    Vertical distribution of PAHs in marine sediments (particulate fraction and interstitial water) from eight Iberian Mediterranean areas

    Polycyclic aromatic hydrocarbons (PAHs) have been determined in sediment cores from eight Iberian Mediterranean coastal areas (Barcelona, Tarragona, Ebro Delta, Valencia, Castellón, Cartagena, Almería y Málaga), considering both interstitial water and solid fraction. Three sediment cores were taken in three different sampling sites per area by using a box corer. Cores were cut into 1-cm-thick sections between 0 and 18-cm depth, interstitial water was obtained by centrifugation and samples from every site were pooled. Fourteen polycyclic aromatic hydrocarbons (fluorene, phenanthrene, anthracene, fluoranthene, pyrene, benz[a]anthracene, crysene, benzo[e]pyrene, benzo[b]fluoranthene, benzo[k]fluoranthene, benzo[a]pyrene, benzo[g,h,i]perylene, dibenzo[a,h]anthracene and indeno[1,2,3-c,d]pyrene) were determined in interstitial water by stir bar sorptive extraction coupled to GC/MS and solid fraction by Soxhlet extraction and HPLC with fluorescence detection. As result of PAHs hydrofobicity, their concentrations were several magnitude order higher in sediment (solid phase) than interstitial water. Total PAH concentration in sediment varied from 1 to 1321 g•kg-1 d.w and from 4.9 to 274 ng•L-1 in interstitial water. Vertical PAHs distributions in sediments were homogeneous in some areas, however, sharp concentration decreases were observed at deeper layers from some sampling sites.     Levels and spatial distribution of organochlorinated compounds and polycyclic aromatic hydrocarbons in fishes from a Mediterranean coastal lagoon

    The seasonal input and distribution of organic contaminants in the Mar Menor lagoon have been recently characterized in seawater and sediments, showing the significant impact of flash flood events. Mar Menor is a hypersaline (42-47 psu) coastal lagoon located in the Cartagena Field area at the South East of Spain subjected mainly to intensive agriculture, seasonal tourism and recreational activities. The bioaccumulation of polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs) and organochlorinated pesticides (OCPs) has been also determined in bivalves, but no information was available about their accumulation in fishes. In this study the concentrations of PAHs, PCBs and OCPs in golden grey mullet (Liza aurata) and red mullet (Mullus barbatus) muscle were determined. Their spatial distribution throughout the Mar Menor lagoon was characterized considering 9 sampling areas and two different sampling periods, spring and autumn in 2010 for golden grey mullet and only one (autumn) for red mullet. PAHs and organochlorinated pollutants were extracted using specific Soxhlet procedures. After cleanup and purification steps the final extracts were analyzed by HPLC for PAHs, and by GC-MS for PCBs and OCPs. The higher concentrations of all pollutants were found close to main urban and port areas of this lagoon and in the influence area of El Albujón watercourse. PAHs concentrations (sum of 14 congeners) in both species varied between 1 and 20.7 g/kg w.w. being pyrene and phenanthrene the most abundant ones. p,p’DDE showed higher concentrations than PAHs in golden grey mullet (concentration range: 0.2-32.7 g/kg w.w.), specially close to the main navigation channel between this lagoon and the Mediterranean Sea.This work has been supported by the Spanish Inter-Ministerial Science and Technology Commission through ‘DECOMAR’ project (CICYT, CTM2008-01832) and by Seneca Foundation (Region of Murcia, Spain) through ‘BIOMARO’ project (15398/PI/10).     Subtly modulating glycogen synthase kinase 3 ß: Allosteric inhibitor development and their potential for the treatment of chronic diseases

    Glycogen synthase kinase 3 ß (GSK-3ß) is a central target in several unmet diseases. To increase the specificity of GSK-3ß inhibitors in chronic treatments, we developed small molecules allowing subtle modulation of GSK-3ß activity. Design synthesis, structure¿activity relationships, and binding mode of quinoline-3-carbohydrazide derivatives as allosteric modulators of GSK-3ß are presented here. Furthermore, we show how allosteric binders may overcome the ß-catenin side effects associated with strong GSK-3ß inhibition. The therapeutic potential of some of these modulators has been tested in human samples from patients with congenital myotonic dystrophy type 1 (CDM1) and spinal muscular atrophy (SMA) patients. We found that compound 53 improves delayed myogenesis in CDM1 myoblasts, while compounds 1 and 53 have neuroprotective properties in SMA-derived cells. These findings suggest that the allosteric modulators of GSK-3ß may be used for future development of drugs for DM1, SMA, and other chronic diseases where GSK-3ß inhibition exhibits therapeutic effects.This work was financially supported by MINECO (grant no.     Long-Term Dynamic in Nutrients, Chlorophyll a, and Water Quality Parameters in a Coastal Lagoon During a Process of Eutrophication for Decades, a Sudden Break and a Relatively Rapid Recovery

    Coastal lagoons are considered among the marine habitats with the highest biological productivity, and support a great variety of human activities and pressures that make them especially vulnerable to trophic imbalances. While dystrophic crises are common in many lagoons, others like the Mar Menor show homeostatic mechanisms, high resilience, and clear waters. This paper analyses the water column descriptors dynamic during the last 22 years in this coastal lagoon, in the context of a eutrophication process produced by an increase in nutrient inputs, mainly derived from agriculture. Despite water column nitrate concentration increased by one order of magnitude, the lagoon maintained homeostatic regulation for two decades, keeping the water transparency and relatively low levels of nutrients and chlorophyll a (Prebreak phase), followed by a sudden change of state in 2016 with an abrupt increase in average nutrients and chlorophyll a concentration and loss of water transparency (Break phase), and a relatively rapid recovery after the reduction of nutrient discharges (Recovery phase). The activation of the regulation mechanisms seems to manifest through an ammonium production in the water column, as a consequence of the activity in the trophic web. The low correlation between chlorophyll a and nutrients concentration, mainly at small spatio-temporal scales, is in disagreement with eutrophication traditional models, and suggests a rapid response of primary producers to nutrient inputs and a zooplankton control in the short-term, which in turn is controlled by the rest of the trophic web components. Homeostatic properties that in the Mar Menor lagoon have provided resistance to eutrophication are based on several mechanisms: channeling its production toward the benthic system (maintaining high biomasses of primary producers, filter feeders, and detritivores), a top-down control of the pelagic trophic web exerted by ichthyoplankton and jellyfish, and exporting surplus production outside the system. Resilience of the system would be based on the high turnover in the species composition related to the restricted connectivity to the sea, the spatio-temporal variability of the environmental conditions, and the multiplicity of spatial–temporal scales involved in lagoon processes. TRIX index was sensitive to the trophic and water quality changes. However, in our study, its current score does not allow to anticipate or alert the eutrophication risk and the trophic breakpoint of the system

    Synthesis, Structural Analysis, and Biological Evaluation of Thioxoquinazoline Derivatives as Phosphodiesterase 7 Inhibitors

    PDE7 inhibitors regulate pro-inflammatory and immune T-cell functions, and are a potentially novel class of drugs especially useful in the treatment of a wide variety of immune and inflammatory disorders. Starting from our lead family of thioxoquinazolines, we designed, synthesized, and characterized a novel series of thioxoquinazoline derivatives. Many of these compounds showed inhibitory potencies at sub-micromolar levels against the catalytic domain of PDE7A1 and at the micromolar level against PDE4D2. Cell-based studies showed that these compounds not only increased intracellular cAMP levels, but also had interesting anti-inflammatory properties within a therapeutic window. The in silico data predict that these compounds are capable of the crossing the blood–brain barrier. The X-ray crystal structure of the PDE7A1 catalytic domain in complex with compound 15 at a resolution of 2.4 Å demonstrated that hydrophobic interactions at the active site pocket are a key feature. This structure, together with molecular modeling, provides insight into the selectivity of the PDE inhibitors and a template for the discovery of new PDE7 or PDE7/PDE4 dual inhibitors

    Se integra información hidrográfica, geomorfológica, sedimentológica y biológica, para la caracterización de los ecosistemas marinos en el punto de vertido y área adyacente previa al inicio de las obras de dragado del Puerto de Maó.RESUMEN: En este documento se presentan los trabajos científicos realizados por el Instituto Español de Oceanografía, dentro del Plan de Vigilancia Ambiental del dragado del Puerto de Maó (Menorca, Islas Baleares), para la caracterización de los ecosistema marino en el punto de vertido y área adyacente, previa al inicio de las obras. Se incluyen los resultados y las conclusiones de los estudios realizados por diversos grupos de investigación, principalmente en Enero-Marzo 2014, en relación al fondo marino, la hidrodinámica, las praderas de Posidonia oceanica y el molusco bivalvo Pinna nobilis, el macro-bentos de los fondos circalitorales blandos y los contaminantes en agua, sedimentos y biota, así como en especies de interés comercial para el consumo humano.     Effectiveness of an intervention for improving drug prescription in primary care patients with multimorbidity and polypharmacy:Study protocol of a cluster randomized clinical trial (Multi-PAP project)

    This study was funded by the Fondo de Investigaciones Sanitarias ISCIII (Grant Numbers PI15/00276, PI15/00572, PI15/00996), REDISSEC (Project Numbers RD12/0001/0012, RD16/0001/0005), and the European Regional Development Fund ("A way to build Europe").Background: Multimorbidity is associated with negative effects both on people's health and on healthcare systems. A key problem linked to multimorbidity is polypharmacy, which in turn is associated with increased risk of partly preventable adverse effects, including mortality. The Ariadne principles describe a model of care based on a thorough assessment of diseases, treatments (and potential interactions), clinical status, context and preferences of patients with multimorbidity, with the aim of prioritizing and sharing realistic treatment goals that guide an individualized management. The aim of this study is to evaluate the effectiveness of a complex intervention that implements the Ariadne principles in a population of young-old patients with multimorbidity and polypharmacy. The intervention seeks to improve the appropriateness of prescribing in primary care (PC), as measured by the medication appropriateness index (MAI) score at 6 and 12months, as compared with usual care. Methods/Design: Design:pragmatic cluster randomized clinical trial. Unit of randomization: family physician (FP). Unit of analysis: patient. Scope: PC health centres in three autonomous communities: Aragon, Madrid, and Andalusia (Spain). Population: patients aged 65-74years with multimorbidity (≥3 chronic diseases) and polypharmacy (≥5 drugs prescribed in ≥3months). Sample size: n=400 (200 per study arm). Intervention: complex intervention based on the implementation of the Ariadne principles with two components: (1) FP training and (2) FP-patient interview. Outcomes: MAI score, health services use, quality of life (Euroqol 5D-5L), pharmacotherapy and adherence to treatment (Morisky-Green, Haynes-Sackett), and clinical and socio-demographic variables. Statistical analysis: primary outcome is the difference in MAI score between T0 and T1 and corresponding 95% confidence interval. Adjustment for confounding factors will be performed by multilevel analysis. All analyses will be carried out in accordance with the intention-to-treat principle. Discussion: It is essential to provide evidence concerning interventions on PC patients with polypharmacy and multimorbidity, conducted in the context of routine clinical practice, and involving young-old patients with significant potential for preventing negative health outcomes.
