    Estudio de la citotoxicidad de cerámicas biomórficas de SiC recubiertas con vidrio bioactivo

    La necesidad de desarrollar nuevos implantes basados en materiales bioactivos que sean capaces de soportar grandes cargas mecánicas ha llevado a la producción de sustratos metálicos recubiertos con cerámicas bioactivas. Recientemente se ha propuesto un dispositivo alternativo que consiste en un sustrato de carburo de silicio (SiC) biomórfico recubierto con vidrio bioactivo, mediante la técnica de Depósito por Láser Pulsado (PLD), y que dispone de la resistencia mecánica adecuada, además de gran ligereza y una porosidad intrínseca muy favorable de cara a la implantación. En este trabajo se presenta un estudio interdisciplinar de este nuevo material centrado en la morfología y porosidad de sustratos de SiC provenientes de diferentes maderas, la bioactividad de los recubrimientos producidos por PLD y en la evaluación in vitro con células de osteosarcoma MG-63 con la que se ha determinado la citotoxicidad de estos materiales y se ha estudiado la influencia de los mismos en la adhesión y la proliferación celular.In the past years there was a need to develop new tough bioactive materials capable to resist high loads when implanted in the body, that led to the production of bioactive coatings on metallic substrates. A new approach, which consists of biomorphic silicon carbide (SiC) coated with bioactive glass by Pulsed Laser Deposition (PLD), was recently presented. This new material joins the high mechanical strength, lightness and porosity of biomorphic SiC and the bioactive properties of PLD glass films. In this work, a multiple evaluation of this new material is presented starting from the biomorphic SiC morphology and porosity, following with the bioactivity in simulated body fluid of the coatings, and ending with a deep in vitro study with MG-63 cells.     Extensive studies on biomorphic SiC ceramics properties for medical applications

    Biomorphic silicon carbide ceramics are light, tough and high-strength materials with interesting biomedical applications. The fabrication method of the biomorphic SiC is based in the infiltration of molten-Si in carbon preforms with open porosity. The final product is a biostructure formed by a tangle of SiC fibers. This innovative process allows the fabrication of complex shapes and the tailoring of SiC ceramics with optimised properties and controllable microstructures that will match the biomechanical requirements of the natural host tissue. An interdisciplinary approach of the biomorphic SiC fabricated from beech, sapelly and eucalyptus is presented. Their mechanical properties, microstructure and chemical composition were evaluated. The biocompatible behaviour of these materials has been tested in vitro

    Conservation and management implications of nest-site selection of the sympatric crocodilians Melanosuchus niger and Caiman crocodilus in Central Amazonia, Brazil

    Management of crocodilians is often based on source-sink dynamics, protecting breeding habitat and concentrating hunting in other areas. Nest distributions shed light on habitat use by breeding populations, which might be used as a basis for monitoring and management. The heterogeneous spatial distribution of Melanosuchus niger and Caiman crocodilus in Amazonia has been suggested to reflect past hunting pressure, often underestimating the natural ecological peculiarities of these species. Ground nest surveys combined with satellite imagery allowed us to evaluate whether nest-site use by M. niger and C. crocodilus reflects environmental constraints or is a result of hunting pressure. Our results indicate that there is little evidence that hunting pressure shapes nest-site use of these species in our study areas. M. niger nests mainly on the shores of stable, temporally impounded floodplain lakes isolated from the early stages of the annual rising water of main rivers. This behavior facilitates the identification of suitable nesting sites using moderate-resolution remote-sensing tools and should guide monitoring efforts and the protection of these areas. In contrast, C. crocodilus is a generalist species, able to nest hundreds of meters inside the forest far from permanent water. This makes the occurrence and distribution of nesting-sites unpredictable using Landsat images. Although nests of this species can be found around lakes where nests of M. niger also occur, the protection of these sites might help to preserve only a small portion of C. crocodilus nesting females. Thus, conservation strategies for C. crocodilus should probably be based on different approaches. © 2010 Elsevier Ltd

    Humanized medium (h7H) allows long-term primary follicular thyroid cultures from human normal thyroid, benign neoplasm, and cancer

    Mechanisms of thyroid physiology and cancer are principally studied in follicular cell lines. However, human thyroid cancer lines were found to be heavily contaminated by other sources, and only one supposedly normal-thyroid cell line, immortalized with SV40 antigen, is available. In primary culture, human follicular cultures lose their phenotype after passage. We hypothesized that the loss of the thyroid phenotype could be related to culture conditions in which human cells are grown in medium optimized for rodent culture, including hormones with marked differences in its affinity for the relevant rodent/human receptor.|The objective of the study was to define conditions that allow the proliferation of primary human follicular thyrocytes for many passages without losing phenotype.|Concentrations of hormones, transferrin, iodine, oligoelements, antioxidants, metabolites, and ethanol were adjusted within normal homeostatic human serum ranges. Single cultures were identified by short tandem repeats. Human-rodent interspecies contamination was assessed.|We defined an humanized 7 homeostatic additives medium enabling growth of human thyroid cultures for more than 20 passages maintaining thyrocyte phenotype. Thyrocytes proliferated and were grouped as follicle-like structures; expressed Na+/I- symporter, pendrin, cytokeratins, thyroglobulin, and thyroperoxidase showed iodine-uptake and secreted thyroglobulin and free T3. Using these conditions, we generated a bank of thyroid tumors in culture from normal thyroids, Grave's hyperplasias, benign neoplasms (goiter, adenomas), and carcinomas.|Using appropriate culture conditions is essential for phenotype maintenance in human thyrocytes.     Temporal variation in black-caiman-nest predation in varzea of central Brazilian amazonia

    On the Amazon floodplain, the main predators of black caiman (Melanosuchus niger) eggs are jaguars (Panthera onca), tegu lizards (Tupinambis teguixim), capuchin monkeys (Sapajus macrocephalus) and humans (Homo sapiens). In this study, we investigated the relationship between predator attacks on nests and incubation period, and evaluated the influence of initial predation on subsequent predation in the Mamirauá Sustainable Development Reserve. We also evaluated the influence of presence of females near the nests and manipulation of nests on the occurrence of attacks. We compared results from data obtained with camera traps and vestiges left by predators on estimates of rates of predation by different predators. Egg predation was recorded in 32% of the 658 black caiman nests monitored during two years. Our results suggest that the probability of predation on black caiman eggs is relatively constant throughout the incubation period and that predation on eggs was lower when adults, presumably females, were present. Careful opening of nests and handling of eggs did not increase the number of attacks on black caiman nests. Nest opening by a predator appeared to increase the chances of a subsequent attack because most of the attacks on nests occurred soon after a predator first opened the nest. However, attacks by another species of predator do not appear to be necessary to initiate attacks by any other species of predator. Results based on camera traps and vestiges differed, but use of vestiges was adequate for identifying the principal predators on eggs in black caiman nests and, in many circumstances, the vestiges may be better for estimating predation by humans.     Age and date for early arrival of the Acheulian in Europe (Barranc de la Boella, la Canonja, Spain)

    The first arrivals of hominin populations into Eurasia during the Early Pleistocene are currently considered to have occurred as short and poorly dated biological dispersions. Questions as to the tempo and mode of these early prehistoric settlements have given rise to debates concerning the taxonomic significance of the lithic assemblages, as trace fossils, and the geographical distribution of the technological traditions found in the Lower Palaeolithic record. Here, we report on the Barranc de la Boella site which has yielded a lithic assemblage dating to ,1 million years ago that includes large cutting tools (LCT). We argue that distinct technological traditions coexisted in the Iberian archaeological repertoires of the late Early Pleistocene age in a similar way to the earliest sub-Saharan African artefact assemblages. These differences between stone tool assemblages may be attributed to the different chronologies of hominin dispersal events. The archaeological record of Barranc de la Boella completes the geographical distribution of LCT assemblages across southern Eurasia during the EMPT (Early-Middle Pleistocene Transition, circa 942 to 641 kyr). Up to now, chronology of the earliest European LCT assemblages is based on the abundant Palaeolithic record found in terrace river sequences which have been dated to the end of the EMPT and later.
