The combined approach of specific surface area (SSA), porosity, microprobe analysis (EMPA), transmission electron microscopy (TEM), scanning electron microscopy (SEM) equipped with EDX and infrared spectroscopy (FTIR) provided the mica mineral physico-chemical and morphological characterisation. The electrostatic surface properties were assessed through the determination of the Point of Zero Charge (pHPZC) by the drift method and the electrokinetic mica mineral features represented by the Isoelectric Point (pHIEP) which was carried out through zeta potential measurements. Adsorption tests were performed to correlate the surface charge behaviour of the mica mineral and its influence on the adsorption efficiency of two different dyes, namely: Safranin Orange (SO), as a cationic dye and Reactive Black 5 (RB5), as an anionic dye. The higher adsorption capacity SO dye was observed at pH 9 and achieved almost 83% of removal, while RB5 dye adsorption on mica surface had the highest result, about 45% of removal efficiency, on pH of 3. In both cases, the main mechanism identified that drove this results is the electrostatic force of attraction between the adsorbent edge surface charge (pHdependent) and the ionic nature (anionic or cationic) of the pollutant dyes particles. The preliminary adsorption experiments demonstrated that the raw grounded mica mineral has a greater potential associated with its application on cationic dye removal in wastewater. The present study aimed to detail the main characteristics of the mica mineral in order to evaluate the potential use of such mineral residues in the removal efficiency of contaminated wastewaterinfo:eu-repo/semantics/publishedVersio

    Do inflammasome impact COVID-19 severity?

COVID-19 pandemic has proven to be a dramatic challenge, introducing huge clinical differences that demand extensive investigations. Severe and critical patients may present coagulopathies and microthrombi, which results in varied complications, or acute respiratory distress syndrome that leads to fatality. Although the lung to be the major site of clinical manifestations, COVID-19 has shown extrapulmonary manifestations, especially on the heart and kidney, directly linked to worse disease outcomes. According to the fast-moving of clinical description and scientific publications, the injuries in multiple organs and systemic inflammation appear to be caused by a deregulated immune response, and the NLRP3 inflammasome could be a relevant influencer in this imbalance. However, until now, the precise drivers of the pathophysiology of these injuries remain unknown. In this review, we discuss how inflammasome seems to be directly involved in the clinical profile of patients infected with SARS-CoV-2 and shed light on the mechanisms that lead to fatality. This research was supported by Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES), Brazil.Scopu

Effects of Ouabain in Ehrlich Tumor Development in vitro and in vivo

    No full text
Ouabain (OUA) is a cardiotonic steroid with an immunomodulatory and anti-inflammatory role in different experimental models. Currently, the potential antineoplastic effect of OUA has been studied, however, research is needed to better understand OUA role during tumor development. Therefore, our aim was to investigate the OUA effects on Ehrlich tumor (ET) development in vitro and in vivo. To evaluate the cytotoxic effects of OUA on ET in vitro the cells were incubated with different concentrations of OUA during 24h and 48h and our results showed that only the [1000 ?M] decreased the number and viability of ET cells in the two analyzed times. To study the OUA effects on ET in vivo, Swiss mice were pretreated with 0.56 mg/kg of OUA intraperitoneally (i.p.) for three consecutive days. To develop ET in the solid form, one hour after the last day of pretreatment, ET cells were inoculated subcutaneously into the footpad and the animals were monitored for 13 days. To develop the ascitic form, ET cells were inoculated (i.p.) and the animals were monitored for 3 days. OUA was able to reduce the thickness and weight of the tumor paw, in addition to reduce the weight of the popliteal lymph node. In the ascitic tumor, OUA reduced the number of neutrophils and macrophages and increased the lymphocytes in the peritoneum. Thus, we demonstrated that OUA affects ET development both in vitro and in vivo. Our results suggest a new perspective in the anti-inflammatory, immunomodulatory and a possible anti-cancer role of ouabain and brings new concepts about the pathophysiological role of this substance

Clinical manifestations of intermediate allele carriers in Huntington disease

    No full text
Objective: There is controversy about the clinical consequences of intermediate alleles (IAs) in Huntington disease (HD). The main objective of this study was to establish the clinical manifestations of IA carriers for a prospective, international, European HD registry. Methods: We assessed a cohort of participants at risk with <36 CAG repeats of the huntingtin (HTT) gene. Outcome measures were the Unified Huntington's Disease Rating Scale (UHDRS) motor, cognitive, and behavior domains, Total Functional Capacity (TFC), and quality of life (Short Form-36 [SF-36]). This cohort was subdivided into IA carriers (27-35 CAG) and controls (<27 CAG) and younger vs older participants. IA carriers and controls were compared for sociodemographic, environmental, and outcome measures. We used regression analysis to estimate the association of age and CAG repeats on the UHDRS scores. Results: Of 12,190 participants, 657 (5.38%) with <36 CAG repeats were identified: 76 IA carriers (11.56%) and 581 controls (88.44%). After correcting for multiple comparisons, at baseline, we found no significant differences between IA carriers and controls for total UHDRS motor, SF-36, behavioral, cognitive, or TFC scores. However, older participants with IAs had higher chorea scores compared to controls (p 0.001). Linear regression analysis showed that aging was the most contributing factor to increased UHDRS motor scores (p 0.002). On the other hand, 1-year follow-up data analysis showed IA carriers had greater cognitive decline compared to controls (p 0.002). Conclusions: Although aging worsened the UHDRS scores independently of the genetic status, IAs might confer a late-onset abnormal motor and cognitive phenotype. These results might have important implications for genetic counseling. ClinicalTrials.gov identifier: NCT01590589
