Editorial: 3D Modelling of Mammalian Embryos and Organs

The main scope of this Special issue was to gain understanding on how tissues and organs are arranged into integrative hierarchical levels of complexity, from the molecular to the morphological organization. To understand the underlying complexity of the relationships among these levels during morphogenesis or in the adult we must learn how to resolve single-cell spatial relationships in the three-dimensional (3D) organisation of tissues, organs and even of the whole organisms.Fil: Garagna, Silvia.

Histone deacetylase inhibitors induce invasion of human melanoma cells in vitro via differential regulation of N-cadherin expression and RhoA activity

Background: Histone deacetylase inhibitors (HDACi) exert multiple cytotoxic actions on cancer cells. Currently, different synthetic HDACi are in clinical use or clinical trials; nevertheless, since both pro-invasive and anti-invasive activities have been described, there is some controversy about the effect of HDACi on melanoma cells. Methods: Matrigel and Collagen invasion assays were performed to evaluate the effect of several HDACi (Butyrate, Trichostatin A, Valproic acid and Vorinostat) on two human melanoma cell line invasion (A375 and HT-144). The expression of N- and E-Cadherin and the activity of the RhoA GTPase were analyzed to elucidate the mechanisms involved in the HDACi activity. Results: HDACi showed a pro-invasive effect on melanoma cells in vitro. This effect was accompanied by an up-regulation of N-cadherin expression and an inhibition of RhoA activity. Moreover, the down-regulation of N-cadherin through blocking antibodies or siRNA abrogated the pro-invasive effect of the HDACi and, additionally, the inhibition of the Rho/ROCK pathway led to an increase of melanoma cell invasion similar to that observed with the HDACi treatments. Conclusion: These results suggest a role of N-cadherin and RhoA in HDACi induced invasion and call into question the suitability of some HDACi as antitumor agents for melanoma patients

With the exception of barley and rice, little is known about the existence of hemoglobins (Hbs) in cereals. This work reports the cloning and analysis of hb genes from maize (Zea mays ssp. mays) and teosinte (Zea mays ssp. parviglumis). Coding sequences of maize and teosinte hb genes (hbm and hbt, respectively) are highly similar to each other and are interrupted by three introns located at identical positions as other plant hb genes. Sequences of predicted Hbm and Hbt proteins are identical. The hydropathic profile of Hbm and Hbt is highly similar to that of rice Hb1, suggesting that Hbm, Hbt and Hb1 have the same tertiary structure and biochemical properties. Expression analysis showed that low levels of Hb transcripts, but considerable levels of Hb proteins exist in maize embryonic organs. No Hb transcripts and proteins were detected in teosinte embryonic organs. Low levels of Hb proteins, but no Hb transcripts, were detected in maize and teosinte vegetative organs. These observations suggest that the regulation of hb genes is different in maize and teosinte embryonic organs, and that the expression of hb genes is down- or up-regulated in maize and teosinte, respectively, from germination to vegetative growing

Espainiako argitalpen zientifiko profesionalen egungo egoera eta dituzten arazo nagusiak aztertzendira artikulu honetan, eta era berean arazo horietarako litezkeen konponbideez hitz egiten da. Interes berezia eskaintzen zaie nazioarteko zientzia hizkuntza bakarraren erabilerari, eskuizkribuen hautaketa egokiarieta adierazle bibliometrikoen erabilera zuzenari. We herein present an evaluation of the present state of professional scientific publications in Spain, and propose solutions for some of the fundamental problems which this enterprise is facing. Particular emphasis is placed on the use of an effective international scientific language, on the importance of rigorous evaluation of papers to be published and on the correct use of bibliometric indicators
