    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    Plant communities and their possible exploitation during Late Prehistory are studied based on charcoal data from four archaeological sites. Settlement location in the area appears to have been chosen on the basis of easy access to rivers and good arable land. Querens (deciduous) and Fabaceae appear to have been the main source of wood; this is in agreement with previous data from north-western Portugal. The abundance of Fabaceae, which thrive in open well-lit areas, is seen as a direct consequence of woodland clearance. The co-occurrence of Quercus (deciduous) and Fabaceac appears as a distinctive feature in north-western Portugal during Late Prehistory and Protohistory. Fabaceae have remained a familiar component of the vegetation cover ever since. Pinus pinaster is present as isolat ed individuals, in contrast to its present-day abundance

    PURPOSE: We generated lung morphometry measurements using single-breath diffusion-weighted MRI and three different acinar duct models in healthy participants and patients with emphysema stemming from chronic obstructive lung disease (COPD) and alpha-1 antitrypsin deficiency (AATD). METHODS: Single-breath-inhaled RESULTS: We evaluated 42 participants, including 15 elderly never-smokers (69 ± 5 years), 12 ex-smokers without COPD (67 ± 11 years), 9 COPD ex-smokers (80 ± 6 years), and 6 AATD patients (59 ± 6 years).
