    The determinants of Teachers’ Wellbeing: the mediating role of Mental Fatigue

    In recent years, many studies have focused on the determinants and consequences of teacher stress. One of the most recent theoretical models concerning stress is the Job Demands-Resources (JD-R) model. This study examines one process – namely the energetic process – which supposes that high job demands exhaust employees’ mental and physical resources and therefore cause ill health. Particularly, this study examines the mediating role of mental fatigue between three job demands of teachers (workload, inequity and work/family confl ict) and three consequences of stress: psychological and physical symptoms and work satisfaction. An Italian version of the selfreport Questionnaire for Psychosocial Work Environment and Stress (PWSQ) was administered to 697 teachers belonging to a random sample of 17 school organizations. The results showed that mental fatigue has a mediating role in the relationship between work/family confl ict and the three outcomes considered, namely psychological and physical symptoms, and work satisfaction. This study has an important implication for intervention strategy because mental fatigue is a pre-strain condition which arises before more intense outcomes, such as stress-related diseases occur.     Tackling job insecurity : can a boundaryless career orientation boost job crafting strategies and career competencies?

    none4siThe present study was aimed at investigating the mediating role played by job crafting and career competencies (i.e., knowing why, knowing how, and knowing whom) within the negative relation between boundaryless career orientation and job insecurity. A sample of 267 Italian employees fulfilled an online self-report questionnaire. Results of bootstrapping models with multiple mediators operating in serial indicated that boundaryless career orientation was negatively related to job insecurity through the subsequent mediation first of job crafting, and then of career competencies.     The aim of the present study is to identify the psychosocial factors that are more related to the risk of workplace bullying in the organizational contexts. According to the work environment hypothesis, job demands, job control, perception of equity and leadership have been considered. It has been hypothesized that all these variables infl uence directly negative acts and also their effects are moderated by perception of colleagues. In order to investigate these relationships, a self-report questionnaire has been administered to 148 participants who represented all the population of a structure of private health care. The main results showed that job demands and leadership are the organizational antecedents more related to workplace bullying. Furthermore, the perception of colleagues has a moderating effect especially between job characteristics, like job demand, and negative acts, while this effect is weaker between relational antecedent, as leadership, and bullying. The data seem to con- fi rm that mobbing prevention can be implemented starting from the psychosocial work environment and management of relationships involving leader and colleagues.El objetivo del presente estudio es identificar cuáles son los factores psicosociales que se  asocian a un mayor riesgo de acoso en los contextos laborales. En particular, en línea con la hipótesis situacional se tuvieron en cuenta la demanda laboral, el control, la percepción de equidad y el liderazgo. Específicamente se plantea una influencia directa de estos factores sobre las acciones negativas perpetradas en los lugares de trabajo y un efecto moderador de las percepciones de los colegas entre los factores considerados y el riesgo de acoso laboral. El estudio, llevado a cabo en una institución privada hospitalaria, incluyó la aplicación de un cuestionario de auto-reporte a todos los trabajadores de la organización (N = 148). Los resultados han evidenciado que son, sobre todo, la demanda laboral y el liderazgo los que influencian directamente las acciones negativas, mientras que el efecto es más débil entre los antecedentes de carácter más relacional, como el liderazgo, y el acoso laboral. Los datos confirman que la prevención del acoso laboral debe ser implementada a partir del ambiente psicosocial de trabajo y de la gestión de las relaciones que impliquen a los colegas y a los superiores

    C-peptide: a predictor of cardiovascular mortality in subjects with established atherosclerotic disease

    Aim: Insulin resistance and type 2 diabetes are independent risk factors for cardiovascular diseases. Levels of C-peptide are increased in these patients and its role in the atherosclerosis progression was studied in vitro and in vivo over the past years. To evaluate the possible use of C-peptide as cardiovascular biomarkers, we designed an observational study in which we enrolled patients with mono- or poly-vascular atherosclerotic disease. Methods: We recruited 431 patients with stable atherosclerosis and performed a yearly follow-up to estimate the cardiovascular and total mortality and cardiovascular events. Results: We performed a mean follow-up of 56months on 268 patients. A multivariate Cox analysis showed that C-peptide significantly increased the risk of cardiovascular mortality [Hazard Ratio: 1.29 (95% confidence interval: 1.02-1.65, p<0.03513)] after adjustment for age, sex, diabetes treatment, estimated glomerular filtration rate and known diabetes status. Furthermore, levels of C-peptide were significantly correlated with metabolic parameters and atherogenic factors. Conclusion: C-peptide was associated with cardiovascular mortality independently of known diabetes status in a cohort of patients with chronic atherosclerotic disease. Future studies using C-peptide into a reclassification approach might be undertaken to consider its potential as a cardiovascular disease biomarker

    Effects on human transcriptome of mutated BRCA1 BRCT domain: A microarray study

    BACKGROUND: BRCA1 (breast cancer 1, early onset) missense mutations have been detected in familial breast and ovarian cancers, but the role of these variants in cancer predisposition is often difficult to ascertain. In this work, the molecular mechanisms affected in human cells by two BRCA1 missense variants, M1775R and A1789T, both located in the second BRCT (BRCA1 C Terminus) domain, have been investigated. Both these variants were isolated from familial breast cancer patients and the study of their effect on yeast cell transcriptome has previously provided interesting clues to their possible role in the pathogenesis of breast cancer. METHODS: We compared by Human Whole Genome Microarrays the expression profiles of HeLa cells transfected with one or the other variant and HeLa cells transfected with BRCA1 wild-type. Microarray data analysis was performed by three comparisons: M1775R versus wild-type (M1775RvsWT-contrast), A1789T versus wild-type (A1789TvsWT-contrast) and the mutated BRCT domain versus wild-type (MutvsWT-contrast), considering the two variants as a single mutation of BRCT domain. RESULTS: 201 differentially expressed genes were found in M1775RvsWT-contrast, 313 in A1789TvsWT-contrast and 173 in MutvsWT-contrast. Most of these genes mapped in pathways deregulated in cancer, such as cell cycle progression and DNA damage response and repair. CONCLUSIONS: Our results represent the first molecular evidence of the pathogenetic role of M1775R, already proposed by functional studies, and give support to a similar role for A1789T that we first hypothesized based on the yeast cell experiments. This is in line with the very recently suggested role of BRCT domain as the main effector of BRCA1 tumor suppressor activity

    Abstract Peripheral blood mononuclear cells (PBMCs) derived from a healthy 40-year-old female were successfully transformed into induced pluripotent stem cells (iPSCs) by using the integration-free CytoTune-iPS Sendai Reprogramming method. The resulting iPSCs line exhibits a normal karyotype, expresses stemness markers and displays the differentiation capacity into the three germ layers. This human iPSCs line can be used as healthy control in disease modelling studies

    Human Mutated MYOT and CRYAB Genes Cause a Myopathic Phenotype in Zebrafish

    Myofibrillar myopathies (MFMs) are a group of hereditary neuromuscular disorders sharing common histological features, such as myofibrillar derangement, Z-disk disintegration, and accumulation of degradation products into protein aggregates. They are caused by mutations in several genes that encode either structural proteins or molecular chaperones. Nevertheless, the mechanisms by which mutated genes result in protein aggregation are still unknown. To unveil the role of myotilin and αB-crystallin in the pathogenesis of MFM, we injected zebrafish fertilized eggs at one-cell stage with expression plasmids harboring cDNA sequences of human wildtype or mutated MYOT (p.Ser95Ile) and human wildtype or mutated CRYAB (p.Gly154Ser). We evaluated the effects on fish survival, motor behavior, muscle structure and development. We found that transgenic zebrafish showed morphological defects that were more severe in those overexpressing mutant genes which developed a myopathic phenotype consistent with that of human myofibrillar myopathy including the formation of protein aggregates. Results indicate that pathogenic mutations in myotilin and αB-crystallin genes associated with MFM cause a structural and functional impairment of the skeletal muscle in zebrafish, thereby making this non-mammalian organism a powerful model to dissect disease pathogenesis and find possible druggable targets

    Italian Validation of the 12-Item Version of the Burnout Assessment Tool (BAT-12)

    The Burnout Assessment Tool (BAT) has shown satisfactory validity evidence in several countries, with the 23-item version of the instrument reporting adequate psychometric properties also in the Italian context. This paper is aimed to present results from the Italian validation of the 12-item version of the BAT. Based on a sample of 2277 workers, our results supported the factorial validity of a higher-order model represented by 4 first-order factors corresponding to the core dimensions of burnout, namely exhaustion, mental distance, and emotional and cognitive impairment. The measure invariance of the BAT-12 between data collected before and during the COVID-19 pandemic was supported. However, ANCOVA results suggest a higher score on the second-order burnout factor on data collected during the COVID-19 pandemic in comparison with earlier data. In line with the JD-R model, the BAT-12 total score reported a positive association with job demands (i.e., workload, time pressure, and role conflict) and a negative association with job resources (i.e., job autonomy, coworkers' support) and personal resources (i.e., optimism, social self-efficacy, and task self-efficacy). Additionally, the BAT-12 showed a negative association with work engagement components (i.e., vigor, dedication, and absorption) and positive job attitudes (i.e., job satisfaction, affective commitment). All in all, our results identify the Italian version of the BAT-12 as a brief and reliable tool for measuring burnout among workers

    Structural Plasticity of Dopaminergic Neurons Requires the Activation of the D3R-nAChR Heteromer and the PI3K-ERK1/2/Akt-Induced Expression of c-Fos and p70S6K Signaling Pathway

    We have previously shown that the heteromer composed by the dopamine D3 receptor (D3R) and the nicotinic acetylcholine receptor (nAChR) (D3R-nAChR heteromer) is expressed in dopaminergic neurons, activated by nicotine and represents the molecular unit that, in these neurons, contributes to the modulation of critical events such as structural plasticity and neuroprotection. We now extended this study by investigating the D3R-nAChR heteromer properties using various cell models such as transfected HEK293 cells, primary cultures of mouse dopaminergic neurons and human dopaminergic neurons derived from induced pluripotent stem cells. We found that the D3R-nAChR heteromer is the molecular effector that transduces the remodeling properties not only associated with nicotine but also with D3R agonist stimulation: neither nAChR nor D3R, in fact, when express as monomers, are able to elicit these effects. Moreover, strong and sustained activation of the PI3K-ERK1/2/Akt pathways is coupled with D3R-nAChR heteromer stimulation, leading to the expression of the immediate-early gene c-Fos and to sustained phosphorylation of cytosolic p70 ribosomal S6 kinase (p70S6K), critical for dendritic remodeling. By contrast, while D3R stimulation results in rapid and transient activation of both Erk1/2 and Akt, that is PI3K-dependent, stimulation of nAChR is associated with persistent activation of Erk1/2 and Akt, in a PI3K-independent way. Thus, the D3R-nAChR heteromer and its ability to trigger the PI3K-ERK1/2/Akt signaling pathways may represent a novel target for preserving dopaminergic neurons healthy and for conferring neuronal protection against injuries
