    Chloroplasts morphology investigation with diverse microscopy approaches and inter-specific variation in Laurencia species (Rhodophyta)

    The present study described with different microscopy approaches chloroplasts lobes in Laurencia sensu latu (Rhodophyta) species and found inter-specific differences among them. Chloroplasts were investigated using confocal laser scanning microscopy (LSM), transmission electron microscopy (TEM) and high resolution scanning electron microscopy (HRSEM). Using and TEM and HRSEM images we distinguished chloroplasts with lobes than chloroplasts without lobes in Yuzurua poiteaui var. gemmifera (Harvey) M. J. Wynne and Laurencia dendroidea J. Agardh cortical cells. The LSM images showed chloroplasts lobes (CLs) with different morphologies, varying from thicker and longer undulated projections in Y. poiteaui var. and L. dendroidea to very small and thin tubules as in Laurencia translucida Fujii & Cordeiro-Marino. The diameter and length of CLs from Y poiteaui var. and L. dendroidea were significantly higher than L. translucida CLs (p \u3c 0.01). Based on LSM observations, we suggest that lobes morphology has a taxonomic validity only to characterize L. translucida species

    Switching from a protease inhibitor-based regimen to a dolutegravir-based regimen : a randomized clinical trial to determine the effect on peripheral blood and ileum biopsies from antiretroviral therapy-suppressed human immunodeficiency virus-infected individuals

    Background: Optimization of combination antiretroviral therapy (cART) can impact the human immunodeficiency virus (HIV) reservoir. We evaluated the effect on the HIV reservoir in peripheral blood and ileum biopsies in patients switching from boosted protease inhibitor (PI/r)-based therapy to dolutegravir (DTG)-based therapy. Methods: Impact of Integrase-inhibitor DOlutegravir On the viral Reservoir (INDOOR) is a phase 4 open-label clinical trial that randomly included 42 HIV type 1-infected individuals on effective cART: 20 who switched from PI/r-based to DTG-based cART (switch group), and 22 who remained in PI/r-based regimens (control group). We analyzed blood and ileum biopsies to quantify episomal, total, and integrated HIV DNA, cell-associated HIV RNA, residual plasma viremia, T-cell subsets, cell activation, and inflammation markers. Results: There were no related adverse events or treatment discontinuations due to drug intolerance. The HIV reservoir was consistently larger in ileal than in peripheral CD4(+) T cells in both groups (P <.01). Residual viremia in plasma decreased in the switch group (P =.03). However, we did not observe significant longitudinal changes in low-level viral replication, total and integrated HIV reservoir, HIV transcription, T-cell maturation subsets, immunoactivation markers, inflammatory soluble proteins, or cellular markers of latently infected cells. Conclusions: The INDOOR study is the first evaluation of changes in HIV reservoir size in ileum biopsies and in peripheral blood in individuals switched from PI/r- to DTG-based cART. Although this switch was safe and well tolerated, it had no impact on a large array of immunological and inflammatory markers or on HIV reservoir markers in peripheral or in ileal CD4(+) T cells

    La detección del sedentarismo en adultos mediante instrumentos sencillos y útiles es un objetivo primordial tanto en salud pública como en el ámbito clínico, ya que este factor de riesgo es una de las causas más importantes de enfermedad no transmisible en el mundo y es muy prevalente en las sociedades desarrolladas como la española. Se describen 2 instrumentos validados, sencillos y útiles para detectar y/o manejar el sedentarismo en adultos: a) el cuestionario internacional de actividad física en su versión corta, y b) el podómetro, para cuantificar el número de pasos realizados en un día. Aumentar los niveles de actividad física es importante para la prevención primaria de algunas enfermedades crónicas (cardiopatía coronaria, diabetes tipo 2, osteoporosis, cáncer de colon) y para mejorar la calidad de vida. El personal sanitario debe conocer el grado de motivación y la disponibilidad del paciente y su familia para el cambio de conducta hacia la actividad física. Las actividades físicas de intensidad moderada no presentan prácticamente ninguna contraindicación y los riesgos son muy escasos.The detection of physical inactivity in adults, using simple and useful tools is primary objective in both public health and in clinical settings, since this risk factor is one of the major causes of non-communicable disease in the world, and is very prevalent in developed societies such as in Spain. Two validated instruments are described that are simple and useful for detecting and/or monitoring physical inactivity in adults: (i) the international physical activity questionnaire in its short version, and (ii) the pedometer to measure the number of steps taken in a day. Increased levels of physical activity are important for the primary prevention of some chronic diseases (coronary heart disease, type 2 diabetes, osteoporosis, colon cancer) and to improve the quality of life. Medical personnel must determine the motivation level and the availability of patients and their families to change their behavior towards physical activity. Moderate-intensity physical activities have hardly any contraindications and the risks are few

    Sensitive quantification of the HIV-1 reservoir in gut-associated lymphoid tissue

    Background: The implementation of successful strategies to achieve an HIV cure has become a priority in HIV research. However, the current location and size of HIV reservoirs is still unknown since there are limited tools to evaluate HIV latency in viral sanctuaries such as gut-associated lymphoid tissue (GALT). As reported in the so called "Boston Patients", despite undetectable levels of proviral HIV-1 DNA in blood and GALT, viral rebound happens in just few months after ART interruption. This fact might imply that current methods are not sensitive enough to detect residual reservoirs. Showing that, it is imperative to improve the detection and quantification of HIV-1 reservoir in tissue samples. Herein, we propose a novel non-enzymatic protocol for purification of Lamina Propria Leukocytes (LPL) from gut biopsies combined to viral HIV DNA (vDNA) quantification by droplet digital PCR (ddPCR) to improve the sensitivity and accuracy of viral reservoir measurements (LPL-vDNA assay). Methods: Endoscopic ileum biopsies were sampled from 12 HIV-1-infected cART-suppressed subjects. We performed a DTT/EDTA-based treatment for epithelial layer removal followed by non-enzymatic disruption of the tissue to obtain lamina propria cell suspension (LP). CD45+ cells were subsequently purified by flow sorting and vDNA was determined by ddPCR. Results: vDNA quantification levels were significantly higher in purified LPLs (CD45+) than in bulk LPs (p<0.01). The levels of vDNA were higher in ileum samples than in concurrent PBMC from the same individuals (p = 0.002). As a result of the increased sensitivity of this purification method, the Poisson 95% confidence intervals of the vDNA quantification data from LPLs were narrower than that from bulk LPs. Of note, vDNA was unambiguously quantified above the detection limit in 100% of LPL samples, while only in 58% of bulk LPs. Conclusion: We propose an innovative combined protocol for a more sensitive detection of the HIV reservoir in gut-associated viral sanctuaries, which might be used to evaluate any proposed eradication strategy

    Estilos docentes y discentes: consideraciones pedagógicas a la luz de la neurociencia

    Este estudio se puede registrar dentro de una matriz disciplinaria que intenta mostrar sus posibilidades, en el presente, en el marco de una nueva ciencia pedagógica. Se trata de una matriz neuropedagógica que exige, a la luz de la neurociencia, más atención para los fenómenos neuroeducativos. Si el proceso educativo tiene que ver con una formación integral del ser humano, entonces la pedagogía no puede renunciar, como ciencia, a los importantes avances derivados de la investigación en neurociencia en este momento, precisamente porque esos avances, o logros, están ayudando para ampliar tal educación / entrenamiento humano. Entre estos logros, no puede olvidarse algo como la asimetría cerebral, la especialización hemisférica ... y, especialmente, la preferencia individual por percibir la realidad externa que es objeto de conocimiento. De hecho, estudios recientes muestran que algunas personas prefieren abordar el procesamiento de información a través del canal visual (hemisferio derecho) mientras que los otros prefieren hacerlo a través del canal auditivo (hemisferio izquierdo). Este último punto de vista fue adoptado por la pedagogía tradicional y verbalista, contribuyendo de esta manera y desde las aulas al desarrollo de las personas unidimensionales en sus relaciones de aprendizaje. Esta es la razón por la cual defendemos un modelo holístico de educación/enseñanza. Un modelo que tenga en cuenta las estrategias de enseñanza compensatoria; es decir, estrategias correctivas respecto a la tendencia espontánea en cada maestro dependiendo de su propio orzuelo de aprendizaje

    Single-shot d-scan technique for ultrashort laser pulse characterization using transverse second-harmonic generation in random nonlinear crystals

    We demonstrate a novel dispersion-scan (d-scan) scheme for single-shot temporal characterization of ultrashort laser pulses. The novelty of this method relies on the use of a highly dispersive crystal featuring antiparallel nonlinear domains with a random distribution and size. This crystal, capable of generating a transverse second-harmonic signal, acts simultaneously as the dispersive element and the nonlinear medium of the d-scan device. The resulting in-line architecture makes the technique very simple and robust, allowing the acquisition of single-shot d-scan traces in real time. The retrieved pulses are in very good agreement with independent frequency-resolved optical grating measurements. We also apply the new single-shot d-scan to a terawatt-class laser equipped with a programmable pulse shaper, obtaining an excellent agreement between the applied and the d-scan retrieved dispersions

    Origin and fate of a bloom of Skeletonema costatum during a winter upwelling/downwelling sequence in the Ría de Vigo (NW Spain)

    Original research paperThe onset, development and decay of a winter bloom of the marine diatom Skeletonema costatum was monitored during a 10 d period in the coastal upwelling system of the Rı´a de Vigo (NW Spain). The succession of upwelling, relaxation and downwelling-favorable coastal winds with a frequency of 10 –20 d is a common feature of the NW Iberian shelf. The onset of the bloom occurred during an upwelling-favorable 1⁄2 wk period under winter thermal inversion conditions. The subsequent 1⁄2 wk coastal wind relaxation period allowed development of the bloom (gross primary production reached 8gCm–2 d–1) utilizing nutrients upwelled during the previous period.     Gene Expression Profiling and Association Studies Implicate the Neuregulin Signaling Pathway in Behçet's Disease Susceptibility

    Behçet's disease (BD) is a complex disease with genetic and environmental risk factors implicated in its etiology; however, its pathophysiology is poorly understood. To decipher BD's genetic underpinnings, we combined gene expression profiling with pathway analysis and association studies. We compared the gene expression profiles in peripheral blood mononuclear cells (PBMCs) of 15 patients and 14 matched controls using Affymetrix microarrays and found that the neuregulin signaling pathway was over-represented among the differentially expressed genes. The Epiregulin (EREG), Amphiregulin (AREG), and Neuregulin-1 (NRG1) genes of this pathway stand out as they are also among the top differentially expressed genes. Twelve haplotype tagging SNPs at the EREG-AREG locus and 15 SNPs in NRG1 found associated in at least one published BD genome-wide association study were tested for association with BD in a dataset of 976 Iranian patients and 839 controls. We found a novel association with BD for the rs6845297 SNP located downstream of EREG, and replicated three associations at NRG1 (rs4489285, rs383632, and rs1462891). Multifactor dimensionality reduction analysis indicated the existence of epistatic interactions between EREG and NRG1 variants. EREG-AREG and NRG1, which are members of the epidermal growth factor (EGF) family, seem to modulate BD susceptibility through main effects and gene–gene interactions. These association findings support a role for the EGF/ErbB signaling pathway inBD pathogenesis that warrants further investigation and highlight the importance of combining genetic and genomic approaches to dissect the genetic architecture of complex diseases
