) or avalanche photodiode (APD) detectors. The statistical characteristics of photodetection are reviewed, and photon count distributions of solar background signals and laser backscatter signals are examined using airborne lidar observations at 532 nm using a photon-counting mode APD. Both distributions appear to be Poisson, indicating that the arrival at the photodetector of photons for these signals is a Poisson stochastic process. For Poisson-distributed signals, a proportional, one-to-one relationship is known to exist between the mean of a distribution and its variance. Although the multiplied photocurrent no longer follows a strict Poisson distribution in analog-mode APD and PMT detectors, the proportionality still exists between the mean and the variance of the multiplied photocurrent. We make use of this relationship by introducing the noise scale factor (NSF), which quantifies the constant of proportionality that exists between the root-mean-square of the random noise in a measurement and the square root of the mean signal. Using the NSF to estimate random errors in lidar measurements due to shot noise provides a significant advantage over the conventional error estimation techniques, in that with the NSF uncertainties can be reliably calculated from/for a single data sample. Methods for evaluating the NSF are presented. Algorithms to compute the NSF are developed for the Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observations (CALIPSO) lidar and tested using data from the Lidar In-space Technology Experiment (LITE).     © 2019 The Author(s) The aim of this study is the development of validated HPTLC method for the quantification of vitexin from Passiflora foetida commercial herbal formulations. The developed method was validated, in accordance with ICH guidelines for precision, accuracy, specificity and robustness. The plate was developed using ethyl acetate:methanol:water:formic acid 30:4:2:1(%, v/v/v/v) on 20 × 10 cm glass coated silica gel 60 F254 plates and the developed plate was scanned and quantified densitometrically at λ = 340 nm. Linear regression analysis revealed a good linear relationship between peak area and amount of vitexin in the range of 100–700 ng/spot. The amount of vitexin in nine commercial herbal formulations was successfully quantified by the developed HPTLC method. The developed and validated high performance thin layer chromatographic method offers a new sensitive and reliable tool for quantification of vitexinin in various herbal formulations containing Passiflora foetida

    Towards a Precise Measurement of Matter Clustering: Lyman-alpha Forest Data at Redshifts 2-4

    We measure the filling factor, correlation function, and power spectrum of transmitted flux in a large sample of Lya forest spectra, comprised of 30 Keck HIRES spectra and 23 Keck LRIS spectra. We infer the linear matter power spectrum P(k) from the flux power spectrum P_F(k), using an improved version of the method of Croft et al. (1998) that accounts for the influence of z-space distortions, non- linearity, and thermal broadening on P_F(k). The evolution of the shape and amplitude of P(k) over the range z= 2-4 is consistent with gravitational instability, implying that non-gravitational fluctuations do not make a large contribution. Our fiducial measurement of P(k) comes from data with = 2.72. It has amplitude Delta^2(k_p)=0.74^0.20_-0.16 at wavenumber k_p=0.03 (km/s)^-1 and is well described by a power-law of index -2.43 +/- 0.06 or by a CDM-like power spectrum with shape parameter Gamma'=1.3^+0.7_-0.5*10^-3 (km/s) at z=2.72. For Omega_m=0.4, Omega_Lam=0.6, the best-fit Gamma =0.16 (h^-1mpc)^-1, in good agreement with the 2dF Galaxy Redshift Survey, and the best-fit sigma_8=0.82 (Gamma/0.15)^-0.44. Matching the observed cluster mass function and our Delta^2(k_p) in spatially flat models requires Omega_m=0.38^+0.10_-0.08 + 2.2 (Gamma-0.15). Matching Delta^2(k_p) in COBE-normalized, flat CDM models with no tensor fluctuations requires Omega_m = (0.29 +/-0.04) n^-2.89 h_65^-1.9. The Lya forest complements other probes of P(k) by constraining a regime of redshift and lengthscale not accessible by other means, and the consistency of these inferred parameters with independent estimates provides further support for inflation, cold dark matter, and vacuum energy (abridged).Comment: Revised version fixes error in normalization (now 1 sigma higher).     The Lick AGN Monitoring Project 2011: Dynamical Modeling of the Broad Line Region in Mrk 50

    We present dynamical modeling of the broad line region (BLR) in the Seyfert 1 galaxy Mrk 50 using reverberation mapping data taken as part of the Lick AGN Monitoring Project (LAMP) 2011. We model the reverberation mapping data directly, constraining the geometry and kinematics of the BLR, as well as deriving a black hole mass estimate that does not depend on a normalizing factor or virial coefficient. We find that the geometry of the BLR in Mrk 50 is a nearly face-on thick disk, with a mean radius of 9.6(+1.2,-0.9) light days, a width of the BLR of 6.9(+1.2,-1.1) light days, and a disk opening angle of 25\pm10 degrees above the plane. We also constrain the inclination angle to be 9(+7,-5) degrees, close to face-on. Finally, the black hole mass of Mrk 50 is inferred to be log10(M(BH)/Msun) = 7.57(+0.44,-0.27). By comparison to the virial black hole mass estimate from traditional reverberation mapping analysis, we find the normalizing constant (virial coefficient) to be log10(f) = 0.78(+0.44,-0.27), consistent with the commonly adopted mean value of 0.74 based on aligning the M(BH)-{\sigma}* relation for AGN and quiescent galaxies. While our dynamical model includes the possibility of a net inflow or outflow in the BLR, we cannot distinguish between these two scenarios.Comment: Accepted for publication in ApJ. 8 pages, 6 figure

    Convective distribution of tropospheric ozone and tracers in the Central American ITCZ region: Evidence from observations during TC4

    During the Tropical Composition, Clouds and Climate Coupling (TC4) experiment that occurred in July and August of 2007, extensive sampling of active convection in the ITCZ region near Central America was performed from multiple aircraft and satellite sensors. As part of a sampling strategy designed to study cloud processes, the NASA ER-2, WB-57 and DC-8 flew in stacked “racetrack patterns” in convective cells. On July 24, 2007, the ER-2 and DC-8 probed an actively developing storm and the DC-8 was hit by lightning. Case studies of this flight, and of convective outflow on August 5, 2007 reveal a significant anti-correlation between ozone and condensed cloud water content. With little variability in the boundary layer and a vertical gradient, low ozone in the upper troposphere indicates convective transport. Because of the large spatial and temporal variability in surface CO and other pollutants in this region, low ozone is a better convective indicator. Lower tropospheric tracers methyl hydrogen peroxide, total organic bromine and calcium substantiate the ozone results. OMI measurements of mean upper tropospheric ozone near convection show lower ozone in convective outflow. A mass balance estimation of the amount of convective turnover below the tropical tropopause transition layer (TTL) is 50%, with an altitude of maximum convective outflow located between 10 and 11 km, 4 km below the cirrus anvil tops. It appears that convective lofting in this region of the ITCZ is either a two-stage or a rapid mixing process, because undiluted boundary layer air is never sampled in the convective outflow

    Automatic wide complex tachycardia differentiation using mathematically synthesized vectorcardiogram signals

    BACKGROUND: Automated wide complex tachycardia (WCT) differentiation into ventricular tachycardia (VT) and supraventricular wide complex tachycardia (SWCT) may be accomplished using novel calculations that quantify the extent of mean electrical vector changes between the WCT and baseline electrocardiogram (ECG). At present, it is unknown whether quantifying mean electrical vector changes within three orthogonal vectorcardiogram (VCG) leads (X, Y, and Z leads) can improve automated VT and SWCT classification. METHODS: A derivation cohort of paired WCT and baseline ECGs was used to derive five logistic regression models: (i) one novel WCT differentiation model (i.e., VCG Model), (ii) three previously developed WCT differentiation models (i.e., WCT Formula, VT Prediction Model, and WCT Formula II), and (iii) one all-inclusive model (i.e., Hybrid Model). A separate validation cohort of paired WCT and baseline ECGs was used to trial and compare each model\u27s performance. RESULTS: The VCG Model, composed of WCT QRS duration, baseline QRS duration, absolute change in QRS duration, X-lead QRS amplitude change, Y-lead QRS amplitude change, and Z-lead QRS amplitude change, demonstrated effective WCT differentiation (area under the curve [AUC] 0.94) for the derivation cohort. For the validation cohort, the diagnostic performance of the VCG Model (AUC 0.94) was similar to that achieved by the WCT Formula (AUC 0.95), VT Prediction Model (AUC 0.91), WCT Formula II (AUC 0.94), and Hybrid Model (AUC 0.95). CONCLUSION: Custom calculations derived from mathematically synthesized VCG signals may be used to formulate an effective means to differentiate WCTs automatically

    Impact of Scotland’s comprehensive, smoke-free legislation on stroke

    &lt;p&gt;Background: Previous studies have reported a reduction in acute coronary events following smoke-free legislation. Evidence is lacking on whether stroke is also reduced. The aim was to determine whether the incidence of stroke, overalland by sub-type, fell following introduction of smoke-free legislation across Scotland on 26 March 2006.&lt;/p&gt; &lt;p&gt;Methods and Findings: A negative binomial regression model was used to determine whether the introduction of smokefree legislation resulted in a step and/or slope change in stroke incidence. The model was adjusted for age-group, sex, socioeconomic deprivation quintile, urban/rural residence and month. Interaction tests were also performed. Routine hospital administrative data and death certificates were used to identify all hospital admissions and pre-hospital deaths due to stroke (ICD10 codes I61, I63 and I64) in Scotland between 2000 and 2010 inclusive. Prior to the legislation, rates of all stroke, intracerebral haemorrhage and unspecified stroke were decreasing, whilst cerebral infarction was increasing at 0.97% per annum. Following the legislation, there was a dramatic fall in cerebral infarctions that persisted for around 20 months. No visible effect was observed for other types of stroke. The model confirmed an 8.90% (95% CI 4.85, 12.77, p,0.001) stepwise reduction in cerebral infarction at the time the legislation was implemented, after adjustment for potential cofounders.&lt;/p&gt; &lt;p&gt;Conclusions: Following introduction of national, comprehensive smoke-free legislation there was a selective reduction in cerebral infarction that was not apparent in other types of stroke.&lt;/p&gt
