Investigation of Noise Field and Velocity Profiles of an Afterburning Engine
Sound pressure levels, frequency spectrum, and jet velocity profiles are presented for an engine-afterburner combination at various values of afterburner fuel - air ratio. At the high fuel-air ratios, severe low-frequency resonance was encountered which represented more than half the total energy in the sound spectrum. At similar thrust conditions, lower sound pressure levels were obtained from a current fighter air craft with a different afterburner configuration. The lower sound pressure levels are attributed to resonance-free afterburner operation and thereby indicate the importance of acoustic considerations in afterburner design
Children’s experiences of domestic violence and abuse: siblings’ accounts of relational coping
This article explores how young people see their relationships, particularly their sibling relationships, in families affected by domestic violence, and how relationality emerges in their accounts as a resource to build an agentic sense of self. The ‘voice’ of children is largely absent from domestic violence literature, which typically portrays them as passive, damaged and relationally incompetent. Children’s own understandings of their relational worlds are often overlooked, and consequently existing models of children’s social interactions give inadequate accounts of their meaning-making-in-context. Drawn from a larger study of children’s experiences of domestic violence and abuse, this paper uses two case studies of sibling relationships to explore young people’s use of relational resources, for coping with violence in the home. The paper explores how relationality and coping intertwine in young people’s accounts, and disrupts the taken for granted assumption that children’s ‘premature caring’ or ‘parentification’ is (only) pathological in children’s responses to domestic violence. This has implications for understanding young people’s experiences in the present, and supporting their capacity for relationship building in the future
The expression of P-glycoprotein does influence the distribution of novel fluorescent compounds in solid tumour models
Solid tumours display a complex drug resistance phenotype that involves inherent and acquired mechanisms. Multicellular resistance is an inherent feature of solid tumours and is known to present significant barriers to drug permeation in tumours. Given this barrier, do acquired resistance mechanisms such as P-glycoprotein (P-gp) contribute significantly to resistance? To address this question, the multicellular tumour spheroid (MCTS) model was used to examine the influence of P-gp on drug distribution in solid tissue. Tumour spheroids (TS) were generated from either drug-sensitive MCF7WT cells or a drug-resistant, P-gp-expressing derivative MCF7Adr. Confocal microscopy was used to measure time courses and distribution patterns of three fluorescent compounds; calcein-AM, rhodamine123 and BODIPY-taxol. These compounds were chosen because they are all substrates for P-gp-mediated transport, exhibit high fluorescence and are chemically dissimilar. For example, BODIPY-taxol and rhodamine 123 showed high accumulation and distributed extensively throughout the TSWT, whereas calcein-AM accumulation was restricted to the outermost layers. The presence of P-gp in TSAdr resulted in negligible accumulation, regardless of the compound. Moreover, the inhibition of P-gp by nicardipine restored intracellular accumulation and distribution patterns to levels observed in TSWT. The results demonstrate the effectiveness of P-gp in modulating drug distribution in solid tumour models. However, the penetration of agents throughout the tissue is strongly determined by the physico-chemical properties of the individual compounds
A questionnaire to identify patellofemoral pain in the community: an exploration of measurement properties
Background
Community-based studies of patellofemoral pain (PFP) need a questionnaire tool that discriminates between those with and those without the condition. To overcome these issues, we have designed a self-report questionnaire which aims to identify people with PFP in the community.
Methods
Study designs: comparative study and cross-sectional study.
Study population: comparative study: PFP patients, soft-tissue injury patients and adults without knee problems. Cross-sectional study: adults attending a science festival.
Intervention: comparative study participants completed the questionnaire at baseline and two weeks later. Cross-sectional study participants completed the questionnaire once.
The optimal scoring system and threshold was explored using receiver operating characteristic curves, test-retest reliability using Cohen’s kappa and measurement error using Bland-Altman plots and standard error of measurement. Known-group validity was explored by comparing PFP prevalence between genders and age groups.
Results
Eighty-four participants were recruited to the comparative study. The receiver operating characteristic curves suggested limiting the questionnaire to the clinical features and knee pain map sections (AUC 0.97 95 % CI 0.94 to 1.00). This combination had high sensitivity and specificity (over 90 %). Measurement error was less than the mean difference between the groups. Test–retest reliability estimates suggest good agreement (N = 51, k = 0.74, 95 % CI 0.52–0.91). The cross-sectional study (N = 110) showed expected differences between genders and age groups but these were not statistically significant.
Conclusion
A shortened version of the questionnaire, based on clinical features and a knee pain map, has good measurement properties. Further work is needed to validate the questionnaire in community samples
Field dependence of the vortex core size in a multi-band superconductor
The magnetic field dependence of the vortex core size in the multi-band
superconductor NbSe2 has been determined from muon spin rotation measurements.
The spatially extended nature of the quasiparticle core states associated with
the smaller gap leads to a rapid field-induced shrinkage of the core size at
low fields, while the more tightly bound nature of the states associated with
the larger gap leads to a field-independent core size for fields greater than 4
kOe. A simple model is proposed for the density of delocalized core states that
establishes a direct relationship between the field-induced reduction of the
vortex core size and the corresponding enhancement of the electronic thermal
conductivity. We show that this model accurately describes both NbSe2 and the

An improved method for surface immobilisation of RNA: application to small Non-Coding RNA - mRNA pairing
Characterisation of RNA and its intermolecular interactions is increasing in importance as the inventory of known RNA functions continues to expand. RNA-RNA interactions are central to post-transcriptional gene regulation mechanisms in bacteria, and the interactions of bacterial small non-coding RNAs (sRNAs) with their mRNA targets are the subject of much current research. The technology of surface plasmon resonance (SPR) is an attractive approach to studying these interactions since it is highly sensitive, and allows interaction measurements to be recorded in real-time. Whilst a number of approaches exist to label RNAs for surface-immobilisation, the method documented here is simple, quick, efficient, and utilises the high-affinity streptavidin-biotin interaction. Specifically, we ligate a biotinylated nucleotide to the 3' end of RNA using T4 RNA ligase. Although this is a previously recognised approach, we have optimised the method by our discovery that the incorporation of four or more adenine nucleotides at the 3' end of the RNA (a poly-A-tail) is required in order to achieve high ligation efficiencies. We use this method within the context of investigating small non-coding RNA (sRNA)-mRNA interactions through the application of surface technologies, including quantitative SPR assays. We first focus on validating the method using the recently characterised Escherichia coli sRNA-mRNA pair, MicA-ompA, specifically demonstrating that the addition of the poly-A-tail to either RNA does not affect its subsequent binding interactions with partner molecules. We then apply this method to investigate the novel interactions of a Vibrio cholerae Qrr sRNA with partner mRNAs, hapR and vca0939; RNA-RNA pairings that are important in mediating pathogenic virulence. Are tibial angles measured with inertial sensors useful surrogates for frontal plane projection angles using 2-dimensional video analysis during single leg squat tasks? A reliability and agreement analysis in elite football (soccer) players
During single leg squats (SLS), tibial angle (TA) quantification using inertial measurement units (IMU) may offer
a practical alternative to frontal plane projection angle (FPPA) measurement using 2-dimensional (2D) video
analysis. This study determined: (i) the reliability of IMUs and 2D video analysis for TA measurement, and 2D
video analysis for FPPA measurement; (ii) the agreement between IMU TA and both 2D video TA and FPPA
measurements during single leg squats in elite footballers. 18 players were tested on consecutive days. Absolute
TA (ATA) and relative TA (RTA) were measured with IMUs. ATA and FPPA were measured concurrently using
2D video analysis. Within-session reliability for all measurements varied across days (intraclass correlation
coefficient (ICC) range=0.27–0.83, standard error of measurement (SEM) range=2.12–6.23°, minimal detectable
change (MDC) range=5.87–17.26°). Between-sessions, ATA reliability was good for both systems
(ICCs=0.70–0.74, SEMs=1.64–7.53°, MDCs=4.55–7.01°), while IMU RTA and 2D FPPA reliability ranged
from poor to good (ICCs=0.39–0.72, SEMs=2.60–5.99°, MDCs=7.20–16.61°). All limits of agreement exceeded
a 5° acceptability threshold. Both systems were reliable for between-session ATA, although agreement
was poor. IMU RTA and 2D video FPPA reliability was variable. For SLS assessment, IMU derived TAs are not
useful surrogates for 2D video FPPA measures in this population
