    Mesospheric observations with the EISCAT UHF radar during polar cap absorption events: <br>3. Comparison with simultaneous EISCAT VHF measurements

    International audienceMesospheric observations were obtained by the EISCAT UHF and VHF radars during the solar proton event of March 1990. We present the first comparison of incoherent-scatter spectral measurements from the middle mesosphere using simultaneous, co-located observations by the two radars. VHF spectra observed with a vertical antenna were found to be significantly narrower than model predictions, in agreement with earlier UHF results. For antenna pointing directions that were significantly away from the vertical, the wider VHF radar beam gave rise to broadening of the observed spectra due to vertical shears in the horizontal wind. In this configuration, UHF spectral measurements were found to be more suitable for aeronomical applications. Both radar systems provide consistent and reliable estimates of the neutral wind.     Research methodology is the procedural framework within which the research is conducted. This includes the overall approach to a problem that could be put into practice in a research process, from the theoretical underpinning to the collection and analysis of data. Choice of methodology depends on the primary drivers: topic to be researched and the specific research questions. Hence, methodological perspectives of managing stakeholder expectations of PDHR context are composed of research philosophies, research strategy, research design, and research techniques. This research belonged to social constructivism or interpretivism within a philosophical continuum. The nature of the study was more toward subjectivism where human behavior favored voluntary stance. Ontological, methodological, epistemological, and axiological positioning carried the characteristics of idealism, ideographic, anti-positivism, and value laden, respectively. Data collection comprises two phases, preliminary and secondary. Exploratory interviews with construction experts in the United Kingdom and Sri Lanka were carried out to refine the interview questions and identify the case studies. Case study interviews during the secondary phase took place in Sri Lanka. Data collected at the preliminary stage were used to assess the attributes of power, legitimacy/proximity, and urgency of stakeholders to the project using Stakeholder Circle™ software. Moreover, the data collected at secondary phase via case studies will be analyzed with NVivo 8. This article aims to discuss these methodological underpinnings in detail applied in a post-disaster housing reconstruction context in Sri Lanka

    Seismo-acoustic ray model benchmarking against experimental tank data

    Acoustic predictions of the recently developed TRACEO ray model, which accounts for bottom shear properties, are benchmarked against tank experimental data from the EPEE-1 and EPEE-2 (Elastic Parabolic Equation Experiment) experiments. Both experiments are representative of signal propagation in a Pekeris-like shallow-water waveguide over a non-flat isotropic elastic bottom, where significant interaction of the signal with the bottom can be expected. The benchmarks show, in particular, that the ray model can be as accurate as a parabolic approximation model benchmarked in similar conditions. The results of benchmarking are important, on one side, as a preliminary experimental validation of the model and, on the other side, demonstrates the reliability of the ray approach for seismo-acoustic applications.     Identification of clouds and aurorae in optical data images

    In this paper we present an automatic image recognition technique used to identify clouds and aurorae in digital images, taken with a CCD all-sky imager. The image recognition algorithm uses image segmentation to generate a binary block object image. Object analysis is then performed on the binary block image, the results of which are used to assess whether clouds, aurorae and stars are present in the original image. The need for such an algorithm arises because the optical study of particle precipitation into the Earth's atmosphere by the Ionosphere and Radio Propagation Group at Lancaster generates vast data-sets, over 25 000 images/year, making manual classification of all the images impractical

    Maximising Social Interactions and Effectiveness within Distance Learning Courses: Cases from Construction

    Advanced Internet technologies have revolutionised the delivery of distance learning education. As a result, the physical proximity between learners and the learning providers has become less important. However, whilst the pervasiveness of these technological developments has reached unprecedented levels, critics argue that the student learning experience is still not as effective as conventional face-to-face delivery. In this regard, surveys of distance learning courses reveal that there is often a lack of social interaction attributed to this method of delivery, which tends to leave learners feeling isolated due to a lack of engagement, direction, guidance and support by the tutor. This paper defines and conceptualises this phenomenon by investigating the extent to which distance-learning programmes provide the social interactions of an equivalent traditional classroom setting. In this respect, two distance learning case studies were investigated, covering the UK and Slovenian markets respectively. Research findings identified that delivery success is strongly dependent on the particular context to which the specific distance learning course is designed, structured and augmented. It is therefore recommended that designers of distance learning courses should balance the tensions and nuances associated with commercial viability and pedagogic effectiveness

    Goal-oriented a posteriori error estimation for the travel time functional in porous media flows

    In this article we consider the a posteriori error estimation and adaptive mesh refinement for the numerical approximation of the travel time functional arising in porous media flows. The key application of this work is in the safety assessment of radioactive waste facilities; in this setting, the travel time functional measures the time taken for a non-sorbing radioactive solute, transported by groundwater, to travel from a potential site deep underground to the biosphere. To ensure the computability of the travel time functional, we employ a mixed formulation of Darcy's law and conservation of mass, together with Raviart-Thomas H(div) conforming finite elements. The proposed a posteriori error bound is derived based on a variant of the standard Dual-Weighted-Residual approximation, which takes into account the lack of smoothness of the underlying functional of interest. The proposed adaptive refinement strategy is tested on both a simple academic test case and a problem based on the geological units found at the Sellafield site in the UK

    Can BioSAXS detect ultrastructural changes of antifungal compounds in Candida albicans?–an exploratory study

    The opportunistic yeast Candida albicans is the most common cause of candidiasis. With only four classes of antifungal drugs on the market, resistance is becoming a problem in the treatment of fungal infections, especially in immunocompromised patients. The development of novel antifungal drugs with different modes of action is urgent. In 2016, we developed a groundbreaking new medium-throughput method to distinguish the effects of antibacterial agents. Using small-angle X-ray scattering for biological samples (BioSAXS), it is now possible to screen hundreds of new antibacterial compounds and select those with the highest probability for a novel mode of action. However, yeast (eukaryotic) cells are highly structured compared to bacteria. The fundamental question to answer was if the ultrastructural changes induced by the action of an antifungal drug can be detected even when most structures in the cell stay unchanged. In this exploratory work, BioSAXS was used to measure the ultrastructural changes of C. albicans that were directly or indirectly induced by antifungal compounds. For this, the well-characterized antifungal drug Flucytosine was used. BioSAXS measurements were performed on the synchrotron P12 BioSAXS beamline, EMBL (DESY, Hamburg) on treated and untreated yeast C. albicans. BioSAXS curves were analysed using principal component analysis (PCA). The PCA showed that Flucytosine-treated and untreated yeast were separated. Based on that success further measurements were performed on five antifungal peptides {1. Cecropin A-melittin hybrid [CA (1–7) M (2–9)], KWKLFKKIGAVLKVL; 2. Lasioglossin LL-III, VNWKKILGKIIKVVK; 3. Mastoparan M, INLKAIAALAKKLL; 4. Bmkn2, FIGAIARLLSKIFGKR; and 5. optP7, KRRVRWIIW}. The ultrastructural changes of C. albicans indicate that the peptides may have different modes of action compared to Flucytosine as well as to each other, except for the Cecropin A-melittin hybrid [CA (1–7) M (2–9)] and optP7, showing very similar effects on C. albicans. This very first study demonstrates that BioSAXS shows promise to be used for antifungal drug development. However, this first study has limitations and further experiments are necessary to establish this application

    Determinants of voluntary audit and voluntary full accounts in micro- and non-micro small companies in the UK

It examines decisions made in connection with the 2006 accounts following UK's adoption of the maximum EU size thresholds in 2004, and the impact of the proposed Directive on the annual accounts of micro-companies. The research extends the model of cost, management and agency factors associated with voluntary audit, and develops a complementary model for voluntary full accounts. The results show the benefits of placing full audited accounts on public record that outweigh the costs for a significant proportion of companies. In non-micro small companies, voluntary audit is determined by cost and agency factors, whereas in micro-companies it is driven by cost, management and agency factors. In both groups, the predictors of voluntary full accounts include management and agency factors, and choosing voluntary audit is one of the key factors. The study provides models that can be tested in other jurisdictions to provide evidence of the needs of micro-companies, and the discussion of the methodological challenges for small company researchers in the UK makes further contribution to the literature

    Inverse Correlation between Promoter Strength and Excision Activity in Class 1 Integrons

    Class 1 integrons are widespread genetic elements that allow bacteria to capture and express gene cassettes that are usually promoterless. These integrons play a major role in the dissemination of antibiotic resistance among Gram-negative bacteria. They typically consist of a gene (intI) encoding an integrase (that catalyzes the gene cassette movement by site-specific recombination), a recombination site (attI1), and a promoter (Pc) responsible for the expression of inserted gene cassettes. The Pc promoter can occasionally be combined with a second promoter designated P2, and several Pc variants with different strengths have been described, although their relative distribution is not known. The Pc promoter in class 1 integrons is located within the intI1 coding sequence. The Pc polymorphism affects the amino acid sequence of IntI1 and the effect of this feature on the integrase recombination activity has not previously been investigated. We therefore conducted an extensive in silico study of class 1 integron sequences in order to assess the distribution of Pc variants. We also measured these promoters' strength by means of transcriptional reporter gene fusion experiments and estimated the excision and integration activities of the different IntI1 variants. We found that there are currently 13 Pc variants, leading to 10 IntI1 variants, that have a highly uneven distribution. There are five main Pc-P2 combinations, corresponding to five promoter strengths, and three main integrases displaying similar integration activity but very different excision efficiency. Promoter strength correlates with integrase excision activity: the weaker the promoter, the stronger the integrase. The tight relationship between the aptitude of class 1 integrons to recombine cassettes and express gene cassettes may be a key to understanding the short-term evolution of integrons. Dissemination of integron-driven drug resistance is therefore more complex than previously thought
