    Use of psychoactive substances by adolescents: current panorama

    Adolescence is a period of vulnerability to substance use disorders (SUDs). Epidemiological studies indicate that about 23% of Brazilian adolescents use drugs, with alcohol being the most widely consumed substance. The etiology of SUDs is complex, influenced by an interaction of genetic risk, individual development, environmental factors, context of use, and substance used. Clinicians should consider diagnostic criteria and be aware of behavioral changes that may indicate drug use and its consequences in various aspects of adolescent life. Identification and treatment of comorbid conditions is critical to the management of SUDs in this age group. Interventions should restrict access to drugs and facilitate prompt recognition of initial use, preventing progression to serious patterns of abuse or dependence.     Differential Evolution Algorithm for Identification of Structural Damage in Steel Beams

    Problems involving errors and uncertainties from the use of numerical and experimental responses of beams using optimization processes have been studied by many researchers. In this field, to simulate the real behavior of structures, especially in problems involving damage, it is required to have reliable experimental results in order to adjust a numerical model. These difficulties may be associated for example to modeling the connection stiffness, support conditions, or relevant parameters in structures involving damages among others. This paper proposes a new methodology to detect damage in steel beams using the Differential Evolution Technique based on experimental and numerical data. The results show a great potential of the methodology to solve damage detection problems

    Epitaxial Self-Assembly of Interfaces of 2D Metal–Organic Frameworks for Electroanalytical Detection of Neurotransmitters

    This paper identifies the electrochemical properties of individual facets of anisotropic layered conductive metal–organic frameworks (MOFs) based on M3(2,3,6,7,10,11-hexahydroxytriphenylene)2 (M3(HHTP)2) (M = Co, Ni). The electroanalytical advantages of each facet are then applied toward the electrochemical detection of neurochemicals. By employing epitaxially controlled deposition of M3(HHTP)2 MOFs on electrodes, the contribution of the basal plane ({001} facets) and edge sites ({100} facets) of these MOFs can be individually determined using electrochemical characterization techniques. Despite having a lower observed heterogeneous electron transfer rate constant, the {001} facets of the M3(HHTP)2 systems prove more selective and sensitive for the detection of dopamine than the {100} facets of the same MOF, with the limit of detection (LOD) of 9.9 ± 2 nM in phosphate-buffered saline and 214 ± 48 nM in a simulated cerebrospinal fluid. Langmuir isotherm studies accompanied by all-atom MD simulations suggested that the observed improvement in performance and selectivity is related to the adsorption characteristics of analytes on the basal plane versus edge sites of the MOF interfaces. This work establishes that the distinct crystallographic facets of 2D MOFs can be used to control the fundamental interactions between analyte and electrode, leading to tunable electrochemical properties by controlling their preferential orientation through self-assembly

    Padrões de controle de crises em pacientes com epilepsia de lobo temporal com ou sem esclerose hipocampal

    Objective Patients with mesial temporal lobe epilepsy (MTLE) may present unstable pattern of seizures. We aimed to evaluate the occurrence of relapse-remitting seizures in MTLE with (MTLE-HS) and without (MTLE-NL) hippocampal sclerosis. Method We evaluated 172 patients with MTLE-HS (122) or MTLE-NL (50). Relapse-remitting pattern was defined as periods longer than two years of seizure-freedom intercalated with seizure recurrence. Infrequent seizures was considered as up to three seizures per year and frequent seizures as any period of seizures higher than that. Results Thirty-seven (30%) MTLE-HS and 18 (36%) MTLE-NL patients had relapse-remitting pattern (X2, p = 0.470). This was more common in those with infrequent seizures (X2, p < 0.001). Twelve MTLE-HS and one MTLE-NL patients had prolonged seizure remission between the first and second decade of life (X2, p = 0.06). Conclusion Similar proportion of MTLE-HS or MTLE-NL patients present relapse-remitting seizures and this occurs more often in those with infrequent seizures.Patients with mesial temporal lobe epilepsy (MTLE) may present unstable pattern of seizures.     Statistical Analysis in Database of Offshore Naturally Flowing Wells with Abnormal Events

    The development of statistical analysis in the oil and gas industry database represents the importance of seeking improvements in safety and preventing undesirable events. For accident prevention, which may cause damage to the environment and financial losses, fault detection is essential. Modern techniques on the data-driven, such as data analysis and artificial intelligence are currently considered the best options for this purpose. Regardless of advances in techniques for database manipulation, scientists spend an amount of time working on data quality improvement. Thus, the high dimensionality of data introduces computational and statistical challenges such as acquisition, treatment, processing and interpretation of data. In this paper, we used the 3W public dataset published by Vargas et al. (2019), provided by Petróleo Brasileiro S.A. (Petrobras), which contains 8 variables such as pressure, temperature and flow rate in the process of offshore natural flow wells. The database was evaluated by data exploration and transformation that enables statistical analysis. The relationship between the variables was verified using histograms, Spearman's correlation and Principal Component Analysis (PCA). The results showed that at least one variable should be removed and others should be filled in order to complete the database. The analysis, also, revealed that the variables do not follow a normal distribution, and the variables importance rank. Thereby, it was possible to reach a database with useful format

    Test-retest reproducibility of a multi-atlas automated segmentation tool on multimodality brain MRI

    The increasing use of large sample sizes for population and personalized medicine requires high-throughput tools for imaging processing that can handle large amounts of data with diverse image modalities, perform a biologically meaningful information reduction, and result in comprehensive quantification. Exploring the reproducibility of these tools reveals the specific strengths and weaknesses that heavily influence the interpretation of results, contributing to transparence in science. We tested-retested the reproducibility of MRICloud, a free automated method for whole-brain, multimodal MRI segmentation and quantification, on two public, independent datasets of healthy adults. Results The reproducibility was extremely high for T1-volumetric analysis, high for diffusion tensor images (DTI) (however, regionally variable), and low for resting-state fMRI. Conclusion In general, the reproducibility of the different modalities was slightly superior to that of widely used software.     Exames de tomografia computadorizada e cintilografia para diagnóstico e plano de tratamento de osteocondroma condilar: relato de caso

    PURPOSE: To report an uncommon case of osteochondroma affecting the mandibular condyle of a young patient and to illustrate the important contributions of different imaging resources to the diagnosis and treatment planning of this lesion. CASE DESCRIPTION: A 24-year-old female patient with the chief complaint of an increasing facial asymmetry and pain in the left pre-auricular region, revealing a reduced mouth opening, mandibular deviation and posterior cross-bite over a period of 18 months. Panoramic radiography revealed an enlargement of the left condyle, whereas computed tomography (CT) sections and three-dimensional CT showed a well-defined bone growth arising from condylar neck. The scintigraphy exam showed an abnormal osteogenic activity in the left temporomandibular joint. The condyle was surgically removed and after 18 months follow-up the panoramic radiography and CT scans showed no signs of recurrence. CONCLUSION: Although osteochondroma is a benign bone tumor that rarely arises in cranial and maxillofacial region, it should be considered in the differential diagnosis of slow-growing masses of the temporomandibular area and the use of different imaging exams significantly contribute to the correct diagnosis and treatment planning of this pathological condition.OBJETIVO: Relatar um caso incomum de osteocondroma afetando o côndilo mandibular de uma paciente jovem e ilustrar as importantes contribuições dos diferentes exames por imagem para o diagnóstico e plano de tratamento desta lesão. DESCRIÇÃO DO CASO: Paciente do sexo feminino, 24 anos, com queixa principal de uma crescente assimetria facial e dor na região pré-auricular esquerda, além de apresentar redução da abertura bucal, desvio de mandíbula e mordida cruzada posterior, por um período de 18 meses. A radiografia panorâmica revelou um aumento do côndilo esquerdo, enquanto secções de tomografia computadorizada e reconstrução 3D exibiram um crescimento ósseo bem definido originando-se do côndilo. O exame de cintilografia revelou uma atividade osteogênica anormal na região de articulação tempormandibular esquerda. O côndilo foi cirurgicamente removido e após 18 meses de acompanhamento a radiografia panorâmica e a tomografia computadorizada não revelaram nenhum sinal de recorrência. CONCLUSÃO: Apesar de o osteocondroma ser um tumor ósseo benigno que raramente surge na região crânio maxilofacial, ele deve ser considerado como diagnóstico diferencial para massas de crescimento lento na área temporomandibular e o uso de diferentes exames por imagem contribuem significativamente para o correto diagnóstico e plano de tratamento desta condição patológica

    The phase separation-dependent FUS interactome reveals nuclear and cytoplasmic function of liquid–liquid phase separation

    Liquid–liquid phase separation (LLPS) of proteins and RNAs has emerged as the driving force underlying the formation of membrane-less organelles. Such biomolecular condensates have various biological functions and have been linked to disease. The protein Fused in Sarcoma (FUS) undergoes LLPS and mutations in FUS have been causally linked to the motor neuron disease Amyotrophic Lateral Sclerosis (ALS-FUS). LLPS followed by aggregation of cytoplasmic FUS has been proposed to be a crucial disease mechanism. However, it is currently unclear how LLPS impacts the behaviour of FUS in cells, e.g. its interactome. Hence, we developed a method allowing for the purification of LLPS FUS-containing droplets from cell lysates. We observe substantial alterations in the interactome, depending on its biophysical state. While non-LLPS FUS interacts mainly with factors involved in pre-mRNA processing, LLPS FUS predominantly binds to proteins involved in chromatin remodelling and DNA damage repair. Interestingly, also mitochondrial factors are strongly enriched with LLPS FUS, providing a potential explanation for the observed changes in mitochondrial gene expression in mouse models of ALS-FUS. In summary, we present a methodology to investigate the interactomes of phase separating proteins and provide evidence that LLPS shapes the FUS interactome with implications for function and disease
