Manufacturing and use of human placenta-derived mesenchymal stromal cells for phase I clinical trials: Establishment and evaluation of a protocol
Background/Aim. Mesenchymal stromal cells (MSCs) have been utilised in many
clinical trials as an experimental treatment in numerous clinical settings.
Bone marrow remains the traditional source tissue for MSCs but is relatively
hard to access in large volumes. Alternatively, MSCs may be derived from
other tissues including the placenta and adipose tissue. In an initial study
no obvious differences in parameters such as cell surface phenotype,
chemokine receptor display, mesodermal differentiation capacity or
immunosuppressive ability, were detected when we compared human marrow
derived- MSCs to human placenta-derived MSCs. The aim of this study was to
establish and evaluate a protocol and related processes for preparation
placenta-derived MSCs for early phase clinical trials. Methods. A full-term
placenta was taken after delivery of the baby as a source of MSCs. Isolation,
seeding, incubation, cryopreservation of human placentaderived MSCs and used
production release criteria were in accordance with the complex regulatory
requirements applicable to Code of Good Manufacturing Practice manufacturing
of ex vivo expanded cells. Results. We established and evaluated instructions
for MSCs preparation protocol and gave an overview of the three clinical
areas application. In the first trial, MSCs were co-transplanted iv to
patient receiving an allogeneic cord blood transplant as therapy for
treatmentrefractory acute myeloid leukemia. In the second trial, MSCs were
administered iv in the treatment of idiopathic pulmonary fibrosis and without
serious adverse effects. In the third trial, MSCs were injected directly into
the site of tendon damage using ultrasound guidance in the treatment of
chronic refractory tendinopathy. Conclusion. Clinical trials using both
allogeneic and autologous cells demonstrated MSCs to be safe. A described
protocol for human placenta-derived MSCs is appropriate for use in a clinical
setting, relatively inexpensive and can be relatively easily adjusted to a
different set of regulatory requirements, as applicable to early phase
clinical trials
Additions to the marine decapod (Crustacea: Decapoda) fauna of South Africa
This report adds five previously unreported species to the decapod crustacean fauna of South Africa, as well as removing one species previously listed in error. It also documents locality (and/or reference specimen) data for 12 other species, most of which had been depicted in regional field guides, but without reporting when and where they had been collected. Almost all the species added were already known from adjacent African countries and their ranges are here extended into South Africa. Although some of these records are based on photographs, rather than collected specimens, it is argued that such records should be accepted as adequate evidence for inclusion of at least visually-distinctive crustacean species into the regional fauna
Acquiring, archiving, analyzing and exchanging seismic data in real time at the Seismological Research Center of the OGS in Italy
The Centro di Ricerche Sismologiche (CRS, Seismological Research Center) of the Istituto Nazionale di
Oceanografia e di Geofisica Sperimentale (OGS, Italian National Institute for Oceanography and Experimental
Geophysics) in Udine (Italy) after the strong earthquake (magnitude M=6.4) occurred in 1976 in the Italian
Friuli-Venezia Giulia region, started to operate the North-east Italy (NI) seismic network: it currently consists of
11 very sensitive broad band and 23 more simple short period seismic stations, all telemetered to and acquired in
real time at the OGS-CRS data center in Udine.
Real time data exchange agreements in place with other Italian, Slovenian, Austrian and Swiss seismological
institutes lead to a total number of 89 seismic stations acquired in real time, which makes the OGS the reference
institute for seismic monitoring of Northeastern Italy.
Since 2002 OGS-CRS is using the Antelope software suite as the main tool for collecting, analyzing, archiving
and exchanging seismic data in the framework of the EU Interreg IIIA project “Trans-national seismological
networks in the South-Eastern Alps”. SeisComP is also used as a real time data exchange server tool. At OGS-CRS
we then adapted existing programs and created new ones like: a customized web-accessible server to manually
relocate earthquakes, a script for automatic moment tensor determination, scripts for web publishing of earthquake
parametric data, waveforms, state of health parameters and shaking maps, noise characterization by means of
automatic spectra analysis, plus scripts for email/SMS/fax alerting. A new OGS-CRS real time web site has also
been recently designed and made operative in the framework of the DPC-INGV S3 Project

Best Practices for Publishing, Retrieving, and Using Spatial Data on the Web
Data owners are creating an ever richer set of information resources online, and these are being used for more and more applications. With the rapid growth of connected embedded devices, GPS-enabled mobile devices, and various organizations that publish their location-based data (i.e., weather and traffic services), maps and geographical and spatial information (i.e., GIS and open maps), spatial data on the Web is becoming ubiquitous and voluminous. However, the heterogeneity of the available spatial data, as well as some challenges related to spatial data in particular make it difficult for data users, web applications and services to discover, interpret and use the information in large and distributed web systems. This paper summarizes some of the efforts that have been undertaken in the joint W3C/OGC Working Group on Spatial Data on the Web, in particular the effort to describe the best practices for publishing spatial data on the Web. This paper presents the set of principles that guide the selection of these best practices, describes best practices that are employed to enable publishing, discovery and retrieving (querying) this type of data on the Web, and identifies some areas where a best practice has not yet emerged
Incident vertebral fractures and risk factors in the first three years following glucocorticoid initiation among pediatric patients with rheumatic disorders
Vertebral fractures are an important yet underrecognized manifestation of osteoporosis in children with chronic, glucocorticoid-treated illnesses. Our goal was to determine the incidence and clinical predictors of vertebral fractures in the 3 years following glucocorticoid initiation among pediatric patients with rheumatic disorders. Incident vertebral fractures were evaluated according to the Genant semiquantitative method on lateral radiographs at baseline and then annually in the 3 years following glucocorticoid initiation. Extended Cox models were used to assess the association between vertebral fractures and clinical risk predictors. A total of 134 children with rheumatic disorders were enrolled in the study (mean ± standard deviation (SD) age 9.9 ± 4.4 years; 65% girls). The unadjusted vertebral fracture incidence rate was 4.4 per 100 person-years, with a 3-year incidence proportion of 12.4%. The highest annual incidence occurred in the first year (6.0%; 95% confidence interval (CI) 2.9% to 11.7%). Almost one-half of the patients with fractures were asymptomatic. Every 0.5 mg/kg increase in average daily glucocorticoid (prednisone equivalents) dose was associated with a twofold increased fracture risk (hazard ratio (HR) 2.0; 95% CI 1.1 to 3.5). Other predictors of increased vertebral fracture risk included: (1) increases in disease severity scores between baseline and 12 months; (2) increases in body mass index Z-scores in the first 6 months of each 12-month period preceding the annual fracture assessment; and (3) decreases in lumbar spine bone mineral density Z-scores in the first 6 months of glucocorticoid therapy. As such, we observed that a clinically significant number of children with rheumatic disorders developed incident vertebral fractures in the 3 years following glucocorticoid initiation. Almost one-half of the children were asymptomatic and thereby would have been undiagnosed in the absence of radiographic monitoring. In addition, discrete clinical predictors of incident vertebral fractures were evident early in the course of glucocorticoid therapy
High incidence of vertebral fractures in children with acute lymphoblastic leukemia 12 months after the initiation of therapy
Purpose: Vertebral fractures due to osteoporosis are a potential complication of childhood acute lymphoblastic leukemia (ALL). To date, the incidence of vertebral fractures during ALL treatment has not been reported. Patient and Methods: We prospectively evaluated 155 children with ALL during the first 12 months of leukemia therapy. Lateral thoracolumbar spine radiographs were obtained at baseline and 12 months. Vertebral bodies were assessed for incident vertebral fractures using the Genant semiquantitative method, and relevant clinical indices such as spine bone mineral density (BMD), back pain, and the presence of vertebral fractures at baseline were analyzed for association with incident vertebral fractures. Results: Of the 155 children, 25 (16%; 95% CI, 11% to 23%) had a total of 61 incident vertebral fractures, of which 32 (52%) were moderate or severe. Thirteen (52%) of the 25 children with incident vertebral fractures also had fractures at baseline. Vertebral fractures at baseline increased the odds of an incident fracture at 12 months by an odds ratio of 7.3 (95% CI, 2.3 to 23.1; P = .001). In addition, for every one standard deviation reduction in spine BMD Z-score at baseline, there was 1.8-fold increased odds of incident vertebral fracture at 12 months (95% CI, 1.2 to 2.7; P = .006). Conclusion: Children with ALL have a high incidence of vertebral fractures after 12 months of chemotherapy, and the presence of vertebral fractures and reductions in spine BMD Z-scores at baseline are highly associated clinical features. © 2012 by American Society of Clinical Oncology
Incident vertebral fractures among children with rheumatic disorders 12 months after glucocorticoid initiation: A national observational study
Objective. To determine the frequency of incident vertebral fractures (IVF) 12 months after glucocorticoid (GC) initiation in children with rheumatic diseases and to identify children at higher risk. Methods. Children with rheumatic diseases initiating GC were enrolled in a prospective observational study. Annual spine radiographs were evaluated using the Genant semiquantitative method. Spine areal bone mineral density (aBMD) was measured every 6 months. Clinical features, including cumulative GC dose, back pain, disease and physical activity, calcium and vitamin D intake, and spine aBMD Z scores, were analyzed for association with IVF. Results. Seven (6%) of 118 children (95% confidence interval 2.9-11.7%) had IVF. Their diagnoses were: juvenile dermatomyositis (n = 2), systemic lupus erythematosus (n = 3), systemic vasculitis (n = 1), and mixed connective tissue disease (n = 1). One child was omitted from the analyses after 4 months because of osteoporosis treatment for symptomatic IVF. Children with IVF received on average 50% more GC than those without (P = 0.030), had a greater increase in body mass index (BMI) at 6 months (P = 0.010), and had greater decrements in spine aBMD Z scores in the first 6 months (P = 0.048). Four (67%) of 6 children with IVF and data to 12 months had spine aBMD Z scores less than-2.0 at 12 months compared to 16% of children without IVF (P = 0.011). Conclusion. The incidence of VF 12 months following GC initiation was 6%; most children were asymptomatic. Children with IVF received more GC, had greater increases in BMI, and had greater declines in spine aBMD Z scores in the first 6 months. © 2012, American College of Rheumatology
Effects of antiplatelet therapy on stroke risk by brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases: subgroup analyses of the RESTART randomised, open-label trial
Background
Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage. We did subgroup analyses of the RESTART trial to explore whether these brain imaging features modify the effects of antiplatelet therapy
Agricultural Nematology in East and Southern Africa : Problems, Management Strategies and Stakeholder Linkages

Africa will have to increase food production more than 50% in the coming 50 years to meet the nutritional requirements of its growing population. Nowhere is the need to increase agricultural productivity more pertinent than in much of sub-Saharan Africa where it is currently static or declining. Optimal pest management will be essential, because intensification of any system creates heightened selection pressures for pests. Plant-parasitic nematodes and their damage potential are intertwined with intensified systems and can be an indicator of unsustainable practices. As soil pests, nematodes are commonly overlooked or misdiagnosed, particularly where appropriate expertise and knowledge transfer systems are meager or inadequately funded. Nematode damage to roots results in less efficient root systems that are less able to access nutrients and water, which can produce symptoms typical of water or nutrient deficiency, leading to misdiagnosis of the underlying cause. Damage in subsistence agriculture is exacerbated by growing crops on degraded soils and in areas of low water retention where strong root growth is vital. This review focuses on the current knowledge of economically important nematode pests affecting key crops, nematode control methods, and the research and development needs for sustainable management, stakeholder involvement and capacity building in the context of crop security in East and Southern Africa, especially Kenya, Tanzania, Uganda and Zimbabwe.Peer reviewe
