    A numerical study of transient three-dimensional heat conduction problem with a moving source is presented. For numerical solution Douglas-Gunn alternating direction implicit method is applied and for the moving heat source flux distribution Gaussian function is used. An influence on numerical solution of input parameters figuring in flux boundary conditions is examined. This include parameters appearing in Gaussian function and heat transfer coefficient from free convection boundaries. Sensitivity of cooling time from 800 to 500°C with respect to input parameters is also tested

    The three-dimensional transient nonlinear thermal analysis of the hard facing process is performed by using the finite element method. The simulations were executed on the open source Salome platform using the open source finite element solver Code_Aster. The Gaussian double ellipsoid was selected in order to enable greater possibilities for the calculation of the moving heat source. The numerical results were compared with available experimental results

    Including Pathogen Risk in Life Cycle Assessment of Wastewater Management. 1. Estimating the Burden of Disease Associated with Pathogens

    The environmental performance of wastewater and sewage sludge management is commonly assessed using life cycle assessment (LCA), whereas pathogen risk is evaluated with quantitative microbial risk assessment (QMRA). This study explored the application of QMRA methodology with intent to include pathogen risk in LCA and facilitate a comparison with other potential impacts on human health considered in LCA. Pathogen risk was estimated for a model wastewater treatment system (WWTS) located in an industrialized country and consisting of primary, secondary, and tertiary wastewater treatment, anaerobic sludge digestion, and land application of sewage sludge. The estimation was based on eight previous QMRA studies as well as parameter values taken from the literature. A total pathogen risk (expressed as burden of disease) on the order of 0.2–9 disability-adjusted life years (DALY) per year of operation was estimated for the model WWTS serving 28 600 persons and for the pathogens and exposure pathways included in this study. The comparison of pathogen risk with other potential impacts on human health considered in LCA is detailed in part 2 of this article series

    The ciliopathy gene cc2d2a controls zebrafish photoreceptor outer segment development through a role in Rab8-dependent vesicle trafficking

    Ciliopathies are a genetically and phenotypically heterogeneous group of human developmental disorders whose root cause is the absence or dysfunction of primary cilia. Joubert syndrome is characterized by a distinctive hindbrain malformation variably associated with retinal dystrophy and cystic kidney disease. Mutations in CC2D2A are found in ∼10% of patients with Joubert syndrome. Here we describe the retinal phenotype of cc2d2a mutant zebrafish consisting of disorganized rod and cone photoreceptor outer segments resulting in abnormal visual function as measured by electroretinogram. Our analysis reveals trafficking defects in mutant photoreceptors affecting transmembrane outer segment proteins (opsins) and striking accumulation of vesicles, suggesting a role for Cc2d2a in vesicle trafficking and fusion. This is further supported by mislocalization of Rab8, a key regulator of opsin carrier vesicle trafficking, in cc2d2a mutant photoreceptors and by enhancement of the cc2d2a retinal and kidney phenotypes with partial knockdown of rab8. We demonstrate that Cc2d2a localizes to the connecting cilium in photoreceptors and to the transition zone in other ciliated cell types and that cilia are present in these cells in cc2d2a mutants, arguing against a primary function for Cc2d2a in ciliogenesis. Our data support a model where Cc2d2a, localized at the photoreceptor connecting cilium/transition zone, facilitates protein transport through a role in Rab8-dependent vesicle trafficking and fusion

    IFT Proteins Accumulate during Cell Division and Localize to the Cleavage Furrow in Chlamydomonas

    Intraflagellar transport (IFT) proteins are well established as conserved mediators of flagellum/cilium assembly and disassembly. However, data has begun to accumulate in support of IFT protein involvement in other processes elsewhere in the cell. Here, we used synchronous cultures of Chlamydomonas to investigate the temporal patterns of accumulation and localization of IFT proteins during the cell cycle. Their mRNAs showed periodic expression that peaked during S and M phase (S/M). Unlike most proteins that are synthesized continuously during G1 phase, IFT27 and IFT46 levels were found to increase only during S/M phase. During cell division, IFT27, IFT46, IFT72, and IFT139 re-localized from the flagella and basal bodies to the cleavage furrow. IFT27 was further shown to be associated with membrane vesicles in this region. This localization pattern suggests a role for IFT in cell division

    A recent survey commissioned by English Heritage highlights the rich cultural history of the traditional English seaside resort (Brodie and Winter 2007). Emerging in the eighteenth century, these towns grew in significance before the advent of cheaper continental holidays in the 1960s signalled their demise. Nevertheless they retain an affectionate place within English social memory, and are in their own right distinctive maritime communities. Using an archaeological case study and a broadly phenomenological approach this contribution analyses the experience of the resort holiday through reference to place, space and materiality. Further, it seeks to situate the English seaside resort, as a functionally distinctive post-medieval urban and maritime phenomenon, within a global context of the archaeology of tourism
