    Molecular and antigenic characterization of Trypanosoma cruzi TolT proteins

    Background: TolT was originally described as a Trypanosoma cruzi molecule that accumulated on the trypomastigote flagellum bearing similarity to bacterial TolA colicins receptors. Preliminary biochemical studies indicated that TolT resolved in SDS-PAGE as ~3–5 different bands with sizes between 34 and 45 kDa, and that this heterogeneity could be ascribed to differences in polypeptide glycosylation. However, the recurrent identification of TolT-deduced peptides, and variations thereof, in trypomastigote proteomic surveys suggested an intrinsic TolT complexity, and prompted us to undertake a thorough reassessment of this antigen. Methods/Principle findings: Genome mining exercises showed that TolT constitutes a larger-than-expected family of genes, with at least 12 polymorphic members in the T. cruzi CL Brener reference strain and homologs in different trypanosomes. According to structural features, TolT deduced proteins could be split into three robust groups, termed TolT-A, TolT-B, and TolT-C, all of them showing marginal sequence similarity to bacterial TolA proteins and canonical signatures of surface localization/membrane association, most of which were herein experimentally validated. Further biochemical and microscopy-based characterizations indicated that this grouping may have a functional correlate, as TolT-A, TolT-B and TolT-C molecules showed differences in their expression profile, sub-cellular distribution, post-translational modification(s) and antigenic structure. We finally used a recently developed fluorescence magnetic beads immunoassay to validate a recombinant protein spanning the central and mature region of a TolT-B deduced molecule for Chagas disease serodiagnosis. Conclusion/Significance: This study unveiled an unexpected genetic and biochemical complexity within the TolT family, which could be exploited for the development of novel T. cruzi biomarkers with diagnostic/therapeutic applications.Fil: Lobo, Mabel Maite.     Noncommutative field gas driven inflation

    We investigate early time inflationary scenarios in an Universe filled with a dilute noncommutative bosonic gas at high temperature. A noncommutative bosonic gas is a gas composed of bosonic scalar field with noncommutative field space on a commutative spacetime. Such noncommutative field theories was recently introduced as a generalization of quantum mechanics on a noncommutative spacetime. As key features of these theories are Lorentz invariance violation and CPT violation. In the present study we use a noncommutative bosonic field theory that besides the noncommutative parameter θ\theta shows up a further parameter σ\sigma. This parameter σ\sigma controls the range of the noncommutativity and acts as a regulator for the theory. Both parameters play a key role in the modified dispersion relations of the noncommutative bosonic field, leading to possible striking consequences for phenomenology. In this work we obtain an equation of state p=ω(σ,θ;β)ρp=\omega(\sigma,\theta;\beta)\rho for the noncommutative bosonic gas relating pressure pp and energy density ρ\rho, in the limit of high temperature. We analyse possible behaviours for this gas parameters σ\sigma, θ\theta and β\beta, so that 1ω<1/3-1\leq\omega<-1/3, which is the region where the Universe enters an accelerated phase.Comment: Reference added.     El papel de la investigación para identificar oportunidades de mejora del regadío en Mauritania

    Ponencia presentada en el I Congreso en Investigación en Agricultura para el desarrollo, celebrado en Madrid el 17 y 18 de octubre de 2011.-- Gómez Macpherson, H. et. al.La producción agrícola de Mauritania se concentra en el valle del río Senegal donde también se concentra gran parte de la población. Sólo un 0,5 % de la superficie mauritana es cultivable, y la tradicional dependencia de la agricultura pluvial ha provocado una alta inseguridad alimentaria; algo que quedó patente con las hambrunas causadas por los ciclos de sequía de los años 70. Los estados ribereños del río Senegal se organizaron entonces para desarrollar zonas regables con el objetivo de la seguridad alimentaria. En Mauritania, el regadío se orientó de forma exclusiva al cultivo del arroz, prácticamente desconocido en la zona. La ricicultura convive con sistemas tradicionales (pluviales y basados en el ciclo anual de inundación-recesión del río), pero el regadío es el sistema que garantiza la producción año a año. Sorprende por tanto que dos tercios de la superficie inicialmente regada se hayan abandonado. Esta paradoja fue el origen de la colaboración entre el Ministerio de Desarrollo Rural de Mauritania, el Instituto de Agricultura Sostenible y la Universidad Politécnica de Cataluña, concretada en proyectos financiados por AECID, y que tiene como objetivo ahondar en las razones del abandono del regadío y en buscar fórmulas para atajarlo. La colaboración se ha realizado sobre tres ejes principales: 1) comprender la interacción del regadío con los sistemas tradicionales de secano y de recesión de inundaciones; 2) diagnosticar el funcionamiento de los pequeños perímetros de riego; y 3) evaluar cultivos alternativos al arroz en términos de productividad de la tierra y del agua. Entre los cultivos regados de interés potencial se identificó el sorgo por ser del gusto de los habitantes del valle y cultivarse tradicionalmente en secano y tras la recesión del agua en suelos inundados.Los autores agradecen a los técnicos de campo y agricultores mauritanos por el trabajo realizado y al MDR y AECID por la financiación.Peer Reviewe

    Immune-inflammatory and hypothalamic-pituitary-adrenal axis biomarkers are altered in patients with non-specific low back pain: A systematic review.

    This systematic review aimed to investigate immune-inflammatory and hypothalamic-pituitary-adrenal (HPA) axis biomarkers in individuals with non-specific low back pain (NSLBP) compared to healthy control. The search was performed in five databases until 4 November 2021. Two reviewers independently conducted screenings, data extraction, risk of bias, and methodological quality assessment of 14 unique studies. All studies reported the source of the fluid analyzed: nine studies used serum, two used plasma, one used serum and plasma, and two studies used salivary cortisol. We found preliminary and limited evidence (only one study for each biomarker) of increased levels in growth differentiation factor 15 (GDF-15), interleukin-23 (IL-23), transforming growth factor-beta (TGF-β), and soluble tumor necrosis factor receptor 1 (sTNF-R1) in NSLBP. Inconsistent and limited evidence was identified for interleukin-10 (IL-10). Although C-reactive protein (CRP), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-α) levels appear to increase in NSLBP, only one study per each biomarker reported statistically significant differences. Interleukin-1 beta (IL-1β), interleukin-17 (IL-17), interferon gamma (IFN-γ), and high-sensitivity CRP (hsCRP) showed no significant differences. Regarding cortisol, one study showed a significant increase and another a significant decrease. More robust evidence between GDF-15, IL-23, TGF-β, and sTNF-R1 with NSLBP is needed. Moreover, contrary to the findings reported in previous studies, when comparing results exclusively with healthy control, insufficient robust evidence for IL-6, TNF-α, and CRP was found in NSLBP. In addition, cortisol response (HPA-related biomarker) showed a dysregulated functioning in NSLBP, with incongruent evidence regarding its directionality. Therefore, our effort is to find adjusted evidence to conclude which immune-inflammatory and HPA axis biomarkers are altered in NSLBP and how much their levels are affected

    Social inequities in the impact of COVID-19 lockdown measures on the mental health of a large sample of the Colombian population (PSY-COVID study)

    The identification of general population groups particularly vulnerable to the impact of COVID-19 lockdown measures on mental health and the development of healthcare policies are priority challenges in the current and future pandemics. This study aimed to identify the personal and social determinants of the impact of COVID-19 lockdown measures on mental health in a large sample of the Colombian population. In this cross-sectional study, an anonymous online survey was answered by 18,061 participants from the general population residing in Colombia during the first wave of the COVID-19 outbreak (from 20 May to 20 June 2020). The risk of depression, anxiety, and somatization disorders were measured using the Patient Health Questionnaire (PHQ-2), Generalized Anxiety Disorder Scale (GAD-2), and Somatic Symptom Questionnaire (SSQ-5), respectively. Overall, 35% of participants showed risk of depression, 29% of anxiety, and 31% of somatization. According to the analysis of social determinants of health, the most affected groups were people with low incomes, students, and young adults (18-29 years). Specifically, low-income young females were the most at-risk population group. These findings show how the lockdown measures affected the general population's mental health in Colombia and highlight some social risk factors in health

    Robotic repair of vesicovaginal fistulae with the transperitoneal-transvaginal approach: A case series

    ABSTRACT ARTICLE INFO ______________________________________________________________ ______________________ Objective: To describe a novel technique of repairing the VVF using the transperitoneal-transvaginal approach. Materials and Methods: From June 2011 to October 2013, four patients with symptoms of urine leakage in the vagina underwent robotic repair of VVF with the transperitoneal-transvaginal approach. Cystoscopy revealed the fistula opening on the bladder. A ureteral stent was placed through the fistulous tract. After trocar placement, the omental flap was prepared and mobilized robotically. The vagina was identified and incised. The fistulous tract was excised. Cystorrhaphy was performed in two layers in an interrupted fashion. The vaginal opening was closed with running stitches. The omentum was interposed and anchored between the bladder and vagina. Finally, the ureteral catheters were removed in case they have been placed, and an 18 Fr urethral catheter was removed on the 14th postoperative day. Results: The mean age was 46 years (range: 41 to 52 years). The mean fistula diameter was 1.5 cm (range 0.3 to 2 cm). The mean operative time was 117.5 min (range: 100 to 150 min). The estimated blood loss was 100 mL (range: 50 to 150 mL). The mean hospital stay was 1.75 days (range: 1 to 3 days). The mean Foley catheter duration was 15.75 days (range: 10 to 25 days). There was no evidence of recurrence in any of the cases. Conclusions: The robot-assisted laparoscopic transperitoneal transvaginal approach for VVF is a feasible procedure when the fistula tract is identified by first intentionally opening the vagina, thereby minimizing the bladder incision and with low morbidity

    Cuidados en el puerperio durante la Crisis sanitaria por CoVid-19

    Introducción: la infección por el nuevo coronavirus COVID-19, es actualmente una emergencia de salud pública internacional y ha sido declarada pandemia por la OMS. Aún se desconocen ciertos datos sobre su etiopatogenia, surgiendo la necesidad de analizar la influencia de la enfermedad por coronavirus durante la etapa puerperal y sus conse-cuencias. Objetivo: sintetizar el conocimiento y las últimas recomendaciones disponibles acerca de los cuidados específicos durante el periodo del puerperio, en relación con la crisis sanitaria por COVID-19. Metodología: se llevó a cabo una búsqueda sistemática en la literatura científica para localizar los documentos de evidencia científica disponibles en las principales bases de datos referenciales: Pubmed, Biblioteca Cochrane y Science Direct. Resultados: la lactancia materna aporta numerosos beneficios al recién nacido, entre ellos, la protección frente a infecciones respiratorias como el coronavirus. Si el estado materno y neonatal lo permite, se recomienda promover la lactancia materna durante el periodo de riesgo infeccioso. La SEGO y la SETH recomiendan la administra-ción de heparina de bajo peso molecular a dosis profilácticas para evitar la enfermedad tromboembólica. La evidencia actual indica que el riesgo de transmisión a través de la lactancia materna al recién nacido es muy poco probable. Discusión/conclusiones: la limitación principal fue la escasez de literatura científica sobre el COVID-19 en relación con el periodo puerperal. Es fundamental que cualquier puérpera con una infección por COVID-19 y su recién nacido sean valorados de manera individualizada, precozmente y evaluada por un equipo multidisciplinar para prevenir resultados adversosIntroduction: the new coronavirus COVID-19 infection is currently an international pu-blic health emergency and it has been declared a pandemic by the WHO. Certain data about its etiopathogenesis is still unknown, arising the need to analyse the influence of coronavirus disease during the puerperal stage and its consequences. Objective:to synthesize the knowledge and the latest available recommendations about specific care during puerperium period, related to the health crisis due to COVID-19. Methodo-logy: a systematic search was carried out in the scientific literature in order to locate the scientific evidence documents available in the main reference databases: PubMed, Cochrane Library and Science Direct. Results: breastfeeding brings many benefits to newborns, including protection against respiratory infections, such as coronavirus. If maternal and neonatal status allows it, it is recommended promoting breastfeeding during the infectious risk period. Both SEGO and SETH recommend the administration of low molecular weight heparin at prophylactic doses in order to avoid thromboembolic disease. Discussion/conclusions: the main limitation was the paucity of scientific lite-rature about COVID-19 in relation to puerperal period. It is essential that any puerperal woman with COVID-19 infection and her newborn are assessed individually, early and evaluated by a multidisciplinary team to prevent adverse outcome
