New <i>V. cholerae</i> atypical El Tor variant emerged during the 2006 epidemic outbreak in Angola
Background
V. cholerae is the etiological agent of cholera, a major public health concern in most developing countries. Virulence of V. cholerae relies on the powerful cholera toxin, encoded by the CTX prophage. The emergence of new pathogenic variants in the recent years has been mostly associated with new CTX prophage rearrangements.
Results
In this retrospective study, we show that the epidemic V. cholerae O1 El Tor strain responsible for the 2006 outbreak in Angola is clonally and genetically different from El Tor strains circulating in the 1990s in the same area. Strains from 2006 carry ICEVchAng3 of the SXT/R391 family. This ICE is associated with a narrower multidrug resistance profile compared to the one conferred by plasmid p3iANG to strains of the 1990s. The CTX prophage carried by 2006 El Tor strains is characterized by rstRET and ctxBCla alleles organized in a RS1-RS2-Core array on chromosome I. Interestingly, the newly emerging atypical strain belongs to a clade previously known to comprise only clinical isolates from the Indian subcontinent that also contain the same ICE of the SXT/R391 family.
Conclusions
Our findings remark the appearance of a novel V. cholerae epidemic variant in Africa with a new CTXΦ arrangement previously described only in the Indian Subcontinent.</br
Identification of <i>Trichomonas vaginalis</i> alpha-actinin as the most common immunogen recognized by sera of women exposed to the parasite
A study on presence of antibodies to Trichomonis vaginalis in serum was done on a group of 500 pregnant, asymptomatic Angolan women. A serologic screening, done by ELISA, revealed that 41% of the women had IgG and IgM against the parasite. Analysis of sera by immunoblotting revealed that 94.4% of sera with anti-T. vaginalis IgG class antibodies were reactive against a common immunogenic protein of 115 kDa. The common immunogen was identified as the protozoan α-actinin. All sera recognizing the 115-kDa antigen were reactive against both native and recombinant T. vaginalis α-actinin and nonreactive against human α-actinin. The findings presented in this work offer a new tool for epidemiologic studies and open new perspectives for vaccination

As diarreias continuam a ser em Moçambique e em muitos paises
em desenvolvimento um dos problemas de Saúde pública mais importantes.
São doenças transmissiveis não preveniveis por vacina eque
ocorrem com frequencia de forma isolada ou associada à malnutrição
e malaria causando elevadas taxas de mortalidade nos países em
desenvolvimento.
Para cada caso foi escolhido um controle constituido por indivíduos
do mesmo sexo e idade que o casos, e residentes na mesma area,
sem historia de dejecções liquidas nos ultimos 60 dias anteriores á
data de recolha de dados.
A área de estudo foi o Bairro do Xipamanine. Motivou a escolha
desta zona o facto de este bairro ter durante anos consecutivos registado
o maior número de casos das diarreias ocorridas na Cidade de
Maputo. De acordo com dados da Direcção de Saude da Cidade do
Maputo, em 1996, por exemplo, registaram-se 34.095 casos de diarrea
nesta zona, dos quais 25.406 (75%) em crianças do 0-14 anos
Ribavirin Impairs Salivary gland function During Combination Treatment With Pegylated Interferon Alfa-2a In HEpatitis C patients
Background: Xerostomia is a common adverse event of unknown etiology observed during pegylated interferon (PegIFN)/Ribavirin (Rbv) treatment.
Objectives: To assess the frequency and mechanisms of xerostomia during PegIFN/Rbv therapy.
Patients and Methods: Thirty-one naive patients with chronic hepatitis C consecutively received PegIFN-alpha 2a (180 mu g/week) plus Rbv (800-1200 mg/day). The controls were 10 patients with chronic hepatitis B who received PegIFN-alpha 2a (180 mu g/week). During treatment and follow-up, all patients underwent basal and masticatory stimulated sialometry, otorhinolaryngoiatric (ORL) examination, and a questionnaire survey to subjectively assess symptoms of oral dryness.
Results: Twenty-seven patients on PegIFN/Rbv and 4 on PegIFN (87% vs. 40%, P = 0.006) reported xerostomia. Thirty patients on PegIFN/Rbv combination therapy and 2 patients on monotherapy had ORL signs of salivary gland hypofunction (97% vs. 20%, P < 0.0001). Mean basal (A) and stimulated (B) salivary flow rates (mL/min) progressively decreased during PegIFN/Rbv treatment (A, 0.49 at baseline vs. 0.17 at the end of treatment, P < 0.0001; B,1.24 at baseline vs. 0.53 at the end of treatment, P = 0.0004). At week 24 following PegIFN/Rbv treatment, salivary flow rates were similar to baseline (A, 0.53 at the end of follow-up vs. 0.49 at baseline; B,1.19 at the end of follow-up vs. 1.24 at baseline). Salivary function was unaffected in monotherapy patients.
Conclusions: Rbv causes salivary gland hypofunction in hepatitis C patients receiving PegIFN/Rbv therapy, which promptly reverts to normal upon cessation of treatment
Chronic cholesterol administration to the brain supports complete and long-lasting cognitive and motor amelioration in Huntington's disease
: Evidence that Huntington's disease (HD) is characterized by impaired cholesterol biosynthesis in the brain has led to strategies to increase its level in the brain of the rapidly progressing R6/2 mouse model, with a positive therapeutic outcome. Here we tested the long-term efficacy of chronic administration of cholesterol to the brain of the slowly progressing zQ175DN knock-in HD mice in preventing ("early treatment") or reversing ("late treatment") HD symptoms. To do this we used the most advanced formulation of cholesterol loaded brain-permeable nanoparticles (NPs), termed hybrid-g7-NPs-chol, which were injected intraperitoneally. We show that one cycle of treatment with hybrid-g7-NPs-chol, administered in the presymptomatic ("early treatment") or symptomatic ("late treatment") stages is sufficient to normalize cognitive defects up to 5 months, as well as to improve other behavioral and neuropathological parameters. A multiple cycle treatment combining both early and late treatments ("2 cycle treatment") lasting 6 months generates therapeutic effects for more than 11 months, without severe adverse reactions. Sustained cholesterol delivery to the brain of zQ175DN mice also reduces mutant Huntingtin aggregates in both the striatum and cortex and completely normalizes synaptic communication in the striatal medium spiny neurons compared to saline-treated HD mice. Furthermore, through a meta-analysis of published and current data, we demonstrated the power of hybrid-g7-NPs-chol and other strategies able to increase brain cholesterol biosynthesis, to reverse cognitive decline and counteract the formation of mutant Huntingtin aggregates. These results demonstrate that cholesterol delivery via brain-permeable NPs is a therapeutic option to sustainably reverse HD-related behavioral decline and neuropathological signs over time, highlighting the therapeutic potential of cholesterol-based strategies in HD patients. Sex-specific predictors of PCSK9 levels in a European population:The IMPROVE study
Background and aims:
Proprotein convertase subtilisin/kexin type 9 (PCSK9) is one of the key regulators of low-density lipoprotein cholesterol plasma levels and circulating PCSK9, which differs between genders. PCSK9 represents a valid pharmacological target for preventing cardiovascular (CV) events. We aimed to investigate sex-related associations between PCSK9 plasma levels and biochemical and anthropomorphic factors, and familial and personal morbidities, in a large European cohort (n=3,673) of men (47.9%) and women (52.1%).
Methods:
Individuals (aged 54 to 79 years) free of CV diseases were enrolled in 7 centers of five European countries: Finland, France, Italy, the Netherlands, and Sweden. PCSK9 plasma levels were measured by ELISA.
Results:
PCSK9 was higher in women than in men. Multiple linear regression analysis showed that latitude, sex, and treatments with statins and fibrates were the strongest predictors of PCSK9 in the whole group. These variables, together with triglycerides and high-density lipoprotein cholesterol, were also associated with PCSK9 in men or women. Mean corpuscular hemoglobin concentration and pack-years were PCSK9 independent predictors in women, whereas hypercholesterolemia and physical activity were independent predictors in men. The associations between PCSK9 and latitude, uric acid, diabetes, hypercholesterolemia and physical activity were significantly different in men and women (pinteraction <0.05 for all).
Conclusions:
Besides confirming the association with lipids in the whole group, our study revealed previously unknown differences in PCSK9 predictors in men and women. These might be taken into account when defining individual risk for CV events and/or for refining PCSK9 lowering treatments
Roadmap on thermoelectricity
The increasing energy demand and the ever more pressing need for clean technologies of energy conversion pose one of the most urgent and complicated issues of our age. Thermoelectricity, namely the direct conversion of waste heat into electricity, is a promising technique based on a long-standing physical phenomenon, which still has not fully developed its potential, mainly due to the low efficiency of the process. In order to improve the thermoelectric performance, a huge effort is being made by physicists, materials scientists and engineers, with the primary aims of better understanding the fundamental issues ruling the improvement of the thermoelectric figure of merit, and finally building the most efficient thermoelectric devices.
In this Roadmap an overview is given about the most recent experimental and computational results obtained within the Italian research community on the optimization of composition and morphology of some thermoelectric materials, as well as on the design of thermoelectric and hybrid thermoelectric/photovoltaic devices
Ecosistemi per la ricerca Atti Convegno ACNP/NILDE Trieste, 22-23 maggio 2014
Il secondo convegno congiunto ACNP / NILDE: ecosistemi per la ricerca è stato
ospitato dal 22 al 23 maggio 2014 dall’Università di Trieste.
Sotto gli auspici della stessa Università di Trieste e degli altri enti di ricerca
del Friuli Venezia Giulia1 sono stati affrontati in un’ottica internazionale i temi
del rapporto tra cataloghi collettivi e servizi interbibliotecari, e il più generale
ambito dei servizi bibliografici per la ricerca scientifica.
ACNP e NILDE possono essere considerati un vero e proprio ecosistema. Le
biblioteche e i bibliotecari collaborano tra di loro in maniera reciproca e secondo
modalità interconnesse, offrendo agli utenti servizi sempre più evoluti e dinamici.
Questo ecosistema, essendo aperto, mette i propri servizi a disposizione della
ricerca scientifica in senso generale. Il convegno di Trieste ha offerto l’occasione
di investigare e proporre soluzioni innovative, interconnessioni e relazioni nuove
e più proficue.
Il convegno ha presentato alcune rilevanti esperienze internazionali in tema
di servizi interbibliotecari e cataloghi collettivi e la prosecuzione di attività che erano state proposte come spunti di ispirazione nel convegno precedente2 inoltre
si sono condotte delle riflessioni sulle nuove esigenze dell’utenza.
Una ultima parte è dedicata alla illustrazione degli sviluppi tecnici e le prospettive
future di ACNP e di NILDE.
Hanno partecipato all’evento oltre 200 colleghi italiani e stranieri ed i relatori
dei 18 contributi provenivano da Italia, Germania, Austria, Grecia, Slovenia e
Stati Uniti. Il carattere di confronto e di condivisione delle esperienze tipico della
realtà partecipativa di ACNP e NILDE, è emerso anche in questa occasione nella
tavola rotonda - di cui viene riportato un resoconto dettagliato - che ha affrontato
il tema della valutazione della ricerca dal punto di vista delle biblioteche. Inoltre,
la molto partecipata sessione poster ha ospitato 13 lavori- anch’essi riportati nel
volume - incentrati oltre che su ACNP e NILDE anche sul tema ricerca e sistema
biblioteca, dando ottimi spunti di partecipazione, dialogo e confronto sulle diverse
realtà in cui operiamo
MiR-320a as a Potential Novel Circulating Biomarker of Arrhythmogenic CardioMyopathy
Diagnosis of Arrhythmogenic CardioMyopathy (ACM) is challenging and often late after disease onset. No circulating biomarkers are available to date. Given their involvement in several cardiovascular diseases, plasma microRNAs warranted investigation as potential non-invasive diagnostic tools in ACM. We sought to identify circulating microRNAs differentially expressed in ACM with respect to Healthy Controls (HC) and Idiopathic Ventricular Tachycardia patients (IVT), often in differential diagnosis. ACM and HC subjects were screened for plasmatic expression of 377 microRNAs and validation was performed in 36 ACM, 53 HC, 21 IVT. Variable importance in data partition was estimated through Random Forest analysis and accuracy by Receiver Operating Curves. Plasmatic miR-320a showed 0.53\u2009\ub1\u20090.04 fold expression difference in ACM vs. HC (p\u2009<\u20090.01). A similar trend was observed when comparing ACM (n\u2009=\u200913) and HC (n\u2009=\u200917) with athletic lifestyle, a ACM precipitating factor. Importantly, ACM patients miR-320a showed 0.78\u2009\ub1\u20090.05 fold expression change vs. IVT (p\u2009=\u20090.03). When compared to non-invasive ACM diagnostic parameters, miR-320a ranked highly in discriminating ACM vs. IVT and it increased their accuracy. Finally, miR-320a expression did not correlate with ACM severity. Our data suggest that miR-320a may be considered a novel potential biomarker of ACM, specifically useful in ACM vs. IVT differentiation
T follicular helper cells promote a beneficial gut ecosystem for host metabolic homeostasis by sensing microbiota-derived extracellular ATP
The ATP-gated ionotropic P2X7 receptor regulates T follicular helper (Tfh) cell abundance in the Peyer’s patches (PPs) of the small intestine; deletion of P2rx7, encoding for P2X7, in Tfh cells results in enhanced IgA secretion and binding to commensal bacteria. Here, we show that Tfh cell activity is important for generating a diverse bacterial community in the gut and that sensing of microbiota-derived extracellular ATP via P2X7 promotes the generation of a proficient gut ecosystem for metabolic homeostasis. The results of this study indicate that Tfh cells play a role in host-microbiota mutualism beyond protecting the intestinal mucosa by induction of affinity-matured IgA and suggest that extracellular ATP constitutes an inter-kingdom signaling molecule important for selecting a beneficial microbial community for the host via P2X7-mediated regulation of B cell help
