    RadioAstron Space VLBI Imaging of the jet in M87: I. Detection of high brightness temperature at 22 GHz

    We present results from the first 22 GHz space very-long-baseline interferometric (VLBI) imaging observations of M87 by RadioAstron. As a part of the Nearby AGN Key Science Program, the source was observed in Feb 2014 at 22 GHz with 21 ground stations, reaching projected (u,v)(u,v)-spacings up to 11\sim11\,Gλ\lambda. The imaging experiment was complemented by snapshot RadioAstron data of M87 obtained during 2013--2016 from the AGN Survey Key Science Program. Their longest baselines extend up to 25\sim25\,Gλ\lambda. For all these measurements, fringes are detected only up to \sim2.8 Earth Diameter or \sim3 Gλ\lambda baseline lengths, resulting in a new image with angular resolution of 150μ\sim150\,\muas or 20\sim20 Schwarzschild radii spatial resolution. The new image not only shows edge-brightened jet and counterjet structures down to submilliarcsecond scales but also clearly resolves the VLBI core region. While the overall size of the core is comparable to those reported in the literature, the ground-space fringe detection and slightly super-resolved RadioAstron image suggest the presence of substructures in the nucleus, whose minimum brightness temperature exceeds TB,min1012T_{\rm B, min}\sim10^{12}\,K. It is challenging to explain the origin of this record-high TB,minT_{\rm B, min} value for M87 by pure Doppler boosting effect with a simple conical jet geometry and known jet speed. Therefore, this can be evidence for more extreme Doppler boosting due to a blazar-like small jet viewing angle or highly efficient particle acceleration processes occurring already at the base of the outflow.Comment: 27 pages, 13 figures, accepted for publication in Ap

    Genomic and ecogenomic characterisation of Proteus mirabilis bacteriophage

    Proteus mirabilis often complicates the care of catheterized patients through the formation of crystalline biofilms which block urine flow. Bacteriophage therapy has been highlighted as a promising approach to control this problem, but relatively few phages infecting P. mirabilis have been characterized. Here we characterize five phages capable of infecting P. mirabilis, including those shown to reduce biofilm formation, and provide insights regarding the wider ecological and evolutionary relationships of these phages. Transmission electron microscopy (TEM) imaging of phages vB_PmiP_RS1pmA, vB_PmiP_RS1pmB, vB_PmiP_RS3pmA, and vB_PmiP_RS8pmA showed that all share morphologies characteristic of the Podoviridae family. The genome sequences of vB_PmiP_RS1pmA, vB_PmiP_RS1pmB, and vB_PmiP_RS3pmA showed these are species of the same phage differing only by point mutations, and are closely related to vB_PmiP_RS8pmA. Podophages characterized in this study were also found to share similarity in genome architecture and composition to other previously described P. mirabilis podophages (PM16 and PM75). In contrast, vB_PimP_RS51pmB showed morphology characteristic of the Myoviridae family, with no notable similarity to other phage genomes examined. Ecogenomic profiling of all phages revealed no association with human urinary tract viromes, but sequences similar to vB_PimP_RS51pmB were found within human gut, and human oral microbiomes. Investigation of wider host-phage evolutionary relationships through tetranucleotide profiling of phage genomes and bacterial chromosomes, indicated vB_PimP_RS51pmB has a relatively recent association with Morganella morganii and other non-Proteus members of the Morganellaceae family.     A Putative P-Type ATPase Required for Virulence and Resistance to Haem Toxicity in Listeria monocytogenes

    Regulation of iron homeostasis in many pathogens is principally mediated by the ferric uptake regulator, Fur. Since acquisition of iron from the host is essential for the intracellular pathogen Listeria monocytogenes, we predicted the existence of Fur-regulated systems that support infection. We examined the contribution of nine Fur-regulated loci to the pathogenicity of L. monocytogenes in a murine model of infection. While mutating the majority of the genes failed to affect virulence, three mutants exhibited a significantly compromised virulence potential. Most striking was the role of the membrane protein we designate FrvA (Fur regulated virulence factor A; encoded by frvA [lmo0641]), which is absolutely required for the systemic phase of infection in mice and also for virulence in an alternative infection model, the Wax Moth Galleria mellonella. Further analysis of the ΔfrvA mutant revealed poor growth in iron deficient media and inhibition of growth by micromolar concentrations of haem or haemoglobin, a phenotype which may contribute to the attenuated growth of this mutant during infection. Uptake studies indicated that the ΔfrvA mutant is unaffected in the uptake of ferric citrate but demonstrates a significant increase in uptake of haem and haemin. The data suggest a potential role for FrvA as a haem exporter that functions, at least in part, to protect the cell against the potential toxicity of free haem

    Overview of the coordinated ground-based observations of Titan during the Huygens mission

    Coordinated ground-based observations of Titan were performed around or during the Huygens atmospheric probe mission at Titan on 14 January 2005, connecting the momentary in situ observations by the probe with the synoptic coverage provided by continuing ground-based programs. These observations consisted of three different categories: (1) radio telescope tracking of the Huygens signal at 2040 MHz, (2) observations of the atmosphere and surface of Titan, and (3) attempts to observe radiation emitted during the Huygens Probe entry into Titan's atmosphere. The Probe radio signal was successfully acquired by a network of terrestrial telescopes, recovering a vertical profile of wind speed in Titan's atmosphere from 140 km altitude down to the surface. Ground-based observations brought new information on atmosphere and surface properties of the largest Satumian moon. No positive detection of phenomena associated with the Probe entry was reported. This paper reviews all these measurements and highlights the achieved results. The ground-based observations, both radio and optical, are of fundamental imnortance for the interpretatinn of results from the Huygens mission
