Scotin, a novel p53-inducible proapoptotic protein located in the ER and the nuclear membrane
p53 is a transcription factor that induces growth arrest or apoptosis in response to cellular stress. To identify new p53-inducible proapoptotic genes, we compared, by differential display, the expression of genes in spleen or thymus of normal and p53 nullizygote mice after γ-irradiation of whole animals. We report the identification and characterization of human and mouse Scotin homologues, a novel gene directly transactivated by p53. The Scotin protein is localized to the ER and the nuclear membrane. Scotin can induce apoptosis in a caspase-dependent manner. Inhibition of endogenous Scotin expression increases resistance to p53-dependent apoptosis induced by DNA damage, suggesting that Scotin plays a role in p53-dependent apoptosis. Study of the Stability of a Selective Solar Absorber Coating under Air and High Temperature Conditions
Abstract Selective solar absorber material is one of the major components in the Concentrated Solar Power (CSP) technologies. Its optical properties determine the efficiency of the energy conversion from the concentrated sun irradiation to the heat recovered by the heat transfer fluid. Development of the CSP requires costs reduction for this technology. To achieve this goal, one way is the increase of the work temperature up to 400 – 500 °C or more. For economic reasons, the bankable of CSP technology requires a lifetime over 25 years for components and so for solar absorbers. The development of solar receiver, able to operate under air, instead of vacuum conditions, is a challenge to reduce the costs. Along the 25 years of a CSP plant lifetime, solar absorber material is daily exposed to high levels of stresses: high solar flux, high thermal gradient. As the oxidation is one of the main factors involved in degradation processes air stability of the absorber coating under high temperatures should be studied as it could be a critical point in the solar field maintainability in case of accidental vacuum loss. In this article we present a durability study of selective solar absorber from Archimede Solar Energy (ASE) exposed in air at high temperature (up to 500 °C). The results show a good stability of this materials up to 450 °C during 3000h without change of solar absorptance and emittance. Variability of serum markers of erythropoiesis during 6 days of racing in highly trained cyclists
The athlete biological passport for the fight against doping is currently based on longitudinal monitoring for abnormal changes in cellular blood parameters. Serum parameters related to altered erythropoiesis could be considered for inclusion in the passport. The aim of this study was to quantify the changes in such parameters in athletes during a period of intense exercise.
12 highly trained cyclists tapered for 3 days before 6 days of simulated intense stage racing. Morning and afternoon blood samples were taken on most days and analysed for total protein, albumin, soluble transferrin receptor and ferritin concentrations. Plasma volume was determined via total haemoglobin mass measured by carbon-monoxide rebreathing. Percent changes in means from baseline and percent standard errors of measurement (analytical error plus intra-athlete variation) on each measurement occasion were estimated with mixed linear modelling of log-transformed measures.
Means of all variables changed substantially in the days following the onset of racing, ranging from −13% (haemoglobin concentration) to +27% (ferritin). After the second day, errors of measurement were generally twice those at baseline.
Plasma variables were affected by heavy exercise, either because of changes in plasma volume (total protein, albumin, haemoglobin), acute phase/inflammatory reactions (ferritin) or both (soluble transferrin receptor). These effects need to be taken into consideration when integrating a plasma parameter into the biological passport model for athletes
Correction, update, and enhancement of vectorial forestry road maps using ALS data, a pathfinder, and seven metrics
Accurate information about forestry roads is a key aspect of forest management in terms of economy (e.g. accessibility, cost, optimal path) and ecology (e.g. wildfire and wildlife protection). In Canada, and in fact, globally, most provincial, state or territory governments maintain vectorial information on the forestry roads under their jurisdiction. However, official maps are not always accurate, may lack road attributes of interest and are not always up-to-date. Airborne Laser Scanning (ALS) has become an established technology to accurately characterize and map broad territories by providing high density 3D point-clouds with, at least, 3 or 4 measurements per square meter.
This paper addresses the problem of the automatic updating, fixing, and enhancement of vectorial forestry road maps over large landscapes (¿10000 km2). For this purpose, we developed a production ready, documented and open-source software. From metrics derived from the point-cloud the method produces a raster of road probability. It then uses an existing, inaccurate, map of the road network to define approximate start and end points for each road. Then, a pathfinder retrieves the accurate road shape by computing the least cost path between the two points on the probability raster. Using the accurate road position given by the algorithm, road width and road state are then estimated based the on characteristics of the point-cloud. We demonstrate that our algorithm retrieves the centrelines of roads in a natively vectorial form with an error below 3 m in 95% of the roads using a fully automatic method. The accuracy of the road location allows us to derive other accurate measurements, including the state of the roads
Monoclonal antibodies against human astrocytomas and their reactivity pattern
The establishment of hybridomas after fusion of X63-Ag8.653 mouse myeloma cells and splenocytes from mice hyperimmunized against human astrocytomas is presented. The animals were primed with 5 × 106 chemically modified uncultured or cultured glioma cells. Six weeks after the last immunization step an intrasplenal booster injection was administrated and 3 days later the spleen cells were prepared for fusion experiments. According to the specificity analysis of the generated antibodies 7 hybridoma products (MUC 7-22, MUC 8-22, MUC 10-22, MUC 11-22, MUC 14-22, MUC 15-22 and MUC 2-63) react with gliomas, neuroblastomas and melanomas as well as with embryonic and fetal cells but do not recognize non-neurogenic tumors. The selected monoclonal antibodies (McAbs) of IgG1 and IgG2a isotypes are not extensively characterized but these antibodies have been demonstrated to be reactive with a panel of glioma cell lines with varying patterns of antigen distribution. Using the McAbs described above and a series of cryosections of glioma biopsies and paraffin sections of the same material as well as glioma cultures established from these, variable antigenic profiles among glioma cell populations could be demonstrated. From these results it is evident that there is not only a distinct degree of antigenic heterogeneity among and within brain tumors, but also that the pattern of antigenic expression can change continuously. Some of the glioma associated antigens recognized by the selected antibodies persist after fixation with methanol/acetone and Karnovsky's fixative and probably are oncoembryonic/oncofetal antigen(s). The data suggest that the use of McAbs recognizing tumor associated oncofetal antigens in immunohistochemistry facilitates objective typing of intracranial malignancies and precise analysis of fine needle brain/tumor biopsies in a sensitive and reproducible manner
Yin and yang, or peas in a pod? Individual-sport versus team-sport athletes and altitude training
The question of whether altitude training can enhance subsequent sea-level performance has been well investigated over many decades. However, research on this topic has focused on athletes from individual or endurance sports, with scant number of studies on team-sport athletes. Questions that need to be answered include whether this type of training may enhance team-sport athlete performance, when success in team-sport is often more based on technical and tactical ability rather than physical capacity per se.
This review will contrast and compare athletes from two sports representative of endurance (cycling) and team-sports (soccer). Specifically, we draw on the respective competition schedules, physiological capacities, activity profiles and energetics of each sport to compare the similarities between athletes from these sports and discuss the relative merits of altitude training for these athletes. The application of conventional live-high, train-high; live-high, train-low; and intermittent hypoxic training for team-sport athletes in the context of the above will be presented. When the above points are considered, we will conclude that dependent on resources and training objectives, altitude training can be seen as an attractive proposition to enhance the physical performance of team-sport athletes without the need for an obvious increase in training load
KINEMATIC DIFFERENCES BETWEEN ‘ONE-FOOTED’ AND ‘TWO-FOOTED’ YOUNG SOCCER PLAYERS KICKING WITH THE NON-PREFERRED LEG
The purpose of this study was to examine kinematic differences between ‘one-footed’ and ‘two-footed’ players when kicking with the non-preferred leg at a target and with maximal effort. Eighteen highly-trained young soccer players were categorised as one-footed (n=9) and two-footed (n=9) based on results of a kicking test. Motion analysis data showed that two-footed players run-up straighter and have less pelvic rotation at ball-foot impact than one-footed players and the differences are likely to be meaningful (ES differences of 0.89 and 0.99 respectively). Run-up angle and pelvic rotation angle are significantly correlated (P < 0.1). The study found that two-footed players are significantly smaller in stature than one-footed players (P < 0.1). Practical implications for soccer coaches arose from the study
Preferences and skills of Indian public sector teachers
With a sample of 700 future public sector primary teachers in India, a Discrete Choice Experiment is used to measure job preferences, particularly regarding location. General skills are also tested. Urban origin teachers and women are more averse to remote locations than rural origin teachers and men respectively. Women would require a 26-73 percent increase in salary for moving to a remote location. The results suggest that existing caste and gender quotas can be detrimental for hiring skilled teachers willing to work in remote locations. The most preferred location is home, which supports decentralised hiring, although this could compromise skills
A mixed method multi-Country assessment of barriers to implementing pediatric inpatient care guidelines
Introduction: Accelerating progress in reducing child deaths is needed in order to achieve the Sustainable Development Goal child mortality target. This will require a focus on vulnerable children-including young children, those who are undernourished or with acute illnesses requiring hospitalization. Improving adherence to inpatient guidelines may be an important strategy to reduce child mortality, including among the most vulnerable. The aim of our assessment of nine sub-Saharan African and South Asian hospitals was to determine adherence to pediatric inpatient care recommendations, in addition to capacity for and barriers to implementation of guideline-adherent care prior to commencing the Childhood Acute Illness and Nutrition (CHAIN) Cohort study. The CHAIN Cohort study aims to identify modifiable risk factors for poor inpatient and post discharge outcomes above and beyond implementation of guidelines.Methods: Hospital infrastructure, staffing, durable equipment, and consumable supplies such as medicines and laboratory reagents, were evaluated through observation and key informant interviews. Inpatient medical records of 2-23 month old children were assessed for adherence to national and international guidelines. The records of children with severe acute malnutrition (SAM) were oversampled to reflect the CHAIN study population. Seven core adherence indicators were examined: oximetry and oxygen therapy, fluids, anemia diagnosis and transfusion, antibiotics, malaria testing and antimalarials, nutritional assessment and management, and HIV testing.Results: All sites had facilities and equipment necessary to implement care consistent with World Health Organization and national guidelines. However, stockouts of essential medicines and laboratory reagents were reported to be common at some sites, even though they were mostly present during the assessment visits. Doctor and nurse to patient ratios varied widely. We reviewed the notes of 261 children with admission diagnoses of sepsis (17), malaria (47), pneumonia (70), diarrhea (106), and SAM (119); 115 had multiple diagnoses. Adherence to oxygen therapy, antimalarial, and malnutrition refeeding guidelines was \u3e75%. Appropriate antimicrobials were prescribed for 75% of antibiotic-indicative conditions. However, 20/23 (87%) diarrhea and 20/27 (74%) malaria cases without a documented indication were prescribed antibiotics. Only 23/122 (19%) with hemoglobin levels meeting anemia criteria had recorded anemia diagnoses. HIV test results were infrequently documented even at hospitals with universal screening policies (66/173, 38%). Informants at all sites attributed inconsistent guideline implementation to inadequate staffing.Conclusion: Assessed hospitals had the infrastructure and equipment to implement guideline-consistent care. While fluids, appropriate antimalarials and antibiotics, and malnutrition refeeding adherence was comparable to published estimates from low- and high-resource settings, there were inconsistencies in implementation of some other recommendations. Stockouts of essential therapeutics and laboratory reagents were a noted barrier, but facility staff perceived inadequate human resources as the primary constraint to consistent guideline implementation
