    An auxin-inducible, GAL4-compatible, gene expression system for Drosophila

    The ability to control transgene expression, both spatially and temporally, is essential for studying model organisms. In Drosophila, spatial control is primarily provided by the GAL4/UAS system, whilst temporal control relies on a temperature-sensitive GAL80 (which inhibits GAL4) and drug-inducible systems. However, these are not ideal. Shifting temperature can impact on many physiological and behavioural traits, and the current drug-inducible systems are either leaky, toxic, incompatible with existing GAL4-driver lines, or do not generate effective levels of expression. Here, we describe the auxin-inducible gene expression system (AGES). AGES relies on the auxin-dependent degradation of a ubiquitously expressed GAL80, and therefore, is compatible with existing GAL4-driver lines. Water-soluble auxin is added to fly food at a low, non-lethal, concentration, which induces expression comparable to uninhibited GAL4 expression. The system works in both larvae and adults, providing a stringent, non-lethal, cost-effective, and convenient method for temporally controlling GAL4 activity in Drosophila

    The Conservatives and the Union: The 'New English Toryism' and the Origins of Anglo-Britishness

    The Union has for over a century been one of the cornerstones of Conservative politics, but with the changes to the old union state in the last 20 years, its value has been increasingly questioned. While the Conservative Party remains committed to maintaining the Union, a new English Toryism is emerging, which has a strong continuity with an older English Toryism which was partially buried by the ascendancy of Unionism. English Tories have always considered the Union to be desirable, but it comes second in their thinking to the need to protect the sovereignty of the British state, the core of which is England and its traditional institutions. What is new about the contemporary Conservative Party is that there is within it the revival of an English Toryism which is happy to discard the older clothes of Empire and Union once so important to Conservative identity and which is unabashedly English in its focus. For an increasing number of contemporary Conservatives, there no longer seems to be any passion about defending the Union or even of continuing to think about the United Kingdom in Unionist terms

    Soccer activity profile of altitude versus sea-level natives during acclimatisation to 3600 m (ISA3600)

    Objectives We investigated the effect of high altitude on the match activity profile of elite youth high altitude and sea level residents.Methods Twenty Sea Level (Australian) and 19 Altitude-resident (Bolivian) soccer players played five games, two near sea level (430 m) and three in La Paz (3600 m). Match activity profile was quantified via global positioning system with the peak 5 min period for distance ((D5peak)) and high velocity running (>4.17 m/s, HIVR5peak); as well as the 5 min period immediately subsequent to the peak for both distance (D5sub) and high-velocity running (HIVR5sub) identified using a rolling 5 min epoch. The games at 3600 m were compared with the average of the two near sea-level games.Results The total distance per minute was reduced by a small magnitude in the first match at altitude in both teams, without any change in low-velocity running. There were variable changes in HiVR, D5peak and HiVR5peak from match to match for each team. There were within-team reductions in D5peak in each game at altitude compared with those at near sea level, and this reduction was greater by a small magnitude in Australians than Bolivians in game 4. The effect of altitude on HiVR5peak was moderately lower in Australians compared with Bolivians in game 3. There was no clear difference in the effect of altitude on maximal accelerations between teams.Conclusions High altitude reduces the distance covered by elite youth soccer players during matches. Neither 13 days of acclimatisation nor lifelong residence at high altitude protects against detrimental effects of altitude on match activity profile

    Establishing a composite neonatal adverse outcome indicator using English hospital administrative data.

    OBJECTIVE: We adapted a composite neonatal adverse outcome indicator (NAOI), originally derived in Australia, and assessed its feasibility and validity as an outcome indicator in English administrative hospital data. DESIGN: We used Hospital Episode Statistics (HES) data containing information infants born in the English National Health Service (NHS) between 1 April 2014 and 31 March 2015. The Australian NAOI was mapped to diagnoses and procedure codes used within HES and modified to reflect data quality and neonatal health concerns in England. To investigate the concurrent validity of the English NAOI (E-NAOI), rates of NAOI components were compared with population-based studies. To investigate the predictive validity of the E-NAOI, rates of readmission and death in the first year of life were calculated for infants with and without E-NAOI components. RESULTS: The analysis included 484 007 (81%) of the 600 963 eligible babies born during the timeframe. 114/148 NHS trusts passed data quality checks and were included in the analysis. The modified E-NAOI included 23 components (16 diagnoses and 7 procedures). Among liveborn infants, 5.4% had at least one E-NAOI component recorded before discharge. Among newborns discharged alive, the E-NAOI was associated with a significantly higher risk of death (0.81% vs 0.05%; p<0.001) and overnight hospital readmission (15.7% vs 7.1%; p<0.001) in the first year of life. CONCLUSIONS: A composite NAOI can be derived from English hospital administrative data. This E-NAOI demonstrates good concurrent and predictive validity in the first year of life. It is a cost-effective way to monitor neonatal outcomes

    Technical Performance Reduces during the Extra-Time Period of Professional Soccer Match-Play

    Despite the importance of extra-time in determining progression in specific soccer tournament matches, few studies have profiled the demands of 120-minutes of soccer match-play. With a specific focus on the extra-time period, and using a within-match approach, we examined the influence of prolonged durations of professional soccer match-play on markers of technical (i.e., skilled) performance. In 18 matches involving professional European teams played between 2010 and 2014, this retrospective study quantified the technical actions observed during eight 15-minute epochs (E1: 00:00–14:59 min, E2: 15:00-29:59 min, E3: 30:00-44:59 min, E4: 45:00-59:59 min, E5: 60:00-74:59 min, E6: 75:00-89:59 min, E7: 90:00-104:59 min, E8: 105:00-119:59 min). Analysis of players who completed the demands of the full 120 min of match-play revealed that the cumulative number of successful passes observed during E8 (61±23) was lower than E1-4 (E1: 88±23, P=0.001; E2: 77±21, P=0.005; E3: 79±18, P=0.001; E4: 80±21, P=0.001) and E7 (73±20, P=0.002). Similarly, the total number of passes made in E8 (71±25) was reduced when compared to E1 (102±22, P=0.001), E3 (91±19, P=0.002), E4 (93±22, P≤0.0005) and E7 (84±20, P=0.001). The cumulative number of successful dribbles reduced in E8 (9±4) when compared to E1 (14±4, P=0.001) and E3 (12±4, P≤0.0005) and the total time the ball was in play was less in E8 (504±61 s) compared to E1 (598±70 s, P≤0.0005). These results demonstrate that match-specific factors reduced particular indices of technical performance in the second half of extra-time. Interventions that seek to maintain skilled performance throughout extra-time warrant further investigation

    Match running performance and physical capacity profiles of U8 and U10 soccer players

    Aim This study aimed to characterize match running performance of very young soccer players and evaluate the relationship between these data and physical capacities and technical skills. Methods Distances covered at different speed thresholds were measured during 31 official matches using GPS technology in U10 (n = 12; age 10.1 ± 0.1 years) and U8 (n = 15; age 7.9 ± 0.1 years) national soccer players. Counter movement jump performance (CMJ), 20 m shuttle running (20 m-SR), linear sprint performance (10, 20, 30 m), shuttle (SHDT) and slalom dribble tests (SLDT) were performed to determine the players physical capacities and technical skills. Results Physical capacities and technical skills were higher in U10 versus U8 players [P 0.05, ES: 0.74). The U10 players covered more total (TD) and high-intensity running distance (HIRD) than their younger counterparts did (P 0.05, ES: 0.99). TD and HIRD covered across the three 15 min periods of match play did not decline (P > 0.05, ES: 0.02–0.55). Very large magnitude correlations were observed between the U8 and U10 players performances during the 20 m-SR versus TD (r = 0.79; P < 0.01) and HIRD (r = 0.82; P < 0.01) covered during match play. Conclusions Data demonstrate differences in match running performance and physical capacity between U8 and U10 players, and large magnitude relationships between match play measures and physical test performances. These findings could be useful to sports science staff working within the academies

    Community Worker Perspectives on the Use of New Media to Reconfigure Socio-spatial Relations in Belfast

    Cyber enthusiasts as far back as Rheingold have suggested that cyberspatial technologies such as the Internet have the potential to transform space–time relations and create new social spaces, thus ameliorating social conflict in contested areas. However, a more sceptical view of cyberspatial communication is provided by Hampton, who argues that on-line interactions cannot be artifically separated from their off-line contexts. This article will analyse whether these technologies are changing the nature of territorial disputes and patterns of social interaction between Protestant and Catholic interface communities in Belfast, Northern Ireland. Interviews were conducted with nine community workers to investigate this issue. Focusing on the possibility of using social media to facilitate intergroup contact, the paper argues that on-line interactions alone do not appear to have the potential to build mutual understanding and trust between rival interface communities. Indeed, community workers fear that may young people use these sites to exacerbate intercommunity tensions

    Non-integumentary melanosomes can bias reconstructions of the colours of fossil vertebrates

    The soft tissues of many fossil vertebrates preserve evidence of melanosomes-micron-scale organelles that inform on integumentary coloration and communication strategies. In extant vertebrates, however, melanosomes also occur in internal tissues. Hence, fossil melanosomes may not derive solely from the integument and its appendages. Here, by analyzing extant and fossil frogs, we show that non-integumentary melanosomes have high fossilization potential, vastly outnumber those from the skin, and potentially dominate the melanosome films preserved in some fossil vertebrates. Our decay experiments show that non-integumentary melanosomes usually remain in situ provided that carcasses are undisturbed. Micron-scale study of fossils, however, demonstrates that non-integumentary melanosomes can redistribute through parts of the body if carcasses are disturbed by currents. Collectively, these data indicate that fossil melanosomes do not always relate to integumentary coloration. Integumentary and non-integumentary melanosomes can be discriminated using melanosome geometry and distribution. This is essential to accurate reconstructions of the integumentary colours of fossil vertebrates
