    Population genetic analysis of Bartonella bacilliformis isolates from areas of Peru where Carrion\u27s disease is endemic and epidemic

    Carrion's disease is caused by infection with the α-proteobacterium Bartonella bacilliformis. Distribution of the disease is considered coincident with the distribution of its known vector, the sand fly Lutzomyia verrucarum. Recent epidemics of B. bacilliformis infections associated with atypical symptomatology in nonendemic regions have raised questions regarding the historic and present distribution of this bacterium and the scope of disease that infection causes. Phylogenetic relationships and genomic diversity of 18 B. bacilliformis isolates (10 isolates from a region where Carrion's disease is epidemic, Cuzco, Peru, and 8 isolates from a region where Carrion's disease is endemic, Caraz, Peru) were assessed using genomic data generated by infrequent restriction site PCR and gene sequence analysis of the flagellin gltA and ialB genes. A population genetic analysis of the genomic diversity suggests that what was once considered an epidemic region of Peru did not result from the recent introduction of B. bacilliformis

    Powdery mildew resistance genes in vines: an opportunity to achieve a more sustainable viticulture

    Grapevine (Vitis vinifera) is one of the main fruit crops worldwide. In 2020, the total surface area planted with vines was estimated at 7.3 million hectares. Diverse pathogens affect grapevine yield, fruit, and wine quality of which powdery mildew is the most important disease prior to harvest. Its causal agent is the biotrophic fungus Erysiphe necator, which generates a decrease in cluster weight, delays fruit ripening, and reduces photosynthetic and transpiration rates. In addition, powdery mildew induces metabolic reprogramming in its host, affecting primary metabolism. Most commercial grapevine cultivars are highly susceptible to powdery mildew; consequently, large quantities of fungicide are applied during the productive season. However, pesticides are associated with health problems, negative environmental impacts, and high costs for farmers. In paralleled, consumers are demanding more sustainable practices during food production. Therefore, new grapevine cultivars with genetic resistance to powdery mildew are needed for sustainable viticulture, while maintaining yield, fruit, and wine quality. Two main gene families confer resistance to powdery mildew in the Vitaceae, Run (Resistance to Uncinula necator) and Ren (Resistance to Erysiphe necator).     Seismology Of A Massive Pulsating Hydrogen Atmosphere White Dwarf

    We report our observations of the new pulsating hydrogen atmosphere white dwarf SDSS J132350.28+010304.22. We discovered periodic photometric variations in frequency and amplitude that are commensurate with nonradial g-mode pulsations in ZZ Ceti stars. This, along with estimates for the star's temperature and gravity, establishes it as a massive ZZ Ceti star. We used time-series photometric observations with the 4.1 m SOAR Telescope, complemented by contemporary McDonald Observatory 2.1 m data, to discover the photometric variability. The light curve of SDSS J132350.28+010304.22 shows at least nine detectable frequencies. We used these frequencies to make an asteroseismic determination of the total mass and effective temperature of the star: M-star = 0.88 +/- 0.02 M-circle dot and T-eff = 12,100 +/- 140 K.     Community-Based Participatory Research: Lessons Learned from the Centers for Children’s Environmental Health and Disease Prevention Research

    Over the past several decades there has been growing evidence of the increase in incidence rates, morbidity, and mortality for a number of health problems experienced by children. The causation and aggravation of these problems are complex and multifactorial. The burden of these health problems and environmental exposures is borne disproportionately by children from low-income communities and communities of color. Researchers and funding institutions have called for increased attention to the complex issues that affect the health of children living in marginalized communities—and communities more broadly—and have suggested greater community involvement in processes that shape research and intervention approaches, for example, through community-based participatory research (CBPR) partnerships among academic, health services, public health, and community-based organizations. Centers for Children’s Environmental Health and Disease Prevention Research (Children’s Centers) funded by the National Institute of Environmental Health Sciences and U.S. Environmental Protection Agency were required to include a CBPR project. The purpose of this article is to provide a definition and set of CBPR principles, to describe the rationale for and major benefits of using this approach, to draw on the experiences of six of the Children’s Centers in using CBPR, and to provide lessons learned and recommendations for how to successfully establish and maintain CBPR partnerships aimed at enhancing our understanding and addressing the multiple determinants of children’s health

    El presente trabajo es resultado del proyecto de extensión UNNE res. 109/18 C.S., en el cual participaron integrantes de la FaCENA (UNNE), EBCo (MACN- CONICET), docentes y estudiantes de distintos niveles educativos de la provincia de Corrientes, guardaparques del Parque Pcial San Cayetano (PPSC, San Cayetano, Corrientes) y guías del Aluseo de Ciencias Naturales Amado Bonpland y de Casa Iberá (Corrientes, Corrientes). El objetivo principal fue fortalecer valores ambientales relacionados a temas de biodiversidad y conservación, principalmente mamíferos y sus ambientes naturales. Entre ellos, se trató aquellas especies bajo protección como los monumentos naturales de la provincia (aguará guazú, lobito de río, ciervo de los pantanos, venados de las pampas y oso hormiguero), y otras de importancia ecológica, cultural y/o económica como monos carayá, guazunchos, zorros, carpinchos y tatiíes. Esta experiencia fue un punto de partida para la integración didáctica entre los conocimientos científicos generados en la EBCo y la UNNE y los contenidos del Diseño Curricular de Corrientes, siendo la metodología talleres de educación no formal en las aulas y visitas guiadas al PPSC, Museo de Cs. Naturales Bonpland y/o Casa Iberá.     The Ship of Theseus Puzzle

    Does the Ship of Theseus present a genuine puzzle about persistence due to conflicting intuitions based on “continuity of form” and “continuity of matter” pulling in opposite directions? Philosophers are divided. Some claim that it presents a genuine puzzle but disagree over whether there is a solution. Others claim that there is no puzzle at all since the case has an obvious solution. To assess these proposals, we conducted a cross-cultural study involving nearly 3,000 people across twenty-two countries, speaking eighteen different languages. Our results speak against the proposal that there is no puzzle at all and against the proposal that there is a puzzle but one that has no solution. Our results suggest that there are two criteria—“continuity of form” and “continuity of matter”— that constitute our concept of persistence and these two criteria receive different weightings in settling matters concerning persistence

    Sedentary time in older men and women: an international consensus statement and research priorities

    Sedentary time is a modifiable determinant of poor health, and in older adults, reducing sedentary time may be an important first step in adopting and maintaining a more active lifestyle. The primary purpose of this consensus statement is to provide an integrated perspective on current knowledge and expert opinion pertaining to sedentary behaviour in older adults on the topics of measurement, associations with health outcomes, and interventions. A secondary yet equally important purpose is to suggest priorities for future research and knowledge translation based on gaps identified. A five-step Delphi consensus process was used. Experts in the area of sedentary behaviour and older adults (n=15) participated in three surveys, an in-person consensus meeting, and a validation process. The surveys specifically probed measurement, health outcomes, interventions, and research priorities. The meeting was informed by a literature review and conference symposium, and it was used to create statements on each of the areas addressed in this document. Knowledge users (n=3) also participated in the consensus meeting. Statements were then sent to the experts for validation. It was agreed that self-report tools need to be developed for understanding the context in which sedentary time is accumulated. For health outcomes, it was agreed that the focus of sedentary time research in older adults needs to include geriatric-relevant health outcomes, that there is insufficient evidence to quantify the dose-response relationship, that there is a lack of evidence on sedentary time from older adults in assisted facilities, and that evidence on the association between sedentary time and sleep is lacking. For interventions, research is needed to assess the impact that reducing sedentary time, or breaking up prolonged bouts of sedentary time has on geriatric-relevant health outcomes. Research priorities listed for each of these areas should be considered by researchers and funding agencies

    Advancing uracil-excision based cloning towards an ideal technique for cloning PCR fragments

    The largely unused uracil-excision molecular cloning technique has excellent features in most aspects compared to other modern cloning techniques. Its application has, however, been hampered by incompatibility with proof-reading DNA polymerases. We have advanced the technique by identifying PfuCx as a compatible proof-reading DNA polymerase and by developing an improved vector design strategy. The original features of the technique, namely simplicity, speed, high efficiency and low cost are thus combined with high fidelity as well as a transparent, simple and flexible vector design. A comprehensive set of vectors has been constructed covering a wide range of different applications and their functionality has been confirmed
