    Improved production of lutein and β-carotene by thermal and light intensity upshifts in the marine microalga Tetraselmis sp. CTP4

    The industrial microalga Tetraselmis sp. CTP4 is a promising candidate for aquaculture feed, novel food, cosmeceutical and nutraceutical due to its balanced biochemical profile. To further upgrade its biomass value, carotenogenesis was investigated by testing four environmental factors, namely temperature, light intensity, salinity and nutrient availability over different growth stages. The most important factor for carotenoid induction in this species is a sufficient supply of nitrates leading to an exponential growth of the cells. Furthermore, high temperatures of over 30 degrees C compared to lower temperatures (10 and 20 degrees C) induced the accumulation of carotenoids in this species. Remarkably, the two different branches of carotenoid synthesis were regulated depending on different light intensities. Contents of beta-carotene were 3-fold higher under low light intensities (33 mu mol m(-2) s(-1)) while lutein contents increased 1.5-fold under higher light intensities (170 and 280 mu mol m(-2) s(-1)). Nevertheless, highest contents of carotenoids (8.48 +/- 0.47 mg g(-1) DW) were found upon a thermal upshift from 20 degrees C to 35 degrees C after only two days at a light intensity of 170 mu mol m(-2) s(-1). Under these conditions, high contents of both lutein and beta-carotene were reached accounting for 3.17 +/- 0.18 and 3.21 +/- 0.18 mg g(-1) DW, respectively. This study indicates that Tetraselmis sp.     Detection Of Salmonella Spp. By Conventional Bacteriology And By Quantitative Polymerase-chain Reaction In Commercial Egg Structures

    Conventional bacteriology techniques and quantitative polymerasechain reaction (qPCR) were applied to the eggshell, albumen, and yolk of washed and unwashed commercial white and brown eggs, to detect Salmonella spp. Pooled samples of eggshells, albumen, and yolk of white and brown eggs were collected at the poultry house and at the egg-storage room. Salmonella spp. was detected by conventional bacteriology in 5.4% (21/387) of analyzed samples and in 16% (68/387) by qPCR. In the 114 unwashed white eggs samples of eggshell, albumen and yolk, the bacterium was identified in 2.6% of the eggs (3/114) by conventional bacteriology and in 13.2% (15/114) by qPCR. In the 90 samples of washed eggs, 6.7% (6/90) were contaminated as detected by conventional bacteriology and 10.0% (9/90) by qPCR. In the 81 samples of unwashed brown eggs, Salmonella spp. was detected in 6.1% of the eggs (5/81) by conventional bacteriology and 27.2% (22/81) by qPCR. In the 102 samples of brown washed eggs, 6.9% (7/102) where positive by conventional bacteriology and 35.3% (16/102) by qPCR. All samples detected as positive by conventional bacteriology were also positive by qPCR. Salmonella Agona represented 18.2% (4/22) of identified serovars, Salmonella enterica subs. enterica O: 4.5 18.2% (4/22), Salmonella Schwarzengrund 18.2% (4/22), Salmonella Cerro 13.6% (3/22), Salmonella Anatum 13.6% (3/22), Salmonella Enteritidis 9.1% (2/22), Salmonella Johannesburg 4.5% (1/22), and Salmonella Corvallis 4.5% (1/22). The qPCR method provided better detection of Salmonella spp. in commercial eggs than conventional bacteriology.     Insulin therapy modulates mitochondrial dynamics and biogenesis, autophagy and tau protein phosphorylation in the brain of type 1 diabetic rats

    AbstractThe main purpose of this study was to examine whether streptozotocin (STZ)-induced type 1 diabetes (T1D) and insulin (INS) treatment affect mitochondrial function, fission/fusion and biogenesis, autophagy and tau protein phosphorylation in cerebral cortex from diabetic rats treated or not with INS. No significant alterations were observed in mitochondrial function as well as pyruvate levels, despite the significant increase in glucose levels observed in INS-treated diabetic rats. A significant increase in DRP1 protein phosphorylated at Ser616 residue was observed in the brain cortex of STZ rats. Also an increase in NRF2 protein levels and in the number of copies of mtDNA were observed in STZ diabetic rats, these alterations being normalized by INS. A slight decrease in LC3-II levels was observed in INS-treated rats when compared to STZ diabetic animals. An increase in tau protein phosphorylation at Ser396 residue was observed in STZ diabetic rats while INS treatment partially reversed that effect. Accordingly, a modest reduction in the activation of GSK3β and a significant increase in the activity of phosphatase 2A were found in INS-treated rats when compared to STZ diabetic animals. No significant alterations were observed in caspases 9 and 3 activity and synaptophysin and PSD95 levels. Altogether our results show that mitochondrial alterations induced by T1D seem to involve compensation mechanisms since no significant changes in mitochondrial function and synaptic integrity were observed in diabetic animals. In addition, INS treatment is able to normalize the alterations induced by T1D supporting the importance of INS signaling in the brain

    Nematodes as indicators of environmental changes in a river with different levels of anthropogenic impact

    Abstract Considering that changes in the biodiversity of parasite communities can be used as indicators of ecosystem health, the aim of this study was to investigate the potential use of Geophagus brasiliensis parasites as bioindicators of environmental changes. We established three sample points in the Iguaçu River, each presenting different degrees of environmental impact. Out of the 69 G. brasiliensis specimens analyzed, 32 (46.3%) were parasitized by at least one parasite. We collected a total of 56 specimens of endoparasites belonging to the phylum Nematoda. Fishes collected in point 3 presented a significantly higher abundance of nematode species (moderately degraded) (Kruskal-Wallis2;69 = 8.62; p = 0.01) and species compositions between points were significantly different (F = 6.95, p = 0.002). No significant difference in relative condition factor (Kn) among the points (F2;66 = 2.54; p = 0.08) or correlation in Kn and abundance of nematodes (rs = 0.1; p = 0.4) were indicated. The results presented in this study indicate that the parasitic community of G. brasiliensis is characterized by low diversity in polluted locations, which explains the absence of certain parasite species and the occurrence of nematode species with varied responses to the pollution gradient

    Estudo longitudinal de um programa de reabilitação neuropsicológica dirigido a pacientes com doença de Alzheimer

    Our aim was to study the duration of benefits derived from a neuropsychological rehabilitation program (NRP) for dementia patients. METHOD: The participants in this study were three patients diagnosed as probable Alzheimer's disease in the initial-to-moderate phase; the three were taking anticholinesterases. They were submitted to a neuropsychological evaluation (NE) before the NRP and then revaluated after 12 and 24 months of treatment. The aim of our intervention was to do practical work with implicit and explicit residual memory by training them in everyday life activities, and using compensatory strategies and their intact cognitive abilities. RESULTS: Analysis of quantitative NE data (descriptive measures) after the first year of NRP showed cognitive improvement, functional stabilization and fewer behavioral problems. However, this improvement did not continue in the second year, and the disease maintained its characteristic progression.OBJETIVO: Estudar a duração do beneficio de um programa de reabilitação neuropsicológica (PRN) dirigido a pacientes demenciados. MÉTODO: Participaram deste estudo, três pacientes com diagnóstico de provável doença de Alzheimer em fase inicial a moderada. Todos faziam uso de anti-colinesterásicos e passaram por uma avaliação neuropsicológica (AN) antes de começar o PRN e reavaliação após 12 e 24 meses do tratamento. O alvo de nossa intervenção foi trabalhar de forma prática a memória explicita residual e implícita, através do treino das atividades da vida diária, uso de estratégias compensatórias e habilidades cognitivas ainda preservadas. RESULTADOS: A análise dos dados quantitativos (medidas descritivas) da AN mostrou, que após o primeiro ano do PRN houve uma melhora cognitiva, estabilização funcional e redução dos problemas comportamentais nos pacientes.     Seismic stratigraphy and sediment cores reveal lake-level fluctuations in Lake Iznik (NW Turkey) over the past ∼70 ka

    Our study aims to understand the palaeohydrological history of Lake Iznik and unravel the complex interplay between climatic, tectonic, and environmental factors that have shaped this Turkish basin. Through the analysis of seismic stratigraphy and sediment cores, we reveal a significant lowstand, indicating a lake level 60 m lower than today at ∼70 ka BP. Subsequently, a major phase of stepwise transgression is evidenced by 13 buried palaeoshorelines between ∼70 and 45 ka BP. From 45 to ∼10 ka cal BP, strong currents controlled the sedimentation in the lake, as evidenced by the occurrence of contourite drifts. Between ∼14 and 10 ka cal. BP, a major lowstand indicating a drier climate interrupted the current-controlled sedimentation regime. From ∼10 ka cal. BP, the subsequent increase in lake level occurred at the same time as the reconnection between the Mediterranean and Black seas. Archaeological evidence, including submerged structures of a basilica, establishes a link between lake-level changes and human settlement during the last millennium.     Fingolimod therapy in multiple sclerosis leads to the enrichment of a subpopulation of aged NK cells

    Fingolimod is an approved oral treatment for relapsing–remitting multiple sclerosis (RRMS) that modulates agonistically the sphingosin-1-phosphate receptor (S1PR), inhibiting thereby the egress of lymphocytes from the lymph nodes. In this interventional prospective clinical phase IV trial, we longitudinally investigated the impact of fingolimod on frequencies of NK cell subpopulations by flow cytometry in 17 RRMS patients at baseline and 1, 3, 6, and 12 months after treatment initiation. Clinical outcome was assessed by the Expanded Disability Status Scale (EDSS) and annualized relapse rates (ARR). Over the study period, median EDSS remained stable from month 3 to month 12, and ARR decreased compared to ARR in the 24 months prior treatment. Treatment was paralleled by an increased frequency of circulating NK cells, due primarily to an increase in CD56(dim)CD94(low) mature NK cells, while the CD56(bright) fraction and CD127(+) innate lymphoid cells (ILCs) decreased over time. An unsupervised clustering algorithm further revealed that a particular fraction of NK cells defined by the expression of CD56(dim)CD16(++)KIR(+/-)NKG2A(-)CD94(-)CCR7(+/-)CX(3)CR1(+/-)NKG2C(-)NKG2D(+)NKp46(-)DNAM1(++)CD127(+) increased during treatment. This specific phenotype might reflect a status of aged, fully differentiated, and less functional NK cells. Our study confirms that fingolimod treatment affects both NK cells and ILC. In addition, our study suggests that treatment leads to the enrichment of a specific NK cell subset characterized by an aged phenotype. This might limit the anti-microbial and anti-tumour NK cell activity in fingolimod-treated patients

    Morfoanatomia e histoquímica da semente de sororoca (Phenakospermum guyannense (Rich.) Endl. - Strelitziaceae)

    Phenakospermum guyannense, popularly known in the Amazon as sororoca, is usually found along rivers and in ombrophilous environments. The objective of this study was to describe the morpho-anatomy and histochemistry of mature seeds of P. guyannense collected at the Urubuí Waterfall, Presidente Figueiredo, Amazonas state, Brazil. Seed anatomy was studied using a light and a scanning electron microscopes (SEM). Histochemical tests were performed to identify phenolic compounds, starch, protein and lipids. The mature seed of P. guyannense is stenospermic, with a black seed coat composed of several layers of different cell types, with most containing phenolic compounds. The hilum is punctiform, surrounded by cells, which form the aryl. The endosperm is solid, formed by tetrahedral cells containing starch and protein. The embryo, which is cylindrical and located in the proximal region, is basal capitate, with cells containing lipids and proteins and is composed of a slightly dilated hypocotyl-radicle axis. The haustorium is flattened and located in the distal region
