Tolerance and safety of the potentially probiotic strain Lactobacillus rhamnosus PRSF-L477 : a randomised, double-blind placebo-controlled trial in healthy volunteers
In Europe, the species Lactobacillus rhamnosus is currently on the Qualified Presumption of Safety list used by the European Food Safety Authority (EFSA) for internal safety assessment, but according to the EFSA the species should remain a topic of surveillance. In the present study, the safety and tolerance of the potentially probiotic strain L. rhamnosus PRSF-L477 was investigated in a placebo-controlled double-blind volunteer trial following FAO/WHO guidelines. A total of thirty-four subjects received daily doses of 1 x 10(11) colony-forming units (cfu) of L. rhamnosus PRSF-L477 (n 17) or placebo (n 17) for a period of 3 weeks, followed by a wash-out period of another 3 weeks. A questionnaire on gastrointestinal tolerance and a diary was kept daily to record compliance throughout these 6 weeks. Faecal and blood samples were collected for microbiological and haematological analysis. The recorded gastrointestinal symptoms, defecation frequency and stool consistency were not influenced indicating that L. rhamnosus PRSF-L477 was well tolerated. The species L. rhamnosus was detected in the faeces of sixteen out of seventeen subjects of the probiotic group during the intervention period. Using pulsed-field gel electrophoresis, re-isolates of L. rhamnosus PRSF-L477 were confirmed in nine of these subjects. Antibiotic susceptibility profiles of these re-isolates were unchanged compared with PRSF-L477. No clinically relevant changes in blood parameters such as liver and kidney function and no serious adverse events appeared during and after administration. Therefore, we conclude that L. rhamnosus PRSF-L477 can safely be administrated to healthy subjects at a daily dose of 1 x 10(11) cfu
Novel conopeptides of the I-superfamily occur in several clades of cone snails
The I-superfamily of conotoxins represents a new class of peptides in the venom of some Conus species. These toxins are characterized by four disulfide bridges and inhibit or modify ion channels of nerve cells. When testing venoms from 11 Conus species for a functional characterization, blocking activity on potassium channels (like Kv1.1 and Kv1.3 channels, but not Kv1.2 channels) was detected in the venom of Conus capitaneus, Conus miles, Conus vexillum and Conus virgo. Analysis at the cDNA level of these venoms using primers designed according to the amino acid sequence of a potassium channel blocking toxin (ViTx) from C. virgo confirmed the presence of structurally homologous peptides in these venoms. Moreover, peptides belonging to the I-superfamily, but with divergent amino acid sequences, were found in Conus striatus and Conus imperialis. In all cases, the sequences of the precursors' prepro-regions exhibited high conservation, whereas the sequences of the mature peptides ranged from almost identical to highly divergent between species. We then performed phylogenetic analyses of new and published mitochondrial 16S rDNA sequences representing 104 haplotypes from these and numerous other Conus species, using Bayesian, maximum-likelihood, maximum-parsimony and neighbor-joining methods of inference. Cone snails known to possess I-superfamily toxins were assigned to five different major clades in all of the resulting gene trees. Moreover, I-superfamily conopeptides were detected both in vermivorous and piscivorous species of Conus, thus demonstrating the widespread presence of such toxins in this speciose genus beyond evolutionary and ecological groups

Genetic Diversity of PCR-Positive, Culture-Negative and Culture-Positive Mycobacterium ulcerans Isolated from Buruli Ulcer Patients in Ghana.
Culture of Mycobacterium ulcerans from Buruli ulcer patients has very low sensitivity. Thus confirmation of M. ulcerans infection is primarily based on PCR directed against IS2404. In this study we compare the genotypes obtained by variable number of tandem repeat analysis of DNA from IS2404-PCR positive cultures with that obtained from IS2404 positive, culture-negative tissue. A significantly greater genetic heterogeneity was found among culture-negative samples compared with that found in cultured strains but a single genotype is over-represented in both sample sets. This study provides evidence that both the focal location of bacteria in a lesion as well as differences in the ability to culture a particular genotype may underlie the low sensitivity of culture. Though preliminary, data from this work also suggests that mycobacteria previously associated with fish disease (M. pseudoshottsii) may be pathogenic for humans
Psychiatric Illnesses as Disorders of Network Dynamics
This review provides a dynamical systems perspective on psychiatric symptoms and disease, and discusses its potential implications for diagnosis, prognosis, and treatment. After a brief introduction into the theory of dynamical systems, we will focus on the idea that cognitive and emotional functions are implemented in terms of dynamical systems phenomena in the brain, a common assumption in theoretical and computational neuroscience. Specific computational models, anchored in biophysics, for generating different types of network dynamics, and with a relation to psychiatric symptoms, will be briefly reviewed, as well as methodological approaches for reconstructing the system dynamics from observed time series (like fMRI or EEG recordings). We then attempt to outline how psychiatric phenomena, associated with schizophrenia, depression, PTSD, ADHD, phantom pain, and others, could be understood in dynamical systems terms. Most importantly, we will try to convey that the dynamical systems level may provide a central, hub-like level of convergence which unifies and links multiple biophysical and behavioral phenomena, in the sense that diverse biophysical changes can give rise to the same dynamical phenomena and, vice versa, similar changes in dynamics may yield different behavioral symptoms depending on the brain area where these changes manifest. If this assessment is correct, it may have profound implications for the diagnosis, prognosis, and treatment of psychiatric conditions, as it puts the focus on dynamics. We therefore argue that consideration of dynamics should play an important role in the choice and target of interventions

impulsivity and the impact of contextual cues on instrumental behavior in alcohol dependence
Alcohol-related cues acquire incentive salience through Pavlovian conditioning
and then can markedly affect instrumental behavior of alcohol-dependent
patients to promote relapse. However, it is unclear whether similar effects
occur with alcohol-unrelated cues. We tested 116 early-abstinent alcohol-
dependent patients and 91 healthy controls who completed a delay discounting
task to assess choice impulsivity, and a Pavlovian-to-instrumental transfer
(PIT) paradigm employing both alcohol-unrelated and alcohol-related stimuli.
To modify instrumental choice behavior, we tiled the background of the
computer screen either with conditioned stimuli (CS) previously generated by
pairing abstract pictures with pictures indicating monetary gains or losses,
or with pictures displaying alcohol or water beverages. CS paired to money
gains and losses affected instrumental choices differently. This PIT effect
was significantly more pronounced in patients compared to controls, and the
group difference was mainly driven by highly impulsive patients. The PIT
effect was particularly strong in trials in which the instrumental stimulus
required inhibition of instrumental response behavior and the background CS
was associated to monetary gains. Under that condition, patients performed
inappropriate approach behavior, contrary to their previously formed
behavioral intention. Surprisingly, the effect of alcohol and water pictures
as background stimuli resembled that of aversive and appetitive CS,
respectively. These findings suggest that positively valenced background CS
can provoke dysfunctional instrumental approach behavior in impulsive alcohol-
dependent patients. Consequently, in real life they might be easily seduced by
environmental cues to engage in actions thwarting their long-term goals. Such
behaviors may include, but are not limited to, approaching alcohol
Expertise differences in anticipatory judgements during a temporally and spatially occluded dynamic task
There is contradictory evidence surrounding the role of critical cues in the successful anticipation of penalty kick outcome. In the current study, skilled and less-skilled soccer goalkeepers were required to anticipate spatially (full body; hip region) and temporally (–160 ms, –80 ms before, foot–ball contact) occluded penalty kicks. The skilled group outperformed the less-skilled group in all conditions. Both groups performed better in the full body, compared to hip region condition. Later temporal occlusion conditions were associated with increased performance in the correct response and correct side analysis, but not for correct height. These data suggest that there is enough postural information from the hip region for skilled goalkeepers to make highly accurate predictions of penalty kick direction, however, other regions are needed in order to make predictions of height. These data demonstrate the evolution of cues over time and have implications for anticipation training
Cognitive Bias in Ambiguity Judgements:Using Computational Models to Dissect the Effects of Mild Mood Manipulation in Humans
Positive and negative moods can be treated as prior expectations over future delivery of rewards and punishments. This provides an inferential foundation for the cognitive (judgement) bias task, now widely-used for assessing affective states in non-human animals. In the task, information about affect is extracted from the optimistic or pessimistic manner in which participants resolve ambiguities in sensory input. Here, we report a novel variant of the task aimed at dissecting the effects of affect manipulations on perceptual and value computations for decision-making under ambiguity in humans. Participants were instructed to judge which way a Gabor patch (250ms presentation) was leaning. If the stimulus leant one way (e.g. left), pressing the REWard key yielded a monetary WIN whilst pressing the SAFE key failed to acquire the WIN. If it leant the other way (e.g. right), pressing the SAFE key avoided a LOSS whilst pressing the REWard key incurred the LOSS. The size (0-100 UK pence) of the offered WIN and threatened LOSS, and the ambiguity of the stimulus (vertical being completely ambiguous) were varied on a trial-by-trial basis, allowing us to investigate how decisions were affected by differing combinations of these factors. Half the subjects performed the task in a 'Pleasantly' decorated room and were given a gift (bag of sweets) prior to starting, whilst the other half were in a bare 'Unpleasant' room and were not given anything. Although these treatments had little effect on self-reported mood, they did lead to differences in decision-making. All subjects were risk averse under ambiguity, consistent with the notion of loss aversion. Analysis using a Bayesian decision model indicated that Unpleasant Room subjects were ('pessimistically') biased towards choosing the SAFE key under ambiguity, but also weighed WINS more heavily than LOSSes compared to Pleasant Room subjects. These apparently contradictory findings may be explained by the influence of affect on different processes underlying decision-making, and the task presented here offers opportunities for further dissecting such processes
