    Estágios curriculares para o ensino médio profissonalizante

Outro benefício para o aluno é a identificação mais clara das opções para a organização de seu perfil de profissionalização. Pelo projeto “Práticas de análise de alimentos: consolidando o saber fazer para o ensino médio profissionalizante”, são ofertadas semestralmente, vagas para estágio curricular para alunos de cursos técnicos da área de alimentos. Os estudantes são inseridos nas atividades que estão em curso nos laboratórios do Departamento de Engenharia de Alimentos da Universidade Estadual de Ponta Grossa, acompanhados pelos acadêmicos estagiários do projeto e supervisionados pelas professoras. No primeiro semestre de 2017 foram ofertadas seis vagas para estágio. Os estagiários do projeto receberam treinamento técnico para acompanhar os alunos no desenvolvimento das atividades. Os estagiários, acadêmicos e a coordenação de estágios dos cursos profissionalizantes avaliaram positivamente o projeto. Os alunos reconheceram a importância da oportunidade de estagiar na universidade ressaltando que todas as atividades estavam de acordo com a sua formação escolar e foram importantes para a complementação da formação profissiona

    Computational analysis of the interaction between transcription factors and the predicted secreted proteome of the yeast Kluyveromyces lactis

    <p>Abstract</p> <p>Background</p> <p>Protein secretion is a cell translocation process of major biological and technological significance. The secretion and downstream processing of proteins by recombinant cells is of great commercial interest. The yeast <it>Kluyveromyces lactis </it>is considered a promising host for heterologous protein production. Because yeasts naturally do not secrete as many proteins as filamentous fungi, they can produce secreted recombinant proteins with few contaminants in the medium. An ideal system to address the secretion of a desired protein could be exploited among the native proteins in certain physiological conditions. By applying algorithms to the completed <it>K. lactis </it>genome sequence, such a system could be selected. To this end, we predicted protein subcellular locations and correlated the resulting extracellular secretome with the transcription factors that modulate the cellular response to a particular environmental stimulus.</p> <p>Results</p> <p>To explore the potential <it>Kluyveromyces lactis </it>extracellular secretome, four computational prediction algorithms were applied to 5076 predicted <it>K. lactis </it>proteins from the genome database. SignalP v3 identified 418 proteins with N-terminal signal peptides. From these 418 proteins, the Phobius algorithm predicted that 176 proteins have no transmembrane domains, and the big-PI Predictor identified 150 proteins as having no glycosylphosphatidylinositol (GPI) modification sites. WoLF PSORT predicted that the <it>K. lactis </it>secretome consists of 109 putative proteins, excluding subcellular targeting. The transcription regulators of the putative extracellular proteins were investigated by searching for DNA binding sites in their putative promoters. The conditions to favor expression were obtained by searching Gene Ontology terms and using graph theory.</p> <p>Conclusion</p> <p>A public database of <it>K. lactis </it>secreted proteins and their transcription factors are presented. It consists of 109 ORFs and 23 transcription factors. A graph created from this database shows 134 nodes and 884 edges, suggesting a vast number of relationships to be validated experimentally. Most of the transcription factors are related to responses to stress such as drug, acid and heat resistance, as well as nitrogen limitation, and may be useful for inducing maximal expression of potential extracellular proteins.</p

Diante disto, tem-se observado um crescente interesse pela silvicultura clonal por grandes empresas, investidores e produtores rurais, principalmente decorrente das vantagens advindas deste processo: solução de problemas de doenças, homogeneidade de plantios e produtividade. No que tange a silvicultura clonal de Eucalyptus esta técnica de plantio florestal se destaca por apresentar maior homogeneidade, propiciar controle de pragas e doenças através do plantio de materiais resistentes e maior produtividade, associada ao plantio de materiais superiores selecionados. Esta técnica ganhou força nos últimos anos pelo fato do setor industrial buscar uma forma de suprimento de matérias primas de qualidade, especialmente relacionada à uniformização desta e em quantidades suficientes que não comprometa suas produções. Este tema encontra-se em constantes discussões sendo amplamente difundido e aplicado pelas empresas do setor florestal, desta forma, este estudo tem por objetivo analisar e compreender a importância da silvicultura clonal de Eucaliptus para este setor através de pesquisas bibliográficas. Serão apresentados estudos de casos a respeito da situação de algumas empresas florestais com o objetivo de ilustrar o tema discutido ao longo do trabalho.     Health education as a strategy to promote the development of the prematurely born child: perception of caregivers

    The birth of a premature baby can cause impairment in the child's development, requiring a critical analysis from the health professional of their performance in the care directed to this public and their family. There is lack of scientific literature focused on the promotion of functional development of premature infants, making necessary the use of technologies that help the family in home care. Objective: To understand the knowledge and practices of caregivers of prematurely children, about the functional development mediated by an intervention with an educational technology. This was a qualitative, descriptive, exploratory and intervention research, with 11 mothers of prematurely born children, followed up by a service in a city of Minas Gerais. Performed in three steps. The first still in the follow up service, the second and third consisted of home visits. The analysis originated the categories “Expectations related to the educational material”, “Experiences with the educational material” and “Changes in care aroused by the appropriation of the educational material”. The work of the health educator articulated with an educational technology focused on promoting the child development of premature children contributes to the resignification of knowledge and everyday practices of care directed to this public

    A insegurança do cistema como impasse para a criminalização da homotransfobia no Rio de Janeiro

    The criminalization of homotransphobia represents an advance for the LGBTI+ agenda, however, the protection of those who are victims of such violence is limited to a certain profile and ignores relevant social realities that make it impossible to achieve its goals. Based on data from crime reports collected by the Civil Police of Rio de Janeiro, from Instituto de Segurança Pública, Rio Sem Homofobia and Grupo Arco Íris, which were systematized by Aliança Nacional LGBTI, the research seeks to understand some gaps left by the application of the law that criminalizes racism to these cases, as well as the still existing impasses for the effective protection of LGBTI+ people by public agents. A socio-legal approach is used, based on queer, decolonial and transfeminist theory. Keywords: Homotransphobia. Criminalization. Cystem.La criminalización de la homotransfobia representa un avance para la agenda LGBTI+, sin embargo, la protección de quienes son víctimas de tal violencia se limita a un determinado perfil e ignora realidades sociales relevantes que imposibilitan el logro de sus objetivos. Desde los datos de las denuncias policiales recogidas por la Policía Civil de Río de Janeiro, Instituto de Seguridad Pública, Rio Sem Homofobia y Grupo Arco Íris, que fueron sistematizados por la Aliança Nacional LGBTI, se busca comprender las brechas dejadas por la aplicación de la ley que criminaliza el racismo a estos casos, así como los impasses aún existentes para la protección de personas LGBTI+ por parte de los agentes públicos.     El grupo operativo como instrumento de aprendizaje del cuidado por madres de hijos con deficiencia

    Objective: To understand the meaning attached to the operative group technique by mothers that take care of children with disabilities. Methods: Qualitative research with eight mothers who participate in operative groups, whose children are treated at a specialized program in Minas Gerais. Data were collected through interviews with discursive questions, in 2013, and analyzed using content analysis, interpreted in light of Pichon-Rivière. Results: The study led to the creation of the categories "Embracement space, exchanges and new meanings" and "Inducing strategy for caring for yourself and your child". The results reveal the importance of the operative group in the daily life of the participants, which highlights the contributions that such educational practice brings to the care experience of these mothers. Conclusion: The operative group was a care learning tool, which suggests the need for health services to invest in offering group activities for caregivers of people with disabilities.Objetivo: Compreender os significados atribuídos ao grupo operativo por mães que cuidam do filho com deficiência. Métodos: Pesquisa de abordagem qualitativa, com oito mães participantes de grupos operativos, cujos filhos são acompanhados em um programa especializado, de Minas Gerais. Os dados foram coletados, em 2013, por meio de entrevista com questões abertas, e submetidos à análise de conteúdo, sendo interpretados à luz de Pichon-Rivière. Resultados: Da análise, emergiram as categorias "Espaço de acolhimento, trocas e ressignificações" e "Estratégia indutora do cuidado de si e do filho". Os resultados trazem à tona a importância do grupo operativo no cotidiano das participantes, o que evidencia o valor que esta atividade educativa agrega à experiência de cuidado dessas mães. Conclusão: O grupo operativo constituiu um instrumento de aprendizagem de cuidado, o que sinaliza a necessidade de os serviços de saúde investirem na oferta de atividades grupais destinadas aos cuidadores de pessoas com deficiência

    Zika virus disrupts gene expression in human myoblasts and myotubes: Relationship with susceptibility to infection

    The tropism of Zika virus (ZIKV) has been described in the nervous system, blood, placenta, thymus, and skeletal muscle. We investigated the mechanisms of skeletal muscle susceptibility to ZIKV using an in vitro model of human skeletal muscle myogenesis, in which myoblasts differentiate into myotubes. Myoblasts were permissive to ZIKV infection, generating productive viral particles, while myotubes controlled ZIKV replication. To investigate the underlying mechanisms, we used gene expression profiling. First, we assessed gene changes in myotubes compared with myoblasts in the model without infection. As expected, we observed an increase in genes and pathways related to the contractile muscle system in the myotubes, a reduction in processes linked to proliferation, migration and cytokine production, among others, confirming the myogenic capacity of our system in vitro. A comparison between non-infected and infected myoblasts revealed more than 500 differentially expressed genes (DEGs). In contrast, infected myotubes showed almost 2,000 DEGs, among which we detected genes and pathways highly or exclusively expressed in myotubes, including those related to antiviral and innate immune responses. Such gene modulation could explain our findings showing that ZIKV also invades myotubes but does not replicate in these differentiated cells. In conclusion, we showed that ZIKV largely (but differentially) disrupts gene expression in human myoblasts and myotubes. Identifying genes involved in myotube resistance can shed light on potential antiviral mechanisms against ZIKV infection

    Revisiting the Soybean GmNAC Superfamily

    The NAC (NAM, ATAF, and CUC) genes encode transcription factors involved with the control of plant morph-physiology and stress responses. The release of the last soybean (Glycine max) genome assembly (Wm82.a2.v1) raised the possibility that new NAC genes would be present in the soybean genome. Here, we interrogated the last version of the soybean genome against a conserved NAC domain structure. Our analysis identified 32 putative novel NAC genes, updating the superfamily to 180 gene members. We also organized the genes in 15 phylogenetic subfamilies, which showed a perfect correlation among sequence conservation, expression profile, and function of orthologous Arabidopsis thaliana genes and NAC soybean genes. To validate our in silico analyses, we monitored the stress-mediated gene expression profiles of eight new NAC-genes by qRT-PCR and monitored the GmNAC senescence-associated genes by RNA-seq. Among ER stress, osmotic stress and salicylic acid treatment, all the novel tested GmNAC genes responded to at least one type of stress, displaying a complex expression profile under different kinetics and extension of the response. Furthermore, we showed that 40% of the GmNACs were differentially regulated by natural leaf senescence, including eight (8) newly identified GmNACs. The developmental and stress-responsive expression profiles of the novel NAC genes fitted perfectly with their phylogenetic subfamily. Finally, we examined two uncharacterized senescence-associated proteins, GmNAC065 and GmNAC085, and a novel, previously unidentified, NAC protein, GmNAC177, and showed that they are nuclear localized, and except for GmNAC065, they display transactivation activity in yeast. Consistent with a role in leaf senescence, transient expression of GmNAC065 and GmNAC085 induces the appearance of hallmarks of leaf senescence, including chlorophyll loss, leaf yellowing, lipid peroxidation and accumulation of H2O2. GmNAC177 was clustered to an uncharacterized subfamily but in close proximity to the TIP subfamily. Accordingly, it was rapidly induced by ER stress and by salicylic acid under late kinetic response and promoted cell death in planta. Collectively, our data further substantiated the notion that the GmNAC genes display functional and expression profiles consistent with their phylogenetic relatedness and established a complete framework of the soybean NAC superfamily as a foundation for future analyses
