    The XXZ spin chain at Δ=−1/2\Delta=- {1/2}: Bethe roots, symmetric functions and determinants

    A number of conjectures have been given recently concerning the connection between the antiferromagnetic XXZ spin chain at Δ=−12\Delta = - \frac12 and various symmetry classes of alternating sign matrices. Here we use the integrability of the XXZ chain to gain further insight into these developments. In doing so we obtain a number of new results using Baxter's QQ function for the XXZ chain for periodic, twisted and open boundary conditions. These include expressions for the elementary symmetric functions evaluated at the groundstate solution of the Bethe roots. In this approach Schur functions play a central role and enable us to derive determinant expressions which appear in certain natural double products over the Bethe roots.     Variance and Sample Size Calculations in Quality-of-Life-Adjusted Survival Analysis (Q-TWiST)

    The Quality-Adjusted Time Without Symptoms or Toxicity (Q-TWiST) statistic previously introduced by Glasziou, Simes and Gelber (1990, Statistics in Medicine 9 , 1259–1276) combines toxicity, disease-free survival, and overall survival information in assessing the impact of treatments on the lives of patients. This methodology has received positive reviews from clinicians as intuitive and useful, but to date, the variance of this statistic has remained unspecified. We review aspects of the Q-TWiST method for analyzing clinical trial data, extend the method to accommodate multiple treatment arms, and provide closed-form asymptotic variance formulas. We also provide a framework for designing Q-TWiST clinical trials with sample sizes determined using the derived asymptotic variance formulas.     Hyperspectral imaging based detection of PVC during Sellafield repackaging procedures.

    Traditionally, Special Nuclear Material (SNM) at Sellafield has been stored in multi-layered packages, consisting of metallic cans and an over-layer of plasticized Polyvinyl Chloride (PVC) as an intermediate layer when transitioning between areas of different radiological classification. However, it has been found that plasticized PVC can break down in the presence of both radiation and heat, releasing hydrochloric acid which can corrode these metallic containers. Therefore, internal repackaging procedures at Sellafield have focused recently on the removal of these PVC films from containers, where as much degraded and often adhered PVC as possible is manually removed based on visual inspection. This manual operation is time-consuming and it is possible that residual fragments of PVC could remain, leading to corrosion-related issues in future. In this work, Hyperspectral Imaging (HSI) was evaluated as a new tool for detecting PVC on metallic surfaces. Samples of stainless steel type 1.4404 – also known as 316L, the same as is used to construct SNM cans – and PVC were imaged in our experiments, and Support Vector Machine (SVM) classification models were used to generate detection maps. In these maps, pixels were classified into either PVC or 316L based on their spectral responses in the range 954-1700nm of the electromagnetic spectrum. Results suggest that HSI could be used for an effective automated detection and quantification of PVC during repackaging procedures, detection and quantification that could be extended to other similar applications

    Scaling of Huygens-front speedup in weakly random media

    Front propagation described by Huygens' principle is a fundamental mechanism of spatial spreading of a property or an effect, occurring in optics, acoustics, ecology and combustion. If the local front speed varies randomly due to inhomogeneity or motion of the medium (as in turbulent premixed combustion), then the front wrinkles and its overall passage rate (turbulent burning velocity) increases. The calculation of this speedup is subtle because it involves the minimum-time propagation trajectory. Here we show mathematically that for a medium with weak isotropic random fluctuations, under mild conditions on its spatial structure, the speedup scales with the 4/3 power of the fluctuation amplitude. This result, which verifies a previous conjecture while clarifying its scope, is obtained by reducing the propagation problem to the inviscid Burgers equation with white-in-time forcing.     NIMBUS: The Near-Infrared Multi-Band Ultraprecise Spectroimager for SOFIA

    We present a new and innovative near-infrared multi-band ultraprecise spectroimager (NIMBUS) for SOFIA. This design is capable of characterizing a large sample of extrasolar planet atmospheres by measuring elemental and molecular abundances during primary transit and occultation. This wide-field spectroimager would also provide new insights into Trans-Neptunian Objects (TNO), Solar System occultations, brown dwarf atmospheres, carbon chemistry in globular clusters, chemical gradients in nearby galaxies, and galaxy photometric redshifts. NIMBUS would be the premier ultraprecise spectroimager by taking advantage of the SOFIA observatory and state of the art infrared technologies. This optical design splits the beam into eight separate spectral bandpasses, centered around key molecular bands from 1 to 4 microns. Each spectral channel has a wide field of view for simultaneous observations of a reference star that can decorrelate time-variable atmospheric and optical assembly effects, allowing the instrument to achieve ultraprecise calibration for imaging and photometry for a wide variety of astrophysical sources. NIMBUS produces the same data products as a low-resolution integral field spectrograph over a large spectral bandpass, but this design obviates many of the problems that preclude high-precision measurements with traditional slit and integral field spectrographs.     Phase II Trial of IL-12 Plasmid Transfection and PD-1 Blockade in Immunologically Quiescent Melanoma.

    PurposeTumors with low frequencies of checkpoint positive tumor-infiltrating lymphocytes (cpTIL) have a low likelihood of response to PD-1 blockade. We conducted a prospective multicenter phase II trial of intratumoral plasmid IL-12 (tavokinogene telseplasmid; "tavo") electroporation combined with pembrolizumab in patients with advanced melanoma with low frequencies of checkpoint positive cytotoxic lymphocytes (cpCTL).Patients and methodsTavo was administered intratumorally days 1, 5, and 8 every 6 weeks while pembrolizumab (200 mg, i.v.) was administered every 3 weeks. The primary endpoint was objective response rate (ORR) by RECIST, secondary endpoints included duration of response, overall survival and progression-free survival. Toxicity was evaluated by the CTCAE v4. Extensive correlative analysis was done.ResultsThe combination of tavo and pembrolizumab was well tolerated with adverse events similar to those previously reported with pembrolizumab alone. Patients had a 41% ORR (n = 22, RECIST 1.1) with 36% complete responses. Correlative analysis showed that the combination enhanced immune infiltration and sustained the IL-12/IFNγ feed-forward cycle, driving intratumoral cross-presenting dendritic cell subsets with increased TILs, emerging T cell receptor clones and, ultimately, systemic cellular immune responses.ConclusionsThe combination of tavo and pembrolizumab was associated with a higher than expected response rate in this poorly immunogenic population. No new or unexpected toxicities were observed. Correlative analysis showed T cell infiltration with enhanced immunity paralleling the clinical activity in low cpCTL tumors
