    How Small Polar Molecules Protect Membrane Systems against Osmotic Stress: The Urea−Water−Phospholipid System

    We investigate how a small polar molecule, urea, can act to protect a phospholipid bilayer system against osmotic stress. Osmotic stress can be caused by a dry environment, by freezing, or by exposure to aqueous systems with high osmotic pressure due to solutes like in saline water. A large number of organisms regularly experience osmotic stress, and it is a common response to produce small polar molecules intracellularly. We have selected a ternary system of urea-water-dimyristoyl phosphatidylcholine (DMPC) as a model to investigate the molecular mechanism behind this protective effect, in this case, of urea, and we put special emphasis on the applications of urea in skin care products. Using differential scanning calorimetry, X-ray diffraction, and sorption microbalance measurements, we studied the phase behavior of lipid systems exposed to an excess of solvent of varying compositions, as well as lipid systems exposed to water at reduced relative humidities. From this, we have arrived at a rather detailed thermodynamic characterization. The basic findings are as follows: (i) In excess solvent, the thermally induced lipid phase transitions are only marginally dependent on the urea content, with the exception being that the P phase is not observed in the presence of urea. (ii) For lipid systems with limited access to solvent, the phase behavior is basically determined by the amount (volume) of solvent irrespective of the urea content. (iii) The presence of urea has the effect of retaining the liquid crystalline phase at relative humidities down to 64% (at 27 °C), whereas, in the absence of urea, the transition to the gel phase occurs already at a relative humidity of 94%. This demonstrates the protective effect of urea against osmotic stress. (iv) In skin care products, urea is referred to as a moisturizer, which we find slightly misleading as it replaces the water while keeping the physical properties unaltered. (v) In other systems, urea is known to weaken the hydrophobic interactions, while for the lipid system we find few signs of this loosening of the strong segregation into polar and apolar regions on addition of ure

    Drug Transport in Responding Lipid Membranes Can Be Regulated by an External Osmotic Gradient

    In this paper, we demonstrate, for the first time, how an external osmotic gradient can be used to regulate diffusion of solutes across a lipid membrane. We present experimental and theoretical studies of the transport of different solutes across a monoolein membrane in the presence of an external osmotic gradient. The osmotic gradient introduces phase transformations in the membrane, and it causes nonlinear transport behavior. The external gradient can thus act as a kind of switch for diffusive transport in the skin and in controlled release drug formulations

    Renewed global partnerships and redesigned roadmaps for rabies prevention and control

    Canine rabies, responsible for most human rabies deaths, is a serious global public health concern. This zoonosis is entirely preventable, but by focusing solely upon rabies prevention in humans, this "incurable wound" persists at high costs. Although preventing human deaths through canine rabies elimination is feasible, dog rabies control is often neglected, because dogs are not considered typical economic commodities by the animal health sector. Here, we demonstrate that the responsibility of managing rabies falls upon multiple sectors, that a truly integrated approach is the key to rabies elimination, and that considerable progress has been made to this effect. Achievements include the construction of global rabies networks and organizational partnerships; development of road maps, operational toolkits, and a blueprint for rabies prevention and control; and opportunities for scaling up and replication of successful programs. Progress must continue towards overcoming the remaining challenges preventing the ultimate goal of rabies elimination

    Galaxy luminosity evolution: how much is due to a model choice?

    The cluster and field luminosity functions (LFs) determined on large homogeneous samples (N>2200 galaxies each) are almost indistinguishable, down to M^*+4 in the r and i filters, hence suggesting that the effect of the cluster environment on the galaxy properties does not affect the galaxy luminosity function in red bands. The similarity of the red band LFs in different environments suggests that the galaxy mass function is preserved during the galaxy infall in the cluster. By analyzing a large sample of galaxies in clusters, ideal from many points of view (multicolor data, large size, many clusters, metric magnitudes) we found that luminosity evolution is required by the data but the latter do not unambiguously derive its flavour if a differential luminosity evolution between bright and faint galaxies is allowed. We show that the LF parameters (slope, characteristic magnitude and their evolution) and errors depend on assumptions in a way seldom recognized in literature. We also point out logical inconsistencies between hypothesis assumed in deriving literature LF and presented results, suggesting caution in interpreting similar published results.Comment: A&A, in pres

    Synthesis and characterization of copolymers of alkyl- and azo-thiophenes: chromic properties and photoinduced birefringence

    Polyazothiophene is a type of polythiophene derivative that combines the electrical and luminescent properties of polythiophenes with the photoisomerization property of azopolymers. Extensive efforts have been made to improve the properties of polyazothiophenes, such as solubility, optical, and chromic properties. We report the preparation of copolymers of an alkylthiophene (3-octylthiophene, 3-OT) and an azothiophene (2-[N-ethyl-N-[4-[(4-(4-nitrophenyl)azo]phenyl]amino]ethyl 3-thienylacetate, 3-AzoT) in different ratios as an alternative route to improve these properties. The azosubstituted monomer contents in these copolymers were 6, 9, 12, and 51% (in mol), as evaluated by elemental analysis. Their chemical structures were confirmed by FTIR and 1H-NMR. HPSEC and thermal analysis were used to characterize the polymers. The presence of thermochromic and solvatochromic properties was demonstrated by UV-Vis spectroscopy. Optically induced birefringence was detected only in polymers with 12 and 51 mol % of azo-units. The introduction of different ratios of the azothiophene in the copolymer alters the polymer solubility and emissive properties.     Mouse Papillomavirus L1 and L2 Are Dispensable for Viral Infection and Persistence at Both Cutaneous and Mucosal Tissues.

    Papillomavirus L1 and L2, the major and minor capsid proteins, play significant roles in viral assembly, entry, and propagation. In the current study, we investigate the impact of L1 and L2 on viral life cycle and tumor growth with a newly established mouse papillomavirus (MmuPV1) infection model. MmuPV1 L1 knockout, L2 knockout, and L1 plus L2 knockout mutant genomes (designated as L1ATGko-4m, L2ATGko, and L1-L2ATGko respectively) were generated. The mutants were examined for their ability to generate lesions in athymic nude mice. Viral activities were examined by qPCR, immunohistochemistry (IHC), in situ hybridization (ISH), and transmission electron microscopy (TEM) analyses. We demonstrated that viral DNA replication and tumor growth occurred at both cutaneous and mucosal sites infected with each of the mutants. Infections involving L1ATGko-4m, L2ATGko, and L1-L2ATGko mutant genomes generally resulted in smaller tumor sizes compared to infection with the wild type. The L1 protein was absent in L1ATGko-4m and L1-L2ATGko mutant-treated tissues, even though viral transcripts and E4 protein expression were robust. Therefore, L1 is not essential for MmuPV1-induced tumor growth, and this finding parallels our previous observations in the rabbit papillomavirus model. Very few viral particles were detected in L2ATGko mutant-infected tissues. Interestingly, the localization of L1 in lesions induced by L2ATGko was primarily cytoplasmic rather than nuclear. The findings support the hypothesis that the L2 gene influences the expression, location, transport, and assembly of the L1 protein in vivo

    Stellar populations of early-type galaxies in different environments I. Line-strength indices

    Aims: This paper commences a series devoted to the study of the stellar content of early-type galaxies. The goal of the series is to set constraints on the evolutionary status of these objects. Methods: In this paper we describe the details of the galaxy sample, the observations, and the data reduction. Line-strength indices and velocity dispersions sigma are measured in 98 early-type galaxies drawn from different environments, and the relation of the indices with the velocity dispersion analysed in detail. Results: The present sample indicates that some of the index-sigma relations depend on galaxy environment. In particular, the slope of the relation between Balmer lines and sigma is steeper for galaxies in the Virgo cluster, small groups, and in the field than for galaxies in the Coma cluster. In several indices there is also a significant offset in the zero point between the relations defined by the different subsamples. The slopes of the index-sigma relation for the Virgo and low-density environment galaxies are explained by a variation of both age and metallicity with velocity dispersion, as previously noted in other studies. For the galaxies in the Coma cluster, however, the relation of the indices with sigma only requires a variation of the abundance along the sigma sequence. In agreement with other studies we find that the models that better reproduce the slopes are those in which the alpha elements vary more than the Fe-peak elements along the sigma sequence, while, at a given sigma, older galaxies show an higher alpha/Fe ratio. Conclusions: The results can be explained assuming that galaxies in the Coma cluster have experienced a truncated star formation and chemical enrichment history compared to a more continuous time-extended history for their counterparts in lower density environments.Comment: accepted for publication in A&

    The Structure of 2MASS Galaxy Clusters

    We use a sample of galaxies from the Two Micron All Sky Survey (2MASS) Extended Source Catalog to refine a matched filter method of finding galaxy clusters that takes into account each galaxy's position, magnitude, and redshift if available. The matched filter postulates a radial density profile, luminosity function, and line-of-sight velocity distribution for cluster galaxies. We use this method to search for clusters in the galaxy catalog, which is complete to an extinction-corrected K-band magnitude of 13.25 and has spectroscopic redshifts for roughly 40% of the galaxies, including nearly all brighter than K = 11.25. We then use a stacking analysis to determine the average luminosity function, radial distribution, and velocity distribution of cluster galaxies in several richness classes, and use the results to update the parameters of the matched filter before repeating the cluster search. We also investigate the correlations between a cluster's richness and its velocity dispersion and core radius, using these relations to refine priors that are applied during the cluster search process. After the second cluster search iteration, we repeat the stacking analysis. We find a cluster galaxy luminosity function that fits a Schechter form, with parameters M_K* - 5 log h = -23.64\pm0.04 and \alpha = -1.07\pm0.03. We can achieve a slightly better fit to our luminosity function by adding a Gaussian component on the bright end to represent the brightest cluster galaxy (BCG) population. The radial number density profile of galaxies closely matches a projected Navarro-Frenk-White (NFW) profile at intermediate radii, with deviations at small radii due to well-known cluster centering issues and outside the virial radius due to correlated structure.     COVID-19 Vaccination of Individuals with Down Syndrome—Data from the Trisomy 21 Research Society Survey on Safety, Efficacy, and Factors Associated with the Decision to Be Vaccinated

    Individuals with Down syndrome (DS) are among the groups with the highest risk for severe COVID-19. Better understanding of the efficacy and risks of COVID-19 vaccines for individuals with DS may help improve uptake of vaccination. The T21RS COVID-19 Initiative launched an international survey to obtain information on safety and efficacy of COVID-19 vaccines for individuals with DS. De-identified survey data collected between March and December 2021 were analyzed. Of 2172 individuals with DS, 1973 (91%) had received at least one vaccine dose (57% BNT162b2), 107 (5%) were unvaccinated by choice, and 92 (4%) were unvaccinated for other reasons. Most participants had either no side effects (54%) or mild ones such as pain at the injection site (29%), fatigue (12%), and fever (7%). Severe side effects occurred in <0.5% of participants. About 1% of the vaccinated individuals with DS contracted COVID-19 after vaccination, and all recovered. Individuals with DS who were unvaccinated by choice were more likely to be younger, previously recovered from COVID-19, and also unvaccinated against other recommended vaccines. COVID-19 vaccines have been shown to be safe for individuals with DS and effective in terms of resulting in minimal breakthrough infections and milder disease outcomes among fully vaccinated individuals with DS

    A Review of Direct Neck Measurement in Occupational Settings

    No guidelines are available to orient researchers on the availability and applications of equipment and sensors for recording precise neck movements in occupational settings. In this study reports on direct measurements of neck movements in the workplace were reviewed. Using relevant keywords two independent reviewers searched for eligible studies in the following databases: Cinahal, Cochrane, Embase, Lilacs, PubMed, MEDLINE, PEDro, Scopus and Web of Science. After applying the inclusion criteria, 13 articles on direct neck measurements in occupational settings were retrieved from among 33,666 initial titles. These studies were then methodologically evaluated according to their design characteristics, exposure and outcome assessment, and statistical analysis. The results showed that in most of the studies the three axes of neck movement (flexion-extension, lateral flexion and rotation) were not simultaneously recorded. Deficiencies in available equipment explain this flaw, demonstrating that sensors and systems need to be improved so that a true understanding of real occupational exposure can be achieved. Further studies are also needed to assess neck movement in those who perform heavy-duty work, such as nurses and electricians, since no report about such jobs was identified
