    Electromyographic assessment of muscle fatigue during isometric vibration training at varying frequencies

    Resistance exercise is essential to improve or maintain muscle performance. Vibration training has been suggested as an alternative option for muscle conditioning, aiming especially at improving muscle strength and power. Several studies link the effects of vibration training to enhanced neuromuscular stimulation, measured by electromyography (EMG) and typically ascribed to involuntary reflex mechanisms. However, the underlying mechanisms are still unclear, limiting the use of vibration training. This paper proposes additional methods to analyze the mechanisms involved in vibration training. A dedicated measurement setup was realized to relate vibration parameters to muscle fatigue in the biceps brachii. Fatigue is estimated by EMG mean frequency and conduction velocity assessments as well as by maximum voluntary contraction (MVC) force measurements. A modified maximum likelihood algorithm is proposed for the conduction velocity estimation based on high-density EMG recording. Five volunteers performed four isometric contractions of 50 s at 80% MVC with no vibration (control) and with superimposed vibration at 20, 30, and 40 Hz. Fatigue was estimated from the decay of force, EMG mean frequency, and EMG conduction velocity. 30-Hz vibrations represented the most fatiguing stimulus. Our preliminary results also show a better correlation between force and conduction velocity decay than between force and mean frequency decay, indicating the former as a better EMG indicator of fatigue. The proposed methods provide important advancements for the analysis of vibration exercise and guidance towards the definition of optimal training protocols

    The effect of plant domestication on host control of the microbiota

    Macroorganisms are colonized by microbial communities that exert important biological and ecological functions, the composition of which is subject to host control and has therefore been described as “an ecosystem on a leash”. However, domesticated organisms such as crop plants are subject to both artificial selection and natural selection exerted by the agricultural ecosystem. Here, we propose a framework for understanding how host control of the microbiota is influenced by domestication, in which a double leash acts from domesticator to host and host to microbes.     Clinical relevance of biomarkers in cholangiocarcinoma: critical revision and future directions

    Cholangiocarcinoma (CCA) is a malignant tumour arising from the biliary system. In Europe, this tumour frequently presents as a sporadic cancer in patients without defined risk factors and is usually diagnosed at advanced stages with a consequent poor prognosis. Therefore, the identification of biomarkers represents an utmost need for patients with CCA. Numerous studies proposed a wide spectrum of biomarkers at tissue and molecular levels. With the present paper, a multidisciplinary group of experts within the European Network for the Study of Cholangiocarcinoma discusses the clinical role of tissue biomarkers and provides a selection based on their current relevance and potential applications in the framework of CCA. Recent advances are proposed by dividing biomarkers based on their potential role in diagnosis, prognosis and therapy response. Limitations of current biomarkers are also identified, together with specific promising areas (ie, artificial intelligence, patient-derived organoids, targeted therapy) where research should be focused to develop future biomarkers

    Safety and Efficacy of Subcutaneous Rituximab in Previously Untreated Patients with CD20+ Diffuse Large B-Cell Lymphoma or Follicular Lymphoma: Results from an Italian Phase IIIb Study

    Subcutaneous (SC) rituximab may be beneficial in terms of convenience and tolerability, with potentially fewer and less severe administration-related reactions (ARRs) compared to the intravenous (IV) form. This report presents the results of a phase IIIb study conducted in Italy. The study included adult patients with CD20+ DLBCL or FL having received at least one full dose of IV RTX 375 mg/m2 during induction or maintenance. Patients on induction received ≥4 cycles of RTX SC 1400 mg plus standard chemotherapy and FL patients on maintenance received ≥6 cycles of RTX SC. Overall, 159 patients (73 DLBCL, 86 FL) were enrolled: 103 (54 DLBCL, 49 FL) completed induction and 42 patients with FL completed 12 maintenance cycles. ARRs were reported in 10 patients (6.3%), 3 (4.2%) with DLBCL and 7 (8.1%) with FL, all of mild severity, and resolved without dose delay/discontinuation. Treatment-emergent adverse events (TEAEs) and serious adverse events occurred in 41 (25.9%) and 14 patients (8.9%), respectively. Two patients with DLBCL had fatal events: Klebsiella infection (related to rituximab) and septic shock (related to chemotherapy). Neutropenia (14 patients, 8.9%) was the most common treatment-related TEAE. Two patients with DLBCL (2.8%) and 6 with FL (7.0%) discontinued rituximab due to TEAEs. 65.2% and 69.7% of patients with DLBCL and 67.9% and 73.6% of patients with FL had complete response (CR) and CR unconfirmed, respectively. The median time to events (EFS, PFS, and OS) was not estimable due to the low rate of events. At a median follow-up of 29.5 and 47.8 months in patients with DLBCL and FL, respectively, EFS, PFS, and OS were 70.8%, 70.8%, and 80.6% in patients with DLBCL and 77.9%, 77.9%, and 95.3% in patients with FL, respectively. The switch from IV to SC rituximab in patients with DLBCL and FL was associated with low risk of ARRs and satisfactory response in both groups. This trial was registered with NCT01987505

    Consistent effects of independent domestication events on the plant microbiota

    The effect of plant domestication on plant-microbe interactions remains difficult to prove. In this study, we provide evidence of a domestication effect on the composition and abundance of the plant microbiota. We focused on the genus Phaseolus, which underwent four independent domestication events within two species (P. vulgaris and P. lunatus), providing multiple replicates of a process spanning thousands of years. We targeted Phaseolus seeds to identify a link between domesticated traits and bacterial community composition as Phaseolus seeds have been subject to large and consistent phenotypic changes during these independent domestication events. The seed bacterial communities of representative plant accessions from subpopulations descended from each domestication event were analyzed under controlled and field conditions. The results showed that independent domestication events led to similar seed bacterial community signatures in independently domesticated plant populations, which could be partially explained by selection for common domesticated plant phenotypes. Our results therefore provide evidence of a consistent effect of plant domestication on seed microbial community composition and abundance and offer avenues for applying knowledge of the impact of plant domestication on the plant microbiota to improve microbial applications in agriculture

    Long-range phase synchronization of high-gamma activity in human cortex

    AbstractInter-areal synchronization of neuronal oscillations below 100 Hz is ubiquitous in cortical circuitry and thought to regulate neuronal communication. In contrast, faster activities are generally considered to be exclusively local-circuit phenomena. We show with human intracerebral recordings that 100–300 Hz high-gamma activity (HGA) may be synchronized between widely distributed regions. HGA synchronization was not attributable to artefacts or to epileptic pathophysiology. Instead, HGA synchronization exhibited a reliable cortical connectivity and community structures, and a laminar profile opposite to that of lower frequencies. Importantly, HGA synchronization among functional brain systems during non-REM sleep was distinct from that in resting state. Moreover, HGA synchronization was transiently enhanced for correctly inhibited responses in a Go/NoGo task. These findings show that HGA synchronization constitutes a new, functionally significant form of neuronal spike-timing relationships in brain activity. We suggest that HGA synchronization reflects the temporal microstructure of spiking-based neuronal communication per se in cortical circuits

    Search for the rare decays B0J/ψγB^{0}\to J/\psi \gamma and Bs0J/ψγB^{0}_{s} \to J/\psi \gamma

    A search for the rare decay of a B0B^{0} or Bs0B^{0}_{s} meson into the final state J/ψγJ/\psi\gamma is performed, using data collected by the LHCb experiment in pppp collisions at s=7\sqrt{s}=7 and 88 TeV, corresponding to an integrated luminosity of 3 fb1^{-1}. The observed number of signal candidates is consistent with a background-only hypothesis. Branching fraction values larger than 1.7×1061.7\times 10^{-6} for the B0J/ψγB^{0}\to J/\psi\gamma decay mode are excluded at 90% confidence level. For the Bs0J/ψγB^{0}_{s}\to J/\psi\gamma decay mode, branching fraction values larger than 7.4×1067.4\times 10^{-6} are excluded at 90% confidence level, this is the first branching fraction limit for this decay.Comment: All figures and tables, along with any supplementary material and additional information, are available at https://lhcbproject.web.cern.ch/lhcbproject/Publications/LHCbProjectPublic/LHCb-PAPER-2015-044.htm

    IgE sensitisation in relation to flow-independent nitric oxide exchange parameters

    BACKGROUND: A positive association between IgE sensitisation and exhaled NO levels has been found in several studies, but there are no reports on the compartment of the lung that is responsible for the increase in exhaled NO levels seen in IgE-sensitised subjects. METHODS: The present study comprised 288 adult subjects from the European Community Respiratory Health Survey II who were investigated in terms of lung function, IgE sensitisation (sum of specific IgE), smoking history and presence of rhinitis and asthma. Mean airway tissue concentration of NO (Caw(NO)), airway transfer factor for NO (Daw(NO)), mean alveolar concentration of NO (Calv(NO)) and fractional exhaled concentration of NO at a flow rate of 50 mL s(-1 )(FE(NO 0.05)) were determined using the extended NO analysis. RESULTS: IgE-sensitised subjects had higher levels (geometric mean) of FE(NO 0.05 )(24.9 vs. 17.3 ppb) (p < 0.001), Daw(NO )(10.5 vs. 8 mL s(-1)) (p = 0.02) and Caw(NO )(124 vs. 107 ppb) (p < 0.001) and positive correlations were found between the sum of specific IgE and FE(NO 0.05), Caw(NO )and Daw(NO )levels (p < 0.001 for all correlations). Sensitisation to cat allergen was the major determinant of exhaled NO when adjusting for type of sensitisation. Rhinitis and asthma were not associated with the increase in exhaled NO variables after adjusting for the degree of IgE sensitisation. CONCLUSION: The presence of IgE sensitisation and the degree of allergic sensitisation were related to the increase in airway NO transfer factor and the increase in NO concentration in the airway wall. Sensitisation to cat allergen was related to the highest increases in exhaled NO parameters. Our data suggest that exhaled NO is more a specific marker of allergic inflammation than a marker of asthma or rhinitis

    LHCb Upgraded RICH 1 Engineering Design Review Report

    During the Long Shutdown 2 of the LHC, the LHCb collaboration will replace the upstream Ring Imaging Cherenkov detector (RICH 1). The magnetic shield of the current RICH 1 will be modified, new spherical and plane mirrors will be installed and a new gas enclosure will be manufactured. New photon detectors (multianode photomultiplier tubes) will be used and these, together with their readout electronics, require a new mechanical support system. This document describes the new optical arrangement of RICH 1, its engineering design, installation and alignment. A summary of the project schedule and institute responsibilities is provided
