    Nature’s Lab for Derivatization: New and Revised Structures of a Variety of Streptophenazines Produced by a Sponge-Derived Streptomyces Strain

    Eight streptophenazines (A–H) have been identified so far as products of Streptomyces strain HB202, which was isolated from the sponge Halichondria panicea from the Baltic Sea. The variation of bioactivities based on small structural changes initiated further studies on new derivatives. Three new streptophenazines (I–K) were identified after fermentation in the present study. In addition, revised molecular structures of streptophenazines C, D, F and H are proposed. Streptophenazines G and K exhibited moderate antibacterial activity against the facultative pathogenic bacterium Staphylococcus epidermidis and against Bacillus subtilis. All tested compounds (streptophenazines G, I–K) also showed moderate activities against PDE 4B

    The research on the use of experimental methods in learning characteristics of material and its usefulness toward students\u27 achievements and learning responses of Level 4 primary school students of MIN Tungkob Aceh Besar aims to determine students\u27 learning outcomes and responses toward the use of the method. This study uses experimental research. The data collection techniques were using test and distributing questionnaire to the students. The samples in this experimental study were students of class IV3 totaling 34 people as the experimental class and the class IV4 totaling 31 people as the control class. The data, which is the students\u27 learning outcomes collected from pretest and posttest, were analyzed using t-test formula. The data from the students\u27 questionnaire responses were analyzed using percentage formula.The result was that the students\u27 learning outcomes from the experimental class and the control class showed significant differences. Students\u27 responses toward the use of experimental methods in learning material characteristics and its usefulness were also very positive, where the students are very excited and interested in learning to use the experimental method in the study of material characteristics and its usefulness

    An unusually broad series of seven cyclombandakamines, bridged dimeric naphthylisoquinoline alkaloids from the congolese liana ancistrocladus ealaensis

    A series of seven unusual dimeric naphthylisoquinoline alkaloids was isolated from the leaves of the tropical liana Ancistrocladus ealaensis J. LĂ©onard, named cyclombandakamine A (1), 1-epi-cyclombandakamine A (2), and cyclombandakamines A; 3-7; (3-7). These alkaloids have a chemically thrilling structural array consisting of a twisted dihydrofuran-cyclohexenone-isochromene system. The 1'″-epimer of 4, cyclombandakamine A; 1; (8), had previously been discovered in an unidentified Ancistrocladus species related to A. ealaensis. Both lianas produce the potential parent precursor, mbandakamine A (9), but only A. ealaensis synthesizes the corresponding cyclized form, along with a broad series of slightly modified analogs. The challenging isolation required, besides multi-dimensional chromatography, the use of a pentafluorophenyl stationary phase. Featuring up to six stereocenters and two types of chiral axes, their structures were elucidated by means of 1D and 2D NMR, HRESIMS, in combination with oxidative chemical degradation experiments as well as chiroptical (electronic circular dichroism spectroscopy) and quantum chemical calculations. Compared to the 'open-chain' parent compound 9, these dimers displayed rather moderate antiplasmodial activities

    Biodiversity in Marine Ecosystems—European Developments toward Robust Assessments

    Sustainability of marine ecosystems and their services are dependent on marine biodiversity, which is threatened worldwide. Biodiversity protection is a major target of the EU Marine Strategy Framework Directive, requiring assessment of the status of biodiversity on the level of species, habitats, and ecosystems including genetic diversity and the role of biodiversity in food web functioning and structure. This paper provides a summary of the development of new indicators and refinement of existing ones in order to address some of the observed gaps in indicator availability for marine biodiversity assessments considering genetic, species, habitat, and ecosystem levels. Promising new indicators are available addressing genetic diversity of microbial and benthic communities. Novel indicators to assess biodiversity and food webs associated with habitats formed by keystone species (such as macroalgae) as well as to map benthic habitats (such as biogenic reefs) using high resolution habitat characterization were developed. We also discuss the advances made on indicators for detecting impacts of non-native invasive species and assessing the structure and functioning of marine food-webs. The latter are based on indicators showing the effects of fishing on trophic level and size distribution of fish and elasmobranch communities well as phytoplankton and zooplankton community structure as food web indicators. New and refined indicators are ranked based on quality criteria). Their applicability for various EU and global biodiversity assessments and the need for further development of new indicators and refinement of the existing ones is discussed

    Identification of the Antibacterial Compound Produced by the Marine Epiphytic Bacterium Pseudovibrio sp. D323 and Related Sponge-Associated Bacteria

    Surface-associated marine bacteria often produce secondary metabolites with antagonistic activities. In this study, tropodithietic acid (TDA) was identified to be responsible for the antibacterial activity of the marine epiphytic bacterium Pseudovibrio sp. D323 and related strains. Phenol was also produced by these bacteria but was not directly related to the antibacterial activity. TDA was shown to effectively inhibit a range of marine bacteria from various phylogenetic groups. However TDA-producers themselves were resistant and are likely to possess resistance mechanism preventing autoinhibition. We propose that TDA in isolate D323 and related eukaryote-associated bacteria plays a role in defending the host organism against unwanted microbial colonisation and, possibly, bacterial pathogens

    Ealamines A-H, a Series of Naphthylisoquinolines with the Rare 7,8'-Coupling Site, from the Congolese Liana; Ancistrocladus ealaensis; , Targeting Pancreatic Cancer Cells

    From the twigs and leaves of the Central African liana; Ancistrocladus ealaensis; (Ancistrocladaceae), a series of ten 7,8'-coupled naphthylisoquinoline alkaloids were isolated, comprising eight new compounds, named ealamines A-H (; 4a; ,; 4b; ,; 5; -; 10; ), and two known ones, 6-; O; -demethylancistrobrevine A (; 11; ) and yaoundamine A (; 12; ), which had previously been found in related African; Ancistrocladus; species. Only one of the new compounds within this series, ealamine H (; 10; ), is a typical Ancistrocladaceae-type alkaloid, with 3; S; -configuration at C-3 and an oxygen function at C-6, whereas seven of the new alkaloids are the first 7,8'-linked "hybrid-type" naphthylisoquinoline alkaloids, i.e., 3; R; -configured and 6-oxygenated in the tetrahydroisoquinoline part. The discovery of such a broad series of 7,8'-coupled naphthyltetrahydroisoquinolines is unprecedented, because representatives of this subclass of alkaloids are normally found in Nature quite rarely. The stereostructures of the new ealamines were assigned by HRESIMS, 1D and 2D NMR, oxidative degradation, and experimental and quantum-chemical ECD investigations, and-in the case of ealamine A (; 4a; )-also confirmed by X-ray diffraction analysis. Ealamines A-D exhibited distinct-and specific-antiplasmodial activities, and they displayed pronounced preferential cytotoxic effects toward PANC-1 human pancreatic cancer cells in nutrient-deprived medium, without causing toxicity under normal, nutrient-rich conditions, with ealamine C (; 5; ) as the most potent agent

    Helimeric porphyrinoids: Stereostructure and chiral resolution of meso -tetraarylmorpholinochlorins

    The synthesis and chiral resolution of free-base and Ni(II) complexes of a number of derivatives of meso-tetraphenylmorpholinochlorins, with and without direct ÎČ-carbon-to-o-phenyl linkages to the flanking phenyl groups, is described. The morpholinochlorins, a class of stable chlorin analogues, were synthesized in two to three steps from meso-tetraphenylporphyrin. The conformations and the relative stereostructures of a variety of free-base and Ni(II) complexes of these morpholinochlorins were elucidated by X-ray diffractometry. Steric and stereoelectronic arguments explain the relative stereoarray of the morpholino-substituents, which differ in the free-base and Ni(II) complexes, and in the monoalkoxy, ÎČ-carbon-to-o-phenyl linked morpholinochlorins, and the dialkoxy derivatives. The Ni(II) complexes were all found to be severely ruffled whereas the free-base chromophores are more planar. As a result of the helimeric distortion of their porphyrinoid chromophores, the ruffled macrocycles possess a stable inherent element of chirality. Most significantly, resolution of the racemic mixtures was achieved, both by classical methods via diastereomers and by HPLC on a chiral phase. Full CD spectra were recorded and modeled using quantum-chemical computational methods, permitting, for the first time, an assignment of the absolute configurations of the chromophores. The report expands the range of known pyrrole-modified porphyrins. Beyond this, it introduces large chiral porphyrinoid π-systems that exist in the form of two enantiomeric, stereochemically stable helimers that can be resolved. This forms the basis for possible future applications, for example, in molecular-recognition systems or in materials with chiroptic properties. © 2011 American Chemical Society

    Functionality of Two Origins of Replication in Vibrio cholerae Strains With a Single Chromosome

    Chromosomal inheritance in bacteria usually entails bidirectional replication of a single chromosome from a single origin into two copies and subsequent partitioning of one copy each into daughter cells upon cell division. However, the human pathogen Vibrio cholerae and other Vibrionaceae harbor two chromosomes, a large Chr1 and a small Chr2. Chr1 and Chr2 have different origins, an oriC-type origin and a P1 plasmid-type origin, respectively, driving the replication of respective chromosomes. Recently, we described naturally occurring exceptions to the two-chromosome rule of Vibrionaceae: i.e., Chr1 and Chr2 fused single chromosome V. cholerae strains, NSCV1 and NSCV2, in which both origins of replication are present. Using NSCV1 and NSCV2, here we tested whether two types of origins of replication can function simultaneously on the same chromosome or one or the other origin is silenced. We found that in NSCV1, both origins are active whereas in NSCV2 ori2 is silenced despite the fact that it is functional in an isolated context. The ori2 activity appears to be primarily determined by the copy number of the triggering site, crtS, which in turn is determined by its location with respect to ori1 and ori2 on the fused chromosome

    Germany 2050: For the first time Germany reached a balance between its sources of anthropogenic CO2 to the atmosphere and newly created anthropogenic sinks. This backcasting study presents a fictional future in which this goal was achieved by avoiding (∌645 Mt CO2), reducing (∌50 Mt CO2) and removing (∌60 Mt CO2) carbon emissions. This meant substantial transformation of the energy system, increasing energy efficiency, sector coupling, and electrification, energy storage solutions including synthetic energy carriers, sector-specific solutions for industry, transport, and agriculture, as well as natural-sink enhancement and technological carbon dioxide options. All of the above was necessary to achieve a net-zero CO2 system for Germany by 2050
