Implementation of a Practical Teaching Course on Protein Engineering
Simple Summary Proteins are the workhorses of the cell. With different combinations of the 20 common amino acids and some modifications of these amino acids, proteins have evolved with a staggering array of new functions and capabilities due to Protein Engineering techniques. The practical course presented was offered to undergraduate bioengineering and chemical students at the Faculty of Engineering of the University of Porto (Portugal) and consists of sequential laboratory sessions to learn the basic skills related to the expression and purification of recombinant proteins in bacterial hosts. These experiments were successfully applied by students as all working groups were able to isolate a model recombinant protein (the enhanced green fluorescent protein) from a cell lysate containing a mixture of proteins and other biomolecules produced by an Escherichia coli strain and evaluate the performance of the extraction and purification procedures they learned. Protein Engineering is a highly evolved field of engineering aimed at developing proteins for specific industrial, medical, and research applications. Here, we present a practical teaching course to demonstrate fundamental techniques used to express, purify and analyze a recombinant protein produced in Escherichia coli-the enhanced green fluorescent protein (eGFP). The methodologies used for eGFP production were introduced sequentially over six laboratory sessions and included (i) bacterial growth, (ii) sonication (for cell lysis), (iii) affinity chromatography and dialysis (for eGFP purification), (iv) bicinchoninic acid (BCA) and fluorometry assays for total protein and eGFP quantification, respectively, and (v) sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) for qualitative analysis. All groups were able to isolate the eGFP from the cell lysate with purity levels up to 72%. Additionally, a mass balance analysis performed by the students showed that eGFP yields up to 46% were achieved at the end of the purification process following the adopted procedures. A sensitivity analysis was performed to pinpoint the most critical steps of the downstream processing
Transitional Care Management from Emergency Services to Communities: An Action Research Study
In recent years, nurses have developed projects in the area of hospital to community
transition. The objective of the present study was to analyze the transitional care offered to elderly
people after they used emergency services and were discharged to return to the community. The
action research method was chosen. The participants were nurses, elderly people 70 years old or
older, and their caregivers. The study was carried out from October 2018 to August 2019. The
data were collected by means of semi-structured interviews with the nurses, analysis of medical
records, participatory observation, phone calls to the elderly people and caregivers, and team
meetings. The qualitative data were submitted to Bardin’s content analysis. Statistical treatment
was carried out by applying SPSS version 23.0. The institution’s research ethics committee approved
the research. Only 31.4% of the sample experienced care continuity after discharge, and the rate
of readmission to emergency services during the first 30 days after discharge was 33.4%. The
referral letters lacked data on information provided to patients or caregivers, and nurses mentioned
difficulties in communication between care levels, as well as obstacles to teamwork; they also
mentioned that the lack of health policies and clinical rules to formalize transitional care between the
hospital and the community perpetuated non-coordination of care between the two contexts. The
low level of literacy of patients and their relatives are mentioned as a cause for not understanding
the information regarding seeking primary health care services and handing the discharge letter. It
was concluded that there is an urgent need to mobilize health teams toward action in the patients’
process of returning home, and this factor must be taken into account in care planning.info:eu-repo/semantics/publishedVersio
FERNANDO PESSOA E A “ALGIBEIRA VAZIA”. ESTUDO IMAGÉTICO DO POEMA ANIVERSÁRIO
Gneuropathy: Validation process at clinical environment
Spinal cord injuries are one of the most traumatic situations with a major impact on a person's quality of life. This type of injury have a extremely impact in the performance of daily life activities not only due to motor alterations but also due to the appearance of neuropathic pain Throughout the rehabilitation process the evaluation and intervention methodologies are not very systematic and are not personalized. Thus, to bridge this gap, the VR4NeuroPain was developed a technology that associates virtual reality with a glove "GNeuroPathy". The glove "GNeuroPathy" allows the collection of physiological parameters, namely to identify the electrodermic activity (EDA) while the patient carries out activities in an immersive environment. The main objective of this article is to present the validation process of the "GNeuroPathy" in clinical context. "GNeuroPathy" was applied to a group of 17 individuals with incomplete spinal cord injury. The results showed that "GNeuroPathy" is easy to apply and is suitable for comfort and texture. Data were also collected from EDA and it was found that there is a significant difference in signal amplitude in patients with low and high functionality.preprintpublishe

“ESTRANGEIRO AQUI COMO EM TODA A PARTE” UMA ABORDAGEM DOS POEMAS “LISBON REVISITED”, DE FERNANDO PESSOA.
Nostalgia of childhood marks the poetry of Fernando Pessoa, as relating to mythical and idealization. The objective of this research is to analyze the poems Lisbon Revisited (1923) and Lisbon Revisited (1926) by Alvaro de Campos, in his imagistic components, his revealed confrontations of the emptiness of the self and the connection with the poetic process that links Modernism to memory. Examining the explicit confrontation with the city of Lisbon after generalizing the figure of the foreigner to all the needs of involvement and social commitment (poem, 1923). Interpreting the poems, since the strangeness of the title and the need to inquire about the process of heteronomy, nihilism and the passivity, despite the connection to memory, (poem, 1926).
Learning from Online Voices: A Mixed Methods Approach to Explore Patient Online Reviews of Hospital Care in Portugal
Online patient reviews can offer a rich information source to users of healthcare services, as well as for hospital management and quality monitoring. Whereas in recent years the volume of online patient reviews has been consistently growing, organizations still lack standardized approaches and tools to allow for the systematic monitoring of users’ online comments. Therefore, managers are lagging in the ability to make use of such data from patients’ voices for improving the quality of the services provided. If organizations fail to develop the right capabilities to consider users’ online reviews and feedback, they risk not only to miss important quality failure alerts, as wells as to frustrate their customers’ expectations for service and attention. In this chapter, we present a qualitative analysis of patients’ reviews for healthcare services in Portugal, building on a sample of data extracted from Google for the year of 2019. The chapter reports the major quality management themes addressed by hospital users in their online expressions and offers some guidelines to support a structured analysis and visualization of results from online users’ word of mouth data
