    Garvicins AG1 and AG2 : two novel class IId bacteriocins of lactococcus garvieae Lg-Granada

In silico analysis of plasmid pLG50 of L. garvieae Lg-Granada, an isolate from a patient with endocarditis, revealed the presence of two gene clusters (orf 46–47 and orf 48–49), each one encoding a novel putative bacteriocin, i.e., garvicin AG1 (GarAG1; orf 46) and garvicin AG2 (GarAG2; orf 48), and their corresponding immunity proteins (orf 47 and orf 49). The chemically synthesised bacteriocins GarAG1 and GarAG2 presented inhibitory activity against pathogenic L. garvieae strains, with AG2 also being active against Listeria monocytogenes, Listeria ivanovii and Enterococcus faecalis. Genetic organisation, amino acid sequences and antimicrobial activities of GarAG1 and GarAG2 indicate that they belong to linear non-pediocin-like one-peptide class IId bacteriocins. Gram-positive bacteria that were sensitive to GarAG2 were also able to ferment mannose, suggesting that this bacteriocin could use the mannose phosphotransferase transport system (Man-PTS) involved in mannose uptake as a receptor in sensitive strains. Intriguingly, GarAG1 and GarAG2 were highly active against their own host, L. garvieae Lg-Granada, which could be envisaged as a new strategy to combat pathogens via their own weapons.Publisher PDFPeer reviewe

    E-MIS validity applied to TELMA enhanced learning environment

    Enhanced learning environments are arising with great success within the field of cognitive skills training in minimally invasive surgery (MIS) because they provides multiple benefits since they avoid time, spatial and cost constraints. TELMA [1,2] is a new technology enhanced learning platform that promotes collaborative and ubiquitous training of surgeons. This platform is based on four main modules: an authoring tool, a learning content and knowledge management system, an evaluation module and a professional network. TELMA has been designed and developed focused on the user; therefore it is necessary to carry out a user validation as final stage of the development. For this purpose, e-MIS validity [3] has been defined. This validation includes usability, contents and functionality validities both for the development and production stages of any e-Learning web platform. Using e-MIS validity, the e-Learning is fully validated since it includes subjective and objective metrics. The purpose of this study is to specify and apply a set of objective and subjective metrics using e-MIS validity to test usability, contents and functionality of TELMA environment within the development stage

    Uno de los principales inconvenientes manifestados por los cirujanos a la hora de llevar a cabo su formación continuada es la elevada carga asistencial que les limita la posibilidad de asistir a cursos y estancias presenciales. Por tanto, los cirujanos necesitan otros métodos formativos distintos a la formación in situ y para ello son de gran utilidad las tecnologías web, como es el caso de la plataforma de e-Learning TELMA, que ofrezcan contenidos didácticos adaptados a cada usuario para que puedan complementar su formación con contenidos multimedia online. Estudios previos de validación de TELMA detectaron diferentes problemas respecto a su diseño y usabilidad, propiciando mayores dificultades de accesibilidad para los cirujanos expertos en relación a los cirujanos noveles. Para corregirlos, en este trabajo se han analizado las deficiencias encontradas y se han propuesto una serie de medidas fundamentadas en un conjunto de directivas de diseño web que se deberían adoptar en futuras versiones de la plataforma TELMA para tratar de solventar estas desigualdades, ofreciendo a todos los usuarios tanto expertos como noveles, las mismas facilidades de accesibilidad

    Filling a blank on the map: 60 years of fisheries in Equatorial Guinea

    Despite a scarcity of pertinent information, it has been possible to reconstruct time series of marine fisheries catches for Equatorial Guinea from 1950 to 2010 using per capita fish consumption and population numbers for small-scale fisheries, catch rates and number of vessels for industrial fisheries and discard rates to estimate the discarded bycatch. Small-scale fisheries, industrial large-scale fisheries, domestic and legal and illegal foreign fisheries and their discards are all included. Total catches were estimated at 2.7 million tonnes over the time period considered, of which 653 000 t were caught domestically compared to 187 000 t reported by FAO. This shows that fisheries have more importance for Equatorial Guinea's food security than the official data suggest. In contrast to what is suggested by official figures, fisheries were shown to be strongly impacted by civil and political unrest; notably, they declined overall because of civil and political conflicts, socio-demographic dynamics, and a growing role of the newly discovered oil resources, which directly and indirectly threaten the food security of the people of Equatorial Guinea

    How allosteric control of Staphylococcus aureus penicillin binding protein 2a enables methicillin resistance and physiological function

    6 pags, 4 figs, 1 tabThe expression of penicillin binding protein 2a (PBP2a) is the basis for the broad clinical resistance to the β-lactam antibiotics by methicillin-resistant Staphylococcus aureus (MRSA). The highmolecular mass penicillin binding proteins of bacteria catalyze in separate domains the transglycosylase and transpeptidase activities required for the biosynthesis of the peptidoglycan polymer that comprises the bacterial cell wall. In bacteria susceptible to β-lactam antibiotics, the transpeptidase activity of their penicillin binding proteins (PBPs) is lost as a result of irreversible acylation of an active site serine by the β-lactam antibiotics. In contrast, the PBP2a of MRSA is resistant to β-lactam acylation and successfully catalyzes the DD-transpeptidation reaction necessary to complete the cell wall. The inability to contain MRSA infection with β-lactam antibiotics is a continuing public health concern. We report herein the identification of an allosteric binding domain - a remarkable 60 Å distant from the DD-transpeptidase active site - discovered by crystallographic analysis of a soluble construct of PBP2a. When this allosteric site is occupied, a multiresidue conformational change culminates in the opening of the active site to permit substrate entry. This same crystallographic analysis also reveals the identity of three allosteric ligands: muramic acid (a saccharide component of the peptidoglycan), the cell wall peptidoglycan, and ceftaroline, a recently approved anti-MRSA β-lactam antibiotic. The ability of an anti-MRSA β-lactam antibiotic to stimulate allosteric opening of the active site, thus predisposing PBP2a to inactivation by a second β-lactam molecule, opens an unprecedented realm for β-lactam antibiotic structure-based design.Work in the United States was supported by National Institutes of Health Grants AI090818 and AI104987, and work in Spain was supported by Grants BFU2011-25326 (from the Spanish Ministry of Economy and Competitiveness) and S2010/BMD-2457 (from the Autonomous Government of Madrid)

    Effective Lagrangian Approach to the Theory of Eta Photoproduction in the N(1535)N^{*}(1535) Region

    We investigate eta photoproduction in the N(1535)N^{*}(1535) resonance region within the effective Lagrangian approach (ELA), wherein leading contributions to the amplitude at the tree level are taken into account. These include the nucleon Born terms and the leading tt-channel vector meson exchanges as the non-resonant pieces. In addition, we consider five resonance contributions in the ss- and uu- channel; besides the dominant N(1535)N^{*}(1535), these are: N(1440),N(1520),N(1650)N^{*}(1440),N^{*}(1520),N^{*}(1650) and N(1710)N^{*}(1710). The amplitudes for the π\pi^\circ and the η\eta photoproduction near threshold have significant differences, even as they share common contributions, such as those of the nucleon Born terms. Among these differences, the contribution to the η\eta photoproduction of the ss-channel excitation of the N(1535)N^{*}(1535) is the most significant. We find the off-shell properties of the spin-3/2 resonances to be important in determining the background contributions. Fitting our effective amplitude to the available data base allows us to extract the quantity χΓηA1/2/ΓT\sqrt{\chi \Gamma_\eta} A_{1/2}/\Gamma_T, characteristic of the photoexcitation of the N(1535)N^{*}(1535) resonance and its decay into the η\eta-nucleon channel, of interest to precise tests of hadron models. At the photon point, we determine it to be (2.2±0.2)×101GeV1(2.2\pm 0.2)\times 10^{-1} GeV^{-1} from the old data base, and (2.2±0.1)×101GeV1(2.2\pm 0.1) \times 10^{-1} GeV^{-1} from a combination of old data base and new Bates data. We obtain the helicity amplitude for N(1535)γpN^{*}(1535)\rightarrow \gamma p to be A1/2=(97±7)×103GeV1/2A_{1/2}=(97\pm 7)\times 10^{-3} GeV^{-1/2} from the old data base, and A1/2=(97±6)×103GeV1/2A_{1/2}=(97\pm 6)\times 10^{-3} GeV^{-1/2} from the combination of the old data base and new Bates data, compared with the results of the analysis of pion photoproduction yielding 74±1174\pm 11, in the same units.Comment: 43 pages, RevTeX, 9 figures available upon request, to appear in Phys.     The matter power spectrum in redshift space using effective field theory

    The use of Eulerian 'standard perturbation theory' to describe mass assembly in the early universe has traditionally been limited to modes with k <= 0.1 h/Mpc at z=0. At larger k the SPT power spectrum deviates from measurements made using N-body simulations. Recently, there has been progress in extending the reach of perturbation theory to larger k using ideas borrowed from effective field theory. We revisit the computation of the redshift-space matter power spectrum within this framework, including for the first time for the full one-loop time dependence. We use a resummation scheme proposed by Vlah et al. to account for damping of the baryonic acoustic oscillations due to large-scale random motions and show that this has a significant effect on the multipole power spectra. We renormalize by comparison to a suite of custom N-body simulations matching the MultiDark MDR1 cosmology. At z=0 and for scales k <~ 0.4 h/Mpc we find that the EFT furnishes a description of the real-space power spectrum up to ~ 2%, for the ell=0 mode up to ~ 5% and for the ell = 2, 4 modes up to ~ 25%. We argue that, in the MDR1 cosmology, positivity of the ell = 0 mode gives a firm upper limit of k ~ 0.74 h/Mpc for the validity of the one-loop EFT prediction in redshift space using only the lowest-order counterterm. We show that replacing the one-loop growth factors by their Einstein-de Sitter counterparts is a good approximation for the ell = 0 mode, but can induce deviations as large as 2% for the ell = 2, 4 modes. An accompanying software bundle, distributed under open source licenses, includes Mathematica notebooks describing the calculation, together with parallel pipelines capable of computing both the necessary one-loop SPT integrals and the effective field theory counterterms

    Search for Higgs bosons decaying to tautau pairs in ppbar collisions at sqrt(s) = 1.96 TeV

    We present a search for the production of neutral Higgs bosons decaying into tautau pairs in ppbar collisions at a center-of-mass energy of 1.96 TeV. The data, corresponding to an integrated luminosity of 5.4 fb-1, were collected by the D0 experiment at the Fermilab Tevatron Collider. We set upper limits at the 95% C.L. on the product of production cross section and branching ratio for a scalar resonance decaying into tautau pairs, and we then interpret these limits as limits on the production of Higgs bosons in the minimal supersymmetric standard model (MSSM) and as constraints in the MSSM parameter space.Comment: 7 pages, 5 figures, submitted to PL

    Measurement of three-jet differential cross sections d sigma-3jet / d M-3jet in p anti-p collisions at sqrt(s)=1.96 TeV

    We present the first measurement of the inclusive three-jet differential cross section as a function of the invariant mass of the three jets with the largest transverse momenta in an event in p anti-p collisions at sqrt(s) = 1.96 TeV. The measurement is made in different rapidity regions and for different jet transverse momentum requirements and is based on a data set corresponding to an integrated luminosity of 0.7 fb^{-1} collected with the D0 detector at the Fermilab Tevatron Collider. The results are used to test the three-jet matrix elements in perturbative QCD calculations at next-to-leading order in the strong coupling constant. The data allow discrimination between parametrizations of the parton distribution functions of the proton.Comment: 10 pages, 4 figures, 2 tables, submitted to Phys. Lett.
