Experimental characterization of the thermo-poro-mechanical properties of the Aegion Fault gouge
Advances in constitutive modelling of jointed rock hydro mechanical interactions at laboratory scale
Cette étude s’inscrit dans une recherche multi échelles basée sur des expérimentations in situ et des essais de laboratoire. Les essais, injections radiales d’eau au centre de l’éponte inférieure, sont fait à contrainte normale constante de 0 à 110 MPa et pour des pressions de 0 à 4 MPa. Quatre essais hydromécaniques ont été réalisés au laboratoire L3S sur des échantillons de calcaire fracturé (deux diaclases et deux joints de stratification). L’analyse des mesures montre une corrélation entre les variations de la surface de contact et le débit hydraulique dans la fracture. La modélisation hydromécanique réalisée à l’aide du code 3DEC peut être améliorée à partir de l’analyse précédente à travers la modification de la relation entre l’ouverture hydraulique et la fermeture mécanique du joint. L’écart entre les valeurs mesurées et calculées est lié en particulier à la dégradation des aspérités de la fracture sous forte contrainte et à la turbulence des écoulements non prises en compte dans la modélisation
Physical and optical properties of 2010 Eyjafjallajökull volcanic eruption aerosol: ground-based, Lidar and airborne measurements in France
International audienceDuring the Eyjafjallajökull eruption (14 April to 24 May 2010), the volcanic aerosol cloud was observed across Europe by several airborne in situ and ground-based remote-sensing instruments. On 18 and 19 May, layers of depolarizing particles (i.e. non-spherical particles) were detected in the free troposphere above the Puy de Dôme station, (PdD, France) with a Rayleigh-Mie LIDAR emitting at a wavelength of 355 nm, with parallel and crossed polarization channels. These layers in the free troposphere (FT) were also well captured by simulations with the Lagrangian particle dispersion model FLEXPART, which furthermore showed that the ash was eventually entrained into the planetary boundary layer (PBL). Indeed, the ash cloud was then detected and characterized with a comprehensive set of in situ instruments at the Puy de Dôme station (PdD). In agreement with the FLEXPART simulation, up to 65 μg m−3 of particle mass and 2.2 ppb of SO2 were measured at PdD, corresponding to concentrations higher than the 95 percentile of 2 yr of measurements at PdD. Moreover, the number concentration of particles increased to 24 000 cm−3, mainly in the submicronic mode, but a supermicronic mode was also detected with a modal diameter of 2 μm. The resulting optical properties of the ash aerosol were characterized by a low scattering Ångström exponent (0.98), showing the presence of supermicronic particles. For the first time to our knowledge, the combination of in situ optical and physical characterization of the volcanic ash allowed the calculation of the mass-to-extinction ratio (η) with no assumptions on the aerosol density. The mass-to-extinction ratio was found to be significantly different from the background boundary layer aerosol (max: 1.57 g m−2 as opposed to 0.33 ± 0.03 g m−2). Using this ratio, ash mass concentration in the volcanic plume derived from LIDAR measurements was found to be 655 ± 23 μg m−3 when the plume was located in the FT (3000 m above the sea level - a.s.l.). This ratio could also be used to retrieve an aerosol mass concentration of 579 ± 60 μg m−3 on 19 April, when LIDAR observations detected the ash cloud at 3000 m a.s.l. in correspondence with model simulations (FLEXPART). On 22 April, another ash plume entered the BL, and although it was more diluted than during the May episode, the French research aircraft ATR42 that passed over Clermont-Ferrand in the PBL confirmed the presence of particles with a supermicronic mode, again with a modal diameter at 2 μm. This data set combining airborne, ground-based and remote sensing observations with dispersion model simulations shows an overall very good coherence during the volcanic eruption period, which allows a good confidence in the characteristics of the ash particles that can be derived from this unique data set

Client protein recruitment to the Hsp90 system depends on cochaperones that bind the client and Hsp90 simultaneously and facilitate their interaction. Hsp90 involvement in the assembly of snoRNPs, RNA polymerases, PI3-kinase-like kinases, and chromatin remodeling complexes depends on the TTT (Tel2-Tti1-Tti2), and R2TP complexes-consisting of the AAA-ATPases Rvb1 and Rvb2, Tah1 (Spagh/RPAP3 in metazoa), and Pih1 (Pih1D1 in humans)-that together provide the connection to Hsp90. The biochemistry underlying R2TP function is still poorly understood. Pih1 in particular, at the heart of the complex, has not been described at a structural level, nor have the multiple protein-protein interactions it mediates been characterized. Here we present a structural and biochemical analysis of Hsp90-Tah1-Pih1, Hsp90-Spagh, and Pih1D1-Tel2 complexes that reveal a domain in Pih1D1 specific for binding CK2 phosphorylation sites, and together define the structural basis by which the R2TP complex connects the Hsp90 chaperone system to the TTT complex

Vertical Zoning in Marine Protected Areas: Ecological Considerations for Balancing Pelagic Fishing with Conservation of Benthic Communities
Marine protected areas (MPAs), ideally, manage human uses that threaten ecosystems, or components of ecosystems. During several recent MPA designation processes, concerns have arisen over the scientific justification for no-take MPAs, particularly those that restrict recreational fishing for pelagic species. An important question is: under what conditions might recreational pelagic fishing be compatible with the conservation goals of an MPA that is primarily focused on benthic Communities? In 2005, an expert workshop of fisheries biologists, marine ecologists, MPA managers, and recreational fishermen was convened by NOAAs National MPA Center to evaluate the limited empirical data on benthic-pelagic coupling and to help provide practical advice on this topic. The participants (i) proposed a preliminary conceptual framework for addressing vertical zoning, (ii) developed preliminary guidelines to consider when evaluating whether to allow or restrict pelagic fishing in an MPA, and (iii) identified future research priorities for understanding benthic-pelagic Coupling. A Suite of ecological conditions where recreational pelagic fishing may not be compatible with benthic conservation were identified: (1) high relief habitats, (2) depths shallower than 50-100 m (depending upon the specific location), (3) major topographic and oceanographic features, and (4) spawning areas. Similarly, pelagic fishing is not likely to affect benthic communities adversely in many circumstances. Until further scientific study can shed more light on the issue of how benthic-pelagic linkages affect specific conservation targets, the proposed framework in this manuscript provides practical, easily-applied guidance for using vertical zoning to manage fishing in multiple use MPAs that focus on benthic conservation.Areas Marinas Protegidas (AMP) idealmente, administran el uso humano que amenaza los ecosistemas o sus componentes. Durante el actual proceso de declaracion de AMP, han surgido algunas preocupaciones acerca de la justificacion cientifica para establecer areas de no pesca, particularmente aquellas que restringen la pesca recreativa de especies pelagicas. Una pregunta importante es QQQ ?bajo que condiciones la pesca pelagica recreativa es compatible con los objetivos de conservacion de un AMP que se enfoca principalmente en comunidades bentonicas? En 2005, un taller de expertos en biologia pesquera, ecologfa marina, manejo de AMP y Pescadores recreativos fue convocado por el Centra Nacional de AMP de la NOAA para evaluar los pocos datos empiricos del acoplamiento entre los sistemas pelagico y bentonico, y ofrecer asesoria practica sobre el tema. Los participantes (i) propusieron un marco conceptual preliminar para abordar el tema de la zonacion vertical, (ii) desarrollar directrices preliminares para que cuando se haga una evaluation si se permite o restringe la pesca pelagica dentro de la AMP, y (iii) identificar futuras lineas de investigation para comprender mejor el acoplamiento entre el bentos y el sistema pelagico. Se identifico una serie de condiciones ecologicas en las que la pesca recreativa pelagica puede no ser compatible con la conservacion del bentos: (1) habitat de alto relieve, (2) profundidades menores a 50 m-100 m (dependiendo de la zona), (3) caracteristicas oceanograficas y topograficas sobresalientes, y (4) areas de desove. De igual forma, bajo varias circunstancias, la pesca pelagica puede no afectar las comunidades bentonicas. Hasta que los estudios cientificos brinden mas information acerca de como las relaciones entre el bentos y el ambiente pelagico afectan los objetivos especificos de la conservacion, el contexto propuesto en este trabajo provee una guia practica y de facil aplicacion para utilizar la zonacion vertical en el manejo pesquero en varios aspectos de las AMP que se enfocan en la conservacion del bentos.Keywords: Trophic level, Marine protected areas, Pelagic fisheries, Conservatio

Comportement hydromécanique des joints rocheux (expérimentations et modélisation constitutive)
De nombreuses applications en génie civil dans les massifs rocheux fracturés exigent des études de faisabilité précises. Ces massifs peuvent présenter des défauts de différentes tailles. Ils peuvent contenir d'une part des micro fissures induites par la perturbation du champ de contraintes due à la construction d'un l'ouvrage, et d'autre part des fractures naturelles de plusieurs mètres voire plus. Notre travail est d'apporter des informations supplémentaires nouvelles sur le comportement hydromécanique des joints rocheux naturels et sur la modélisation constitutive de leur comportement hydromécanique. L'étude expérimentale a consisté en une série d'essais hydromécaniques de compression simple avec la machine BCR-3D du laboratoire 3S de Grenoble. Nous avons présenté un modèle hydromécanique constitutif original, qui peut être un moyen efficace pour modéliser le comportement hydromécanique des joints rocheux en compression et en cisaillement, en échelle méso sans prendre en compte l'effet d'échelle. La Boîte de Cisaillement direct pour Roches à 3 Dimensions, appelée BCR3D, a servi à la réalisation de nos essais mécaniques et hydromécaniques sur les interfaces entre géomatériaux. Cette boîte de cisaillement direct particulière a un système d'injection hydraulique afin d'étudier le comportement hydromécanique des joints rocheux et aussi de mesurer l'anisotropie hydraulique des écoulements. A l'aide du modèle de Divoux modifié et de l'approche de Darcy sous forme incrémentale, nous avons détaillé un modèle hydromécanique incrémentaI non linéaire original. Nous avons mis au point la formulation de transmissivité hydraulique des joints rocheux selon deux chemins: tout d'abord, l'évolution de transmissivité en fonction de la contrainte normale (et de l'énergie spécifique normale), et ensuite les changements de transmissivité en fonction du chemin de cisaillement (et de l'énergie spécifique tangentielle). La performance du modèle est discutée à travers la comparaison entre des résultats expérimentaux et les modélisations constitutives. Le modèle hydromécanique incrémentaI non linéaire (LHMIN) pour l'interface, permet d'évaluer soit le débit, soit la transmissivité fonction du niveau de contrainte normale, de l'énergie normale, du niveau de contrainte de cisaillement et de l'énergie de cisaillement imposés au joint rocheux. Nous avons vu qu'il est possible de comparer schématiquement par simulation, le débit percolant entre divers géomatériaux au débit percolant à travers la masse de ces géomatériaux s'agissant d'un bouchon de galerie, nous avons présenté l'évolution de la transmissivité en cas de cisaillement pour quatre valeurs différentes de retrait du béton de scellement. Advances in constitutive modelling of jointed roxk hydro mechanical interactions at laboratory scale
National audienceThis study is a part of a wide multi scale research based on in situ experimentations and laboratory tests. The latest, radial water injections in the centre of the lower joint wall are done at constant normal stress from 0 to 110 MPa and at pressures from 0 to 4 MPa. Four hydro mechanical tests have been performed at the L3S laboratory on fractured limestone samples (two diaclase and two bedding plane). The measurement analysis shows a relation between the contact surface variations and the flow values inside the fracture. The hydro mechanical modelling performed using 3DEC code can be improved from the previous analysis through the modification of the relation between the joint hydraulic opening and the joint mechanical closure. The remaining gap between simulations and measurements can be related to fracture asperity degradation and turbulent flow that have not been taken into account in the modelling.Cette étude s'inscrit dans une recherche multi échelles basée sur des expérimentations in situ et des essais de laboratoire.
