Dans des solutions diluées de mononucléotides (AMP-5’, 8-Br-AMP-5’, cAMP-3’, 5’, cAMP-2’, 3’) la vitesse de relaxation 1/T1 du 31P est la moyenne des vitesses relatives aux diverses espèces Ioniques en présence, pondérée par leurs proportions. L’étude de T1 en fonction de la concentration montre que la relaxation est Indépendante de l’état d’association par liaison H, mais s’accélère lorsqu’interviennent des Interactions par empilement.La position “syn” et “anti” de la base est sans effet notable sur T1 mais sa proximité par rapport au 31P est un facteur de relaxation.Le ralentissement de la relaxation lié à l’augmentation de température est dû à un mécanisme dipôle-dipôle ; la contribution due à l’anisotropie de déplacement chimique, faible à 1,4 T, devient prépondérante à 6,3 T

Le déplacement chimique du 31P observé pour des solutions de monoculéotides (AMP-5’, 8-Br-AMP-5’, cAMP-3’, 5’, cAMP-2’, 3’) est une moyenne pondérée des 5 relatifs aux diverses espèces ioniques. Une méthode d’analyse des valeurs de δ en fonction du pH permet de calculer le 5 de chaque espèce. Les mesures à concentration variable montrent que les variations du déplacement chimique du 31P reflètent l’état d’association par liaison hydrogène ; les mesures à température variable révèlent la variation des proportions des divers conformères, grâce à la dépendance de δ aux angles [math]. Une structure quasi-rigide est mise en évidence pour le monoanion de l’AMP-5’, fortement structuré par liaisons intermoléculaires

Conformational studies of the O-specific polysaccharide of Shigella flexneri 5a and of four related synthetic pentasaccharide fragments using NMR and molecular modeling.

International audienceAs part of a program for the development of synthetic vaccines against the pathogen Shigella flexneri, we used a combination of NMR and molecular modeling methods to study the conformations of the O-specific polysaccharide (O-SP) of S. flexneri 5a and of four related synthetic pentasaccharide fragments. The NMR study, based on the analysis of 1H and 13C chemical shifts, the evaluation of inter-residue distances, and the measurement of one- and three-bond heteronuclear coupling constants, showed that the conformation of one of the four pentasaccharides is similar to that of the native O-SP in solution. Interestingly, inhibition enzyme-linked immunosorbent assay demonstrated that a protective monoclonal antibody specific for S. flexneri 5a has a greater affinity for this pentasaccharide than for the others. We carried out a complete conformational search on the pentasaccharides using the CICADA algorithm interfaced with MM3 force field. We calculated Boltzmann-averaged inter-residue distances and 3JC,H coupling constants for the different conformational families and compared the results with NMR data for all pentasaccharides. Our experimental data are consistent with only one conformational family. We also used molecular modeling data to build models of the O-SP with the molecular builder program POLYS. The models that are in agreement with NMR data adopt right-handed 3-fold helical structures in which the branched glucosyl residue points outwards

Zinc, a novel structural element found in the family of bacterial adenylate kinases

The adk gene from Bacillus stearothermophilus was cloned and overexpressed in Escherichia coli under the control of the lac promoter. The primary structure of B. stearothermophilus adenylate kinase exhibited 76% identity with the enzyme from Bacillus subtilis, 60% identity with the enzyme from Lactococcus lactis, and 42% identity with the enzyme from E. coli. The most striking property of the adenylate kinase from B. stearothermophilus is the presence of a structural zinc atom bound to four cysteines in a zinc finger-like fashion. The ability to coordinate zinc is predicted also for a number of other isoforms of bacterial adenylate kinases. Furthermore, the tightly bound metal ion contributes to the high thermodynamic stability of adenylate kinase from B. stearothermophilus.Peer reviewed: YesNRC publication: Ye
