Identification and Characterization of the Transcriptional Regulator ChrB in the Chromate Resistance Determinant of Ochrobactrum tritici 5bvl1
Ochrobactrum tritici 5bvl1 is able to resist to high concentrations of chromate through the expression of an inducible
chromate-resistant determinant, found in a mobile element (TnOtChr), which carries the genes, chrB, chrA, chrC and chrF.
The regulation of chr operon present in TnOtChr, which is controlled by a transcriptional regulator, ChrB, was characterized
in the current work. Fusions of chr promoter, or chr promoter and chrB gene, upstream of a gfp reporter gene, identified the
most probable promoter sequence within the tnpR-chrB intergenic region. This region contains an AT-rich imperfect
inverted repeat sequence, which overlaps a part of the 210 sequence. The results of the in vitro DNA-binding assays with
purified ChrB (His- or no-tagged) showed that the protein binds directly to the chr promoter region. In order to identify the
ChrB functional domain for sensing chromate stress and for DNA-binding, site-directed mutagenesis of ChrB was performed.
Among several single amino acid mutants, three mutants (R180; R187 and H229) prevented chromate induction without any
modification to the protein’s stability. Interestingly, two ChrB mutants (R18 and R23) were constitutively active, regardless of
chromate stress conditions, indicating that the residues most probably belong to the protein-DNA binding site. As such, the
ChrB was classified as a transcriptional regulator that recognizes a specific DNA sequence, regulating the expression of a

Finite element modelling and analysis of crack shape evolution in mode-I fatigue Middle Cracked Tension specimens
A numerical procedure was employed to study the shape evolution of fatigue cracks in Middle Cracked Tension specimens. This iterative procedure consists of a 3D finite element analysis to obtain the displacement field in the cracked body, calculation of stress intensity factors along crack front and definition of local crack advances considering the Paris law. Numerical predictions were compared with experimental crack shapes with a good agreement. The evolution of crack shape was analysed for different propagation conditions considering robust dependent parameters. Two main propagation stages were identified: an initial transient stage highly dependent on initial crack shape and a stable stage where the crack follows preferred paths. Mathematical models were proposed for transient and stable stages consisting of exponential and polynomial functions, respectively. The transition between both stages was defined considering two criteria: the rate of shape variation and the distance to stable shape. Rephasing Invariants of Quark and Lepton Mixing Matrices
Rephasing invariants of quark and lepton mixing matrices are obtained in the
standard model extended by the seesaw mechanism, and in its low-energy
effective theory with the dimension-five Majorana mass operator. We classify
the basic invariants, discuss non-trivial relations between them, and determine
the independent invariants which characterize all the information in the mixing
matrices in a basis-independent way. We also discuss the restrictions on the
allowed ranges for the mixing phases, and on the rephasing invariants, which
follow from a discrete invariance of the Majorana mass matrix.Comment: Some references added, and typos correcte
Jarlskog-like invariants for theories with scalars and fermions
Within the framework of theories where both scalars and fermions are present,
we develop a systematic prescription for the construction of CP-violating
quantities that are invariant under basis transformations of those matter
fields. In theories with Spontaneous Symmetry Breaking, the analysis involves
the vevs' transformation properties under a scalar basis change, with a
considerable simplification of the study of CP violation in the scalar sector.
These techniques are then applied in detail to the two Higgs-doublet model with
quarks. It is shown that there are new invariants involving scalar-fermion
interactions, besides those already derived in previous analyses for the

An expert-based assessment of global threats and conservation measures for spiders
Despite the prominent role of spiders in most ecosystems, these invertebrates are still notably endangered as well as underrepresented in current conservation efforts. We sent a survey to spider experts and enthusiasts belonging to arachnological societies across the globe to determine the general consensus on globally relevant threats to spiders as well as the most relevant conservation measures. We report that respondents found agriculture, livestock farming & forestry, climate change, urbanisation and pollution (including pesticides) to be the most relevant threats to spider species worldwide. Likewise, land protection and education & awareness were considered the most relevant conservation measures to avoid species declines and extinctions. Although these results tend to be consistent across the biogeographic regions of expertise of respondents, there was significant variation between regions. We discuss the support and justification for the patterns found, their regional variations, and the relevance of threats and conservation measures. This is the first global roadmap for spider species conservation action and research. In general, land should be set aside for species protection, agroforestry practices should be carefully considered, climate change should be mitigated, and the general public should be made more aware of spiders, their importance and the threats they face.Peer reviewe

A Complex Case of Cholestasis in a Patient with ABCB4 and ABCB11 Mutations
The low-phospholipid-associated cholelithiasis (LPAC) syndrome is a form of symptomatic cholelithiasis occurring in young adults, characterized by recurrence of symptoms after cholecystectomy and presence of hepatolithiasis. The case refers to a healthy 39-year-old Caucasian male who presented with abdominal pain and jaundice. His blood tests showed conjugated hyperbilirubinemia and elevated liver enzymes (total bilirubin 6.65 mg/dL, γ-glutamyltransferase 699 IU/L) and abdominal computed tomography revealed dilation of common bile duct and left intrahepatic ducts. Magnetic resonance cholangiopancreatography identified choledocholithiasis, retrieved by endoscopic retrograde cholangiopancreatography, after which there was a worsening of jaundice (total bilirubin 23 mg/dL), which persisted for several weeks, possibly due to ciprofloxacin toxicity. After an extensive workup including liver biopsy, the identification of two foci of hepatolithiasis on reevaluation abdominal ultrasound raised the hypothesis of LPAC syndrome and the patient was started on ursodeoxycholic acid, with remarkable improvement. Genetic testing identified the mutation c.1954A>G (p.Arg652Gly) in ABCB4 gene (homozygous) and c.1331T>C (p.Val444Ala) in ABCB11 gene (heterozygous). In conclusion, we describe the unique case of an adult male with choledocholithiasis, hepatolithiasis, and persistent conjugated hyperbilirubinemia after retrieval of stones, fulfilling the criteria for LPAC syndrome and with possible superimposed drug-induced liver injury, in whom ABCB4 and ABCB11 mutations were found, both of which had not been previously described in association with LPAC.info:eu-repo/semantics/publishedVersio
