Chemistry in Infrared Dark Cloud Clumps: a Molecular Line Survey at 3 mm
We have observed 37 Infrared Dark Clouds (IRDCs), containing a total of 159
clumps, in high-density molecular tracers at 3 mm using the 22-meter ATNF Mopra
Telescope located in Australia. After determining kinematic distances, we
eliminated clumps that are not located in IRDCs and clumps with a separation
between them of less than one Mopra beam. Our final sample consists of 92 IRDC
clumps. The most commonly detected molecular lines are (detection rates higher
than 8%): N2H+, HNC, HN13C, HCO+, H13CO+, HCN, C2H, HC3N, HNCO, and SiO. We
investigate the behavior of the different molecular tracers and look for
chemical variations as a function of an evolutionary sequence based on Spitzer
IRAC and MIPS emission. We find that the molecular tracers behave differently
through the evolutionary sequence and some of them can be used to yield useful
relative age information. The presence of HNC and N2H+ lines do not depend on
the star formation activity. On the other hand, HC3N, HNCO, and SiO are
predominantly detected in later stages of evolution. Optical depth calculations
show that in IRDC clumps the N2H+ line is optically thin, the C2H line is
moderately optically thick, and HNC and HCO+ are optically thick. The HCN
hyperfine transitions are blended, and, in addition, show self-absorbed line
profiles and extended wing emission. These factors combined prevent the use of
HCN hyperfine transitions for the calculation of physical parameters. Total
column densities of the different molecules, except C2H, increase with the
evolutionary stage of the clumps. Molecular abundances increase with the
evolutionary stage for N2H+ and HCO+. The N2H+/HCO+ and N2H+/HNC abudance
ratios act as chemical clocks, increasing with the evolution of the clumps.Comment: Accepted to ApJ. 29 page
Circulating Senescent T Cells Are Linked to Systemic Inflammation and Lesion Size During Human Cutaneous Leishmaniasis
Leishmania (Viannia) braziliensis induces American tegumentary leishmaniasis that
ranges in severity from the milder form, cutaneous (CL) to severe disseminated
cutaneous leishmaniasis. Patients with CL develop a cell-mediated Th1 immune
response accompanied by production of inflammatory cytokines, which contribute to
parasite control and pathogenesis of disease. Here, we describe the accumulation of
circulating T cells with multiple features of telomere dependent-senescence including
elevated expression of CD57, KLRG-1, and γH2AX that have short telomeres and low
hTERT expression during cutaneous L. braziliensis infection. This expanded population
of T cells was found within the CD45RA+CD27− (EMRA) subset and produced high
levels of inflammatory cytokines, analogous to the senescence-associated secretory
profile (SASP) that has been described in senescent non-lymphoid cells. There was
a significant correlation between the accumulation of these cells and the extent of
systemic inflammation, suggesting that they are involved in the inflammatory response
in this disease. Furthermore, these cells expressed high level of the skin homing
receptor CLA and there was a highly significant correlation between the number of
these cells in the circulation and the size of the Leishmania-induced lesions in the
skin. Collectively our results suggest that extensive activation during the early stages
of leishmaniasis drives the senescence of T cells with the propensity to home to the skin.
The senescence-related inflammatory cytokine secretion by these cells may control the
infection but also contribute to the immunopathology in the disease
The effects of fly-bys on planetary systems
Most of the observed extrasolar planets are found on tight and often
eccentric orbits. The high eccentricities are not easily explained by
planet-formation models, which predict that planets should be on rather
circular orbits. Here we explore whether fly-bys involving planetary systems
with properties similar to those of the gas giants in the solar system, can
produce planets with properties similar to the observed planets. Using
numerical simulations, we show that fly-bys can cause the immediate ejection of
planets, and sometimes also lead to the capture of one or more planets by the
intruder. More common, however, is that fly-bys only perturb the orbits of
planets, sometimes leaving the system in an unstable state. Over time-scales of
a few million to several hundred million years after the fly-by, this
perturbation can trigger planet-planet scatterings, leading to the ejection of
one or more planets. For example, in the case of the four gas giants of the
solar system, the fraction of systems from which at least one planet is ejected
more than doubles in 10^8 years after the fly-by. The remaining planets are
often left on more eccentric orbits, similar to the eccentricities of the
observed extrasolar planets. We combine our results of how fly-bys effect
solar-system-like planetary systems, with the rate at which encounters in young
stellar clusters occur. For example, we measure the effects of fly-bys on the
four gas giants in the solar system. We find, that for such systems, between 5
and 15 per cent suffer ejections of planets in 10^8 years after fly-bys in
typical open clusters. Thus, encounters in young stellar clusters can
significantly alter the properties of any planets orbiting stars in clusters.
As a large fraction of stars which populate the solar neighbourhood form in
stellar clusters, encounters can significantly affect the properties of the

Usability, acceptability, and safety analysis of a computer-tailored web-based exercise intervention (exerciseguide) for individuals with metastatic prostate cancer: Multi-methods laboratory-based study
Background: Digital health interventions such as tailored websites are emerging as valuable tools to provide individualized exercise and behavioral change information for individuals diagnosed with cancer. Objective: The aim of this study is to investigate and iteratively refine the acceptability and usability of a web-based exercise intervention (ExerciseGuide) for men with metastatic prostate cancer and determine how well individuals can replicate the video-based exercise prescription. Methods: A laboratory-based multi-methods design was used, incorporating questionnaires, think-aloud tests, interviews, and movement screening among 11 men aged 63 to 82 years with metastatic prostate cancer. Overall, 9 participants were undergoing androgen deprivation therapy, and 2 were completing chemotherapy. Data were collected in two waves, with changes made for quality improvement after participant 5. Results: The intervention\u27s usability score was deemed moderate overall but improved after modifications (from 60, SD 2.9 to 69.6, SD 2.2 out of 100). Overall, the participants found the intervention acceptable, with scores improving from wave 1 (24.2, SD 1.1 out of 30) to wave 2 (26.3, SD 2.1 out of 30). The personalized multimodal exercise prescription and computer-tailored education were seen as valuable. After wave 1, website navigation videos were added, medical terminology was simplified, and a telehealth component was included after expert real-time telehealth support was requested. Wave 2 changes included the added variety for aerobic exercise modes, reduced computer-tailoring question loads, and improved consistency of style and grammar. Finally, the participants could replicate the resistance exercise videos to a satisfactory level as judged by the movement screen; however, additional technique cueing within the videos is recommended to address safety concerns. Conclusions: The acceptability and usability of ExerciseGuide were deemed satisfactory. Various problems were identified and resolved. Notably, the participants requested the inclusion of personalized expert support through telehealth. The resistance training algorithms were shown to provide appropriate content safely, and the users could replicate the exercise technique unaided to a satisfactory level. This study has optimized the ExerciseGuide intervention for further investigation in this population
