    X-ray ptychography with a laboratory source

    X-ray ptychography has revolutionised nanoscale phase contrast imaging at large-scale synchrotron sources in recent years. We present here the first successful demonstration of the technique in a small-scale laboratory setting. We conducted an experiment with a liquid metal-jet X-ray source and a single photon-counting detector with a high spectral resolution. The experiment used a spot size of 5 microns to produce a ptychographic phase image of a Siemens star test pattern with a sub-micron spatial resolution. The result and methodology presented show how high-resolution phase contrast imaging can now be performed at small-scale laboratory sources worldwide

    Transmission of SARS and MERS coronaviruses and influenza virus in healthcare settings: the possible role of dry surface contamination

    Viruses with pandemic potential including H1N1, H5N1, and H5N7 influenza viruses, and severe acute respiratory syndrome (SARS)/Middle East respiratory syndrome (MERS) coronaviruses (CoV) have emerged in recent years. SARS-CoV, MERS-CoV, and influenza virus can survive on surfaces for extended periods, sometimes up to months. Factors influencing the survival of these viruses on surfaces include: strain variation, titre, surface type, suspending medium, mode of deposition, temperature and relative humidity, and the method used to determine the viability of the virus. Environmental sampling has identified contamination in field-settings with SARS-CoV and influenza virus, although the frequent use of molecular detection methods may not necessarily represent the presence of viable virus. The importance of indirect contact transmission (involving contamination of inanimate surfaces) is uncertain compared with other transmission routes, principally direct contact transmission (independent of surface contamination), droplet, and airborne routes. However, influenza virus and SARS-CoV may be shed into the environment and be transferred from environmental surfaces to hands of patients and healthcare providers. Emerging data suggest that MERS-CoV also shares these properties. Once contaminated from the environment, hands can then initiate self-inoculation of mucous membranes of the nose, eyes or mouth. Mathematical and animal models, and intervention studies suggest that contact transmission is the most important route in some scenarios. Infection prevention and control implications include the need for hand hygiene and personal protective equipment to minimize self-contamination and to protect against inoculation of mucosal surfaces and the respiratory tract, and enhanced surface cleaning and disinfection in healthcare settings

    In the Netherlands, the number of cocaine traffickers arrested at Schiphol International Airport peaked in 2003. In May 2003, sentencing guidelines regarding cocaine traffickers were redefined into a smaller number of categories (wider quantity ranges, less specified offender categories) with lower sentences than before. New guidelines allowed criminal courts more flexibility, and judges were expected to sentence cocaine traffickers more often than before in accordance with guidelines. All cases, at all four applicable criminal courts, against cocaine traffickers that had been arrested at Schiphol in 2003 were analyzed (877 simple court and 994 full court cases). In between 30 and 40 percent of cases sentences were not according to the guidelines (mostly lower), and even more frequently than before the new guidelines had been introduced. Experience as a judge (number of years) was not associated with keeping up with guidelines, but male judges sentenced suspects more often than female judges in discordance with guidelines (and consequently below guidelines). In line with American literature, personal characteristics of suspects appeared strong predictors of the extent judges conformed to sentencing guidelines. Female offenders were twice as often as male offenders sentenced lower than the guidelines. The same result was found for offenders born or living in the Netherlands, versus migrants and offenders living abroad

    Expression Patterns of a putative homolog of AGAMOUS, STAG1, from Strawberry

    MADS box genes function to regulate vegetative, floral, and fruit development in plants. Here we characterize the expression pattern of a MADS box gene from strawberry (Fragariaxananassa), designated STAG1. Sequence analysis revealed that STAG1 shared 68-91 mino acid sequence identity to AGAMOUS homologs from a variety of plant species. STAG1 transcripts were detected in stamens, carpels, and developing fruit. In situ hybridization revealed that STAG1 mRNA expression was restricted to the endothelium and the vascular bundles connecting the achenes to the inner part of the receptacle and was not evident in the receptacle of the fruit. Analysis of the expression of a GUS marker gene driven by the STAG1 promoter showed that during floral development, STAG1 was active in stamens, the base of the receptacle and the petals, and in the central pith and vascular tissue. During the ripening stage of fruit development, STAG1 activity was detected in achenes, pith cells, and cortical cells.     Item does not contain fulltextSentinel lymph node status is the most important prognostic factor in primary melanoma. The number of melanoma-associated lymphatic vessels has been associated with sentinel lymph node status and survival. Vascular endothelial growth factor-C (VEGF-C) is found to promote tumour-associated lymphatic vessel growth. In many human neoplasms, VEGF-C expression in neoplastic cells or tumour-associated macrophages (TAMs) has been linked to lymphatic dissemination of tumour cells. Recent studies have suggested a correlation between VEGF-C expression in primary melanoma and the presence of lymph node metastasis. We performed VEGF-C immunohistochemical staining on melanoma tissues of 113 patients with known sentinel lymph node status. We showed that both high VEGF-C expression in melanoma cells and TAMs are positively associated with the presence of a positive sentinel lymph node. No correlation with Breslow thickness, Clark invasion level or ulceration could be detected. VEGF-C expression in melanoma cells was predictive of a shorter overall and disease-free survival, without being an independent predictor of survival.
