Mechanical alterations in airway smooth muscle after interaction with indoor pollutant - A\ud mathematical model

    Get PDF
The viscoelasticity of mammalian lung is determined by the mechanical properties and structural regulation of the airway smooth muscle (ASM). The exposure to polluted air may deteriorate these properties with harmful consequences to individual health. Formaldehyde (FA) is an important indoor pollutant found among volatile organic compounds. This pollutant permeates through the smooth muscle tissue forming covalent bonds between proteins in the extracellular matrix and intracellular protein structure changing mechanical properties of ASM and inducing asthma symptoms, such as airway hyperresponsiveness, even at low concentrations. In the experimental scenario, the mechanical effect of FA is the stiffening of the tissue, but the mechanism behind this effect is not fully w1derstood. Thus, the aim of this study is to reproduce the mechanical behavior of the ASM, such as contraction and stretching, under FA action or not. For this, it was created a two-dimensional viscoelastic network model based on Voronoi tessellation solved using Runge-Kutta method of fourth order. The equilibrium configuration was reached when the forces in different parts of the network were equal. This model simulates the mechanical behavior of ASM through of a network of dashpots and springs. This dashpot-spring mechanical coupling mimics the composition of the actomyosin machinery of ASM through the contraction of springs to a minimum length. We hypothesized that formation of covalent bonds, due to the FA action, can be represented in the model by a simple change in the elastic constant of the springs, while the action of methacholinc (MCh) reduce the equilibrium length of the spring. A sigmoid curve of tension as a function of MCh doses was obtained, showing increased tension when the muscle strip was exposed to FA. Our simulations suggest that FA, at a concentration of 0.1 ppm, can affect the elastic properties of the smooth muscle fibers by a factor of 120%. We also analyze the dynamic mechanical properties, observing the viscous and elastic behavior of the network. Finally, the proposed model, although simple, ir1corporates the phenomenology of both MCh and FA and reproduces experirnental results observed with ir1 vitro exposure of smooth muscle to .FA. Thus, this new mechanical approach incorporates several well know features of the contractile system of the cells ir1 a tissue level model. The model can also be used in different biological scales.CNPqFAPES

Near-Infrared Imaging Polarimetry of the Serpens Cloud Core: Magnetic Field Structure, Outflows, and Inflows in A Cluster Forming Clump

We made deep NIR imaging polarimetry toward the Serpens cloud core. The polarization vector maps enable us to newly detect 24 small IR reflection nebulae with YSOs. Polarization measurements of NIR point sources indicate an hourglass-shaped magnetic field, of which symmetry axis is nearly perpendicular to the elongation of the C18O (J=1-0) or submillimeter continuum emission. The bright part of C18O (J=1-0), submillimeter continuum cores as well as many class 0/I objects are located just toward the constriction region of the hourglass-shaped magnetic field. Applying the CF method, the magnetic field strength was estimated to be ~100 muG, suggesting that the ambient region of the Serpens cloud core is moderately magnetically supercritical. These suggest that the Serpens cloud core first contracted along the magnetic field to be an elongated cloud, which is perpendicular to the magnetic field, and that then the central part contracted cross the magnetic field due to the high density in the central region of the cloud core, where star formation is actively continuing. Comparison of this magnetic field with the previous observations of molecular gas and large-scale outflows suggests a possibility that the cloud dynamics is controlled by the magnetic field, protostellar outflows and gravitational inflows. This appears to be in good agreement with the outflow-driven turbulence model and implies the importance of the magnetic field to continuous star formation in the center region of the cluster forming region.Comment: 36 pages, 11 figures, 2 tables, accepted for publication in the Astrophysical Journa

Cannabidiol antidepressant-like effect in the lipopolysaccharide model in mice: Modulation of inflammatory pathways

Major Depression is a severe psychiatric condition with a still poorly understood etiology. In the last years, evidence supporting the neuroinflammatory hypothesis of depression has increased. In the current clinical scenario, in which the available treatments for depression is far from optimal, there is an urgent need to develop fast-acting drugs with fewer side effects. In this regard, recent pieces of evidence suggest that cannabidiol (CBD), the major non-psychotropic component of Cannabis sativa with anti-inflammatory properties, appears as a drug with antidepressant properties. In this work, CBD 30 mg/kg was administered systemically to mice 30 min before lipopolysaccharide (LPS; 0.83 mg/kg) administration as a neuroinflammatory model, and behavioral tests for depressive-, anhedonic- and anxious-like behavior were performed. NF-?B, I?B? and PPAR? levels were analyzed by western blot in nuclear and cytosolic fractions of cortical samples. IL-6 and TNF? levels were determined in plasma and prefrontal cortex using ELISA and qPCR techniques, respectively. The precursor tryptophan (TRP), and its metabolites kynurenine (KYN) and serotonin (5-HT) were measured in hippocampus and cortex by HPLC. The ratios KYN/TRP and KYN/5-HT were used to estimate indoleamine 2,3-dioxygenase (IDO) activity and the balance of both metabolic pathways, respectively. CBD reduced the immobility time in the tail suspension test and increased sucrose preference in the LPS model, without affecting locomotion and central activity in the open-field test. CBD diminished cortical NF-?B activation, IL-6 levels in plasma and brain, and the increased KYN/TRP and KYN/5-HT ratios in hippocampus and cortex in the LPS model. Our results demonstrate that CBD produced antidepressant-like effects in the LPS neuroinflammatory model, associated to a reduction in the kynurenine pathway activation, IL-6 levels and NF-?B activation. As CBD stands out as a promising antidepressant drug, more research is needed to completely understand its mechanisms of action in depression linked to inflammation.FUNDING AND ACKNOWLEDGMENTS: This research was supported by the Ministerio de Economía y Competitividad (SAF2015-67457-R MINECO/FEDER), the Ministerio de Ciencia, Innovación y Universidades (RTI2018-097534-B-I00), the Instituto de Salud Carlos III (PI19/00170), and Centro de Investigación Biomédica en Red de Salud Mental (CIBERSAM). AbstractAn important part of the constitutional interpretation arises as a result of the trial of amparo, however this does not imply that the Supreme Court of Justice has built a uniform theory of constitutional rights and freedoms. This text has three purposes: (a) show that speech rights court only has foundation on communitarianism justice or utilitarianism, (b) emphasize that the interpretation of legitimate interest in amparo follows the same fate and (c) noted that the incorporation of that procedural tool in the new model of amparo, can include of an egalitarian liberal view on constitutional justice

    Trace semantics via determinization

This paper takes a fresh look at the topic of trace semantics in the theory of coalgebras. The first development of coalgebraic trace semantics used final coalgebras in Kleisli categories, stemming from an initial algebra in the underlying category (see notably~\cite{HasuoJS07}). This approach requires some non-trivial assumptions, like dcpo enrichment, which do not always hold, even in cases where one can reasonably speak of traces (like for weighted automata). More recently, it has been noticed (see~\cite{SBBR10}) that trace semantics can also arise by first performing a determinization construction. In this paper, we develop a systematic approach, in which the two approaches correspond to different orders of composing a functor and a monad, and accordingly, to different distributive laws. The relevant final coalgebra that gives rise to trace semantics does not live in a Kleisli category, but more generally, in a category of Eilenberg-Moore algebras. In order to exploit its finality, we identify an extension operation, that changes the state space of a coalgebra into a free algebra, which abstractly captures determinization of automata. Notably, we show that the two different views on trace semantics are equivalent, in the examples where both approaches are applicable. We are grateful to the anonymous referees for valuable comments.
