    Germination niche of a neotropical dry forest species: seed osmotic stress and recovery

    Abstract: Erythrina velutina Willd is a native Fabaceae with wide occurrence, mainly in the Caatinga Domain with multiple uses. This study aimed to compare the vulnerability/tolerance of seeds of four accessions of E. velutina to osmotic stresses. Four replications were conducted with 25 seeds, germinated in osmotic solutions of NaCl or PEG up to -1.73 MPa. Seeds that did not germinate after 14 days were recovered in distilled water. Obtained data were fitted in Boltzmann curves and t50 and synchrony were evaluated. A probit analysis was performed, and hydrotime and halotime models were constructed. In distilled water, the germination curve was similar for all accessions. When the concentration of salt or PEG in the substrate increased, the accessions differentiated into two groups. Two accessions - Jutaí 2012 and Jutaí 2015 - maintained high germination up to -0.86 MPa NaCl, while Caboclo 2008 and Caiçara 2011 accessions presented a lower germination speed. The hydrotime and halotime analyses separated the germination response of accessions according to their physiological quality and tolerance to osmotic and saline conditions. Seeds viability was maintained at both stresses, since germination was reestablished at distilled water, thus constituting a crucial response for this species’ survival and conservation

    Detection of bacterial samples on the aquatic ecosystems adjacent to Saquarema Lagoon – Rio de Janeiro

    Introduction: Saquarema Lagoon (RJ) has a high ecological and economic value owing to its multiple uses. The population’s constant growth increases the amount of sewage containing bacteria and antimicrobial drugs that are discharged to the environment. Objectives: to detect Gram negative bacilli able to colonize or infect humans and animals and determine their antimicrobial resistance profiles. Methodology:samples were collected in the city centre in April 2010 and at Jaconé (Lagoon’s most preserved site) in February 2011. The total and thermo tolerant coliforms were determined and the isolation of samples was made using agar media containing  32cg/ mL of cephalotin. All samples were tested for antimicrobial susceptibility (AST) and on 16 samples, plasmid DNA was extracted. Results: different Gram negative bacteria were detected, such as: Enterobacter spp, Citrobacter freundii, Klebsiella pneumoniae and Pseudomonas aeruginosa. The coliform results showed that the water quality is proper for Human recreation. AST results demonstrated the existence of bacteria resistant to antimicrobial drugs frequently used in the community. It was possible to detected high molecular weight plasmids and nine samples (56,25%) showed at least one plasmid DNA electrophoresis band. Conclusions: there were not detected resistant samples to antimicrobial drugs normally used in hospital settings, which may possibly refute the idea of a contamination by nosocomial and/or veterinary sewage discharge

    Detección de enterobacterias multirresistentes aisladas en aguas de los ríos que desembocan en la bahía de Guanabara y en muestras de hospitales de Río de Janeiro, Brasil

    Introduction: The use of antibiotics in humans, animal husbandry and veterinary activities induces selective pressure leading to the colonization and infection by resistant strains.Objective: We evaluated water samples collected from rivers of the Guanabara Bay, which have suffered minor and major environmental degradation, and clinical samples of hospital origin to detect evidence of the presence of resistance genes to aminoglycosides, beta-lactam antibiotics and fluoroquinolones in strains of Klebsiella pneumoniae subsp. pneumoniae, K. pneumoniae subsp. ozaenae and Escherichia coli.Materials and methods: For isolation of the water strains we employed culture media containing 32 μg/ml cephalotin and 8 μg/ml gentamicin. The strains from clinical materials were selected using culture media containing 8 μg/ml gentamicin. The strains were identified and subjected to antimicrobial susceptibility testing (AST), plasmid DNA extraction and polymerase chain reaction (PCR) to detect genes encoding enzymes modifying aminoglycosides (EMA), extended-spectrum beta-lactamases (ESBL) and plasmid mechanisms of quinolone resistance (PMQR).Results: The AST of the isolates recovered from water samples showed multidrugresistance profiles similar to those found in isolates recovered from clinical materials. All isolates from water samples and 90% of the isolates from clinical samples showed at least one plasmid band. In the PCR assays, 7.4% of the isolates recovered from water samples and 20% of those from clinical materials showed amplification products for the three antimicrobial classes.Conclusion: We believe that the detection of microorganisms presenting genetic elements in environments such as water is necessary for the prevention and control of their dissemination with potential to infect humans and other animals in eventual contact with these environments.Introducción. El uso de antibióticos en seres humanos, en la industria pecuaria y en las actividades veterinarias induce una presión selectiva que resulta en la colonización e infección con cepas resistentes.Objetivo. Determinar la presencia de genes de resistencia a aminoglucósidos, betalactámicos y fluoroquinolonas en cepas de Klebsiella pneumoniae subsp. pneumoniae, K. pneumoniae subsp. ozaenae y Escherichia coli, obtenidas de muestras de agua de los ríos que desembocan en la bahía de Guanabara y de muestras clínicas de hospitales de Río de Janeiro. Materiales y métodos. En la selección de las cepas resistentes obtenidas de las muestras de agua de los ríos, se emplearon medios de cultivo que contenían 32 μg/ml de cefalotina y 8 μg/ml de gentamicina. En el caso de las muestras de especímenes clínicos, se usaron medios de cultivo que contenían 8 μg/ml de gentamicina. Las cepas se identificaron y se sometieron a pruebas de sensibilidad antimicrobiana, extracción de ADN plasmídico y pruebas de reacción en cadena de la polimerasa (PCR) para detectar los genes que codifican aquellas enzimas que modifican los aminoglucósidos, las betalactamasas de espectro extendido (BLEE) y los mecanismos de resistencia a las quinolonas mediados por plásmidos.Resultados. Se encontraron perfiles de resistencia a los antimicrobianos similares en los dos grupos. En todas las bacterias obtenidas de las muestras de agua y en 90 % de las muestras clínicas, se evidenciaron bandas de plásmidos asociados con la transferencia de genes de resistencia. En las pruebas de PCR, se obtuvieron productos de amplificación de los genes de resistencia para las tres clases de antimicrobianos analizados, en el 7,4 % de las bacterias recuperadas de las muestras de agua y en el 20 % de aquellas recuperadas de las muestras clínicas.Conclusión. La detección de microorganismos con elementos genéticos que confieren resistencia a los antibióticos en ambientes como el agua, es una estrategia necesaria para prevenir y controlar la diseminación de estos agentes patógenos con potencial para infectar a humanos y a otros animales en dichos ambientes

    Psychosocial impact of undergoing prostate cancer screening for men with BRCA1 or BRCA2 mutations.

    OBJECTIVES: To report the baseline results of a longitudinal psychosocial study that forms part of the IMPACT study, a multi-national investigation of targeted prostate cancer (PCa) screening among men with a known pathogenic germline mutation in the BRCA1 or BRCA2 genes. PARTICPANTS AND METHODS: Men enrolled in the IMPACT study were invited to complete a questionnaire at collaborating sites prior to each annual screening visit. The questionnaire included sociodemographic characteristics and the following measures: the Hospital Anxiety and Depression Scale (HADS), Impact of Event Scale (IES), 36-item short-form health survey (SF-36), Memorial Anxiety Scale for Prostate Cancer, Cancer Worry Scale-Revised, risk perception and knowledge. The results of the baseline questionnaire are presented. RESULTS: A total of 432 men completed questionnaires: 98 and 160 had mutations in BRCA1 and BRCA2 genes, respectively, and 174 were controls (familial mutation negative). Participants' perception of PCa risk was influenced by genetic status. Knowledge levels were high and unrelated to genetic status. Mean scores for the HADS and SF-36 were within reported general population norms and mean IES scores were within normal range. IES mean intrusion and avoidance scores were significantly higher in BRCA1/BRCA2 carriers than in controls and were higher in men with increased PCa risk perception. At the multivariate level, risk perception contributed more significantly to variance in IES scores than genetic status. CONCLUSION: This is the first study to report the psychosocial profile of men with BRCA1/BRCA2 mutations undergoing PCa screening. No clinically concerning levels of general or cancer-specific distress or poor quality of life were detected in the cohort as a whole. A small subset of participants reported higher levels of distress, suggesting the need for healthcare professionals offering PCa screening to identify these risk factors and offer additional information and support to men seeking PCa screening

    Cardiomiopatia de Takotsubo: uma breve revisão sistemática: Takotsubo Cardiomiopathy: a brief systematic review

    A cardiomiopatia de Takotsubo é uma nova cardiomiopatia que foi noticiada pela primeira vez em 2001. A doença é definida por disfunção reversível do ventrículo esquerdo e manifesta-se normalmente como uma síndrome coronária aguda. Este estudo teve como objetivo discutir as principais características da cardiomiopatia takotsubo. Para isso, foi desenvolvida uma revisão sistemática de literatura, recorrendo-se às bases de dados Scielo, Medline e Lilacs, selecionando-se estudos publicados nos últimos 5 anos. A partir da análise e interpretação dos dados das fontes foi possível concluir que a cardiomiopatia de takotsubo é desencadeada pelo estresse físico e é vista como uma complicação para outras doenças não cardíacas, se apresentando geralmente em mulheres pós-menopausa, acima dos 70 anos de idade. Manifesta-se com início súbito de dor torácica e dispneia, após um evento emocional estressante que precede o início dos sintomas. Com principais consequências, tem-se choque cardiogênico, obstrução da via de saída do ventrículo esquerdo, trombo da parede ventricular esquerda, arritmias ventriculares, ruptura da parede ventricular e paragem cardíaca, registrando-se alguns casos de morte súbita

    Impact of COVID-19 on cardiovascular testing in the United States versus the rest of the world

    Objectives: This study sought to quantify and compare the decline in volumes of cardiovascular procedures between the United States and non-US institutions during the early phase of the coronavirus disease-2019 (COVID-19) pandemic. Background: The COVID-19 pandemic has disrupted the care of many non-COVID-19 illnesses. Reductions in diagnostic cardiovascular testing around the world have led to concerns over the implications of reduced testing for cardiovascular disease (CVD) morbidity and mortality. Methods: Data were submitted to the INCAPS-COVID (International Atomic Energy Agency Non-Invasive Cardiology Protocols Study of COVID-19), a multinational registry comprising 909 institutions in 108 countries (including 155 facilities in 40 U.S. states), assessing the impact of the COVID-19 pandemic on volumes of diagnostic cardiovascular procedures. Data were obtained for April 2020 and compared with volumes of baseline procedures from March 2019. We compared laboratory characteristics, practices, and procedure volumes between U.S. and non-U.S. facilities and between U.S. geographic regions and identified factors associated with volume reduction in the United States. Results: Reductions in the volumes of procedures in the United States were similar to those in non-U.S. facilities (68% vs. 63%, respectively; p = 0.237), although U.S. facilities reported greater reductions in invasive coronary angiography (69% vs. 53%, respectively; p < 0.001). Significantly more U.S. facilities reported increased use of telehealth and patient screening measures than non-U.S. facilities, such as temperature checks, symptom screenings, and COVID-19 testing. Reductions in volumes of procedures differed between U.S. regions, with larger declines observed in the Northeast (76%) and Midwest (74%) than in the South (62%) and West (44%). Prevalence of COVID-19, staff redeployments, outpatient centers, and urban centers were associated with greater reductions in volume in U.S. facilities in a multivariable analysis. Conclusions: We observed marked reductions in U.S. cardiovascular testing in the early phase of the pandemic and significant variability between U.S. regions. The association between reductions of volumes and COVID-19 prevalence in the United States highlighted the need for proactive efforts to maintain access to cardiovascular testing in areas most affected by outbreaks of COVID-19 infection

    Investigation of tumor-tumor interactions in a double human cervical carcinoma xenograft model in nude mice.

    Tumor-tumor distant interactions within one organism are of major clinical relevance determining clinical outcome. To investigate this poorly understood phenomenon, a double human cervical xenograft model in nude mice was developed. A first tumor was induced subcutaneously by injection of human papillomavirus positive cervical carcinoma cells into the mouse lower right flank and 3 weeks later, animals were challenged with the same tumor cell line injected subcutaneously into the upper left flank. These tumors had no direct physical contact and we found no systemic changes induced by the primary tumor affecting the growth of a secondary tumor. However, ablation of the primary tumor by local treatment with cidofovir, a nucleotide analogue with known antiviral and antiproliferative properties, resulted not only in a local antitumor effect but also in a temporary far-reaching effect leading to retarded growth of the challenged tumor. Cidofovir far-reaching effects were linked to a reduced tumor-driven inflammation, to increased anti-tumor immune responses, and could not be enhanced by co-administration with immune stimulating adjuvants. Our findings point to the potential use of cidofovir in novel therapeutic strategies aiming to kill tumor cells as well as to influence the immune system to fight cancer.status: Published onlin

Considerando o caráter histórico (alguns datam do século XVIII), probatório, informativo e a função científica e cultural desses objetos, serão fixados também metadados específicos voltados ao apoio tanto de sua preservação a longo prazo quanto dos próprios metadados descritivos atribuídos a eles. Cabe ressaltar que, embora sejam tratados distintos objetos (peças museais, itens bibliográficos, documentos arquivísticos e acervos biológicos). A proposta desta comunicação será apresentar a metodologia aplicada para a definição dos metadados integrativos baseados no uso do padrão Dublin Core (DC) e para a definição dos metadados de preservação a longo prazo, fundamentado no Preservation Metadata: Implementation Strategies (PREMIS), de acordo com as premissas de autenticidade, confiabilidade e acessibilidade.From the creation of a multidisciplinary Working Group for the elaboration of normative institutional documents “Digital Preservation Plan” and “Digitalization Manual”, aimed at the implementation of actions related to digital preservation in the scientific and cultural collections of the Fundação Oswaldo Cruz (Fiocruz), it was observed the need to establish minimum standards of descriptive metadata, to integrate and streamline access to the different digital objects that represent the archival, bibliographic, museological and biological collections that make up the institution’s patrimony. Considering the historical character (some dating from the eighteenth century), evidence, informative, and the scientific and cultural function of these objects, specific metadata will also be set to support both their long-term preservation and the very descriptive metadata attributed to them. It should be noted that although different objects (museum pieces, bibliographic items, archival documents and biological collections) are treated, the proposal of this communication will be to present the methodology applied for the definition of integrative metadata based on the use of the Dublin Core (DC) standard and for the definition of long-term preservation metadata, based on the Preservation Metadata: Implementation Strategies (PREMIS) with the authenticity, reliability and accessibility premises.A partir de la creación de un Grupo de trabajo multidisciplinar para la elaboración de los documentos institucionales normativos “Plan de Preservación Digital” y “Manual de Digitalización” orientados a la implementación de acciones referentes a la preservación digital de los acervos científicos y culturales de la Fundação Oswaldo Cruz (Fiocruz), se observó la necesidad de establecer estándares mínimos de metadatos descriptivos, para integrar y dinamizar el acceso a los distintos objetos digitales que representan los acervos archivísticos, bibliográficos, museológicos y de colecciones biológicas que componen el patrimonio de la institución.
