Whole-genome sequencing of 1,171 elderly admixed individuals from Brazil

As whole-genome sequencing (WGS) becomes the gold standard tool for studying population genomics and medical applications, data on diverse non-European and admixed individuals are still scarce. Here, we present a high-coverage WGS dataset of 1,171 highly admixed elderly Brazilians from a census-based cohort, providing over 76 million variants, of which ~2 million are absent from large public databases. WGS enables identification of ~2,000 previously undescribed mobile element insertions without previous description, nearly 5 Mb of genomic segments absent from the human genome reference, and over 140 alleles from HLA genes absent from public resources. We reclassify and curate pathogenicity assertions for nearly four hundred variants in genes associated with dominantly-inherited Mendelian disorders and calculate the incidence for selected recessive disorders, demonstrating the clinical usefulness of the present study. Finally, we observe that whole-genome and HLA imputation could be significantly improved compared to available datasets since rare variation represents the largest proportion of input from WGS. These results demonstrate that even smaller sample sizes of underrepresented populations bring relevant data for genomic studies, especially when exploring analyses allowed only by WGS

Terapias voltadas para o tratamento do transtorno dissociativo de identidade

O transtorno dissociativo de identidade compreende uma condição psicológica complexa provavelmente causada por inúmeros fatores, envolvendo trauma grave na primeira infância, como abuso sexual, físico ou emocional repetitivo e extremo e repetitivo. Este estudo teve como objetivo identificar as terapias voltadas para o tratamento do transtorno dissociativo de identidade. Para isso, foi realizada uma revisão integrativa de literatura, selecionando fontes a partir das bases de dados Medline e Lilacs. A partir da análise qualitativa de dados, concluiu-se que há vários tipos de terapias para o tratamento de pessoas transtorno dissociativo de identidade, devendo essas serem aplicadas conforme cada realidade. Nos estudos, foram identificados os modelos de tratamento psicanalítico relacional, fásico, psicoativo e psicotraumatológico. Em todos esses, foram registrados resultados satisfatórios, tais como a diminuição na dissociação e o aumento do funcionamento adaptativo do paciente, revelando a possibilidade de desconstruir crenças solidamente cultivadas e trazendo esperança aos pacientes no sentido de amenizar ou superar esse transtorno e garantir uma boa interação social

Taking the pulse of Earth's tropical forests using networks of highly distributed plots

Tropical forests are the most diverse and productive ecosystems on Earth. While better understanding of these forests is critical for our collective future, until quite recently efforts to measure and monitor them have been largely disconnected. Networking is essential to discover the answers to questions that transcend borders and the horizons of funding agencies. Here we show how a global community is responding to the challenges of tropical ecosystem research with diverse teams measuring forests tree-by-tree in thousands of long-term plots. We review the major scientific discoveries of this work and show how this process is changing tropical forest science. Our core approach involves linking long-term grassroots initiatives with standardized protocols and data management to generate robust scaled-up results. By connecting tropical researchers and elevating their status, our Social Research Network model recognises the key role of the data originator in scientific discovery. Conceived in 1999 with RAINFOR (South America), our permanent plot networks have been adapted to Africa (AfriTRON) and Southeast Asia (T-FORCES) and widely emulated worldwide. Now these multiple initiatives are integrated via ForestPlots.net cyber-infrastructure, linking colleagues from 54 countries across 24 plot networks. Collectively these are transforming understanding of tropical forests and their biospheric role. Together we have discovered how, where and why forest carbon and biodiversity are responding to climate change, and how they feedback on it. This long-term pan-tropical collaboration has revealed a large long-term carbon sink and its trends, as well as making clear which drivers are most important, which forest processes are affected, where they are changing, what the lags are, and the likely future responses of tropical forests as the climate continues to change. By leveraging a remarkably old technology, plot networks are sparking a very modern revolution in tropical forest science. Action audio-visual for the promotion of between mother attachment seropositivity for HIV and his son

The study aimed at evaluating the effects of the educational video use to promote the attachment between HIV positive mother and her son after six months of contact between them.This is an exploratory-descriptive study with a qualitative approach. Data was collected through semi-structured interviews. The sampling was intentional. The plan of analysis consisted of the following steps: the constitution of the corpus, definition of unit analysis, data categorization and analysis through categories. This study showed that, despite the difficulties and obstacles imposed by HIV / AIDS, by the treatment and the doubts present in daily life, HIV mothers develop strategies that enable them to live and better deal with the disease, without forgetting their role as a mother

El objetivo de estudio fue evaluar los efectos de la utilización de vídeo educativo para promover el apego entre madre seropositiva para VIH y su hijo después de seis meses de interacción entre ambos. Estudio descriptivo, exploratorio, con enfoque cualitativo. Los datos fueron recolectados a través de entrevista semiestructurada. La muestra fue intencional. El plan de análisis siguió los siguientes pasos: formación del corpus, definición de las unidades de análisis, clasificación, análisis de los datos por categorías. A pesar de las dificultades y limitaciones impuestas por el VIH/SIDA, por su tratamiento e inseguridad en la vida cotidiana, las madres con VIH desarrollaron estrategias que permitieron vivir y lidiar mejor con la enfermedad, pero sin olvidar de su papel de madre

Protective Effect of Cashew Gum (<i>Anacardium occidentale</i> L.) on 5-Fluorouracil-Induced Intestinal Mucositis

Intestinal mucositis is a common complication associated with 5-fluorouracil (5-FU), a chemotherapeutic agent used for cancer treatment. Cashew gum (CG) has been reported as a potent anti-inflammatory agent. In the present study, we aimed to evaluate the effect of CG extracted from the exudate of Anacardium occidentale L. on experimental intestinal mucositis induced by 5-FU. Swiss mice were randomly divided into seven groups: Saline, 5-FU, CG 30, CG 60, CG 90, Celecoxib (CLX), and CLX + CG 90 groups. The weight of mice was measured daily. After treatment, the animals were euthanized and segments of the small intestine were collected to evaluate histopathological alterations (morphometric analysis), levels of malondialdehyde (MDA), myeloperoxidase (MPO), and glutathione (GSH), and immunohistochemical analysis of interleukin 1 beta (IL-1&#946;) and cyclooxygenase-2 (COX-2). 5-FU induced intense weight loss and reduction in villus height compared to the saline group. CG 90 prevented 5-FU-induced histopathological changes and decreased oxidative stress through decrease of MDA levels and increase of GSH concentration. CG attenuated inflammatory process by decreasing MPO activity, intestinal mastocytosis, and COX-2 expression. Our findings suggest that CG at a concentration of 90 mg/kg reverses the effects of 5-FU-induced intestinal mucositis
