    Characterizing the Natural System: Toward Sustained, Integrated Coastal Ocean Acidification Observing Networks to Facilitate Resource Management and Decision Support

    Coastal ocean ecosystems have always served human populations they provide food security, livelihoods, coastal protection, and defense. Ocean acidification is a global threat to these ecosystem services, particularly when other local and regional stressors combine with it to jeopardize coastal health. Monitoring efforts call for a coordinated global approach toward sustained, integrated coastal ocean health observing networks to address the region-specific mix of factors while also adhering to global ocean acidification observing network principles to facilitate comparison among regions for increased utility and understanding. Here, we generalize guidelines for scoping and designing regional coastal ocean acidification observing networks and provide examples of existing efforts. While challenging in the early stages of coordinating the design and prioritizing the implementation Of these observing networks, it is essential to actively engage all of the relevant stakeholder groups from the outset, including private industries, public agencies, regulatory bodies, decision makers, and the general public. The long-term sustainability of these critical observing networks will rely on leveraging of resources and the strength of partnerships across the consortium of stakeholders and those implementing coastal ocean health observing networks

    Exoskeleton dissolution with mechanoreceptor damage in larval Dungeness crab related to severity of present-day ocean acidification vertical gradients

    Ocean acidification (OA) along the US West Coast is intensifying faster than observed in the global ocean. This is particularly true in nearshore regions (<200 m) that experience a lower buffering capacity while at the same time providing important habitats for ecologically and economically significant species. While the literature on the effects of OA from laboratory experiments is voluminous, there is little understanding of present-day OA in-situ effects on marine life. Dungeness crab (Metacarcinus magister) is perennially one of the most valuable commercial and recreational fisheries. We focused on establishing OA-related vulnerability of larval crustacean based on mineralogical and elemental carapace to external and internal carapace dissolution by using a combination of different methods ranging from scanning electron microscopy, energy dispersive X-ray spectroscopy, elemental mapping and X-ray diffraction. By integrating carapace features with the chemical observations and biogeochemical model hindcast, we identify the occurrence of external carapace dissolution related to the steepest Ω calcite gradients (∆Ωcal,60) in the water column. Dissolution features are observed across the carapace, pereopods (legs), and around the calcified areas surrounding neuritic canals of mechanoreceptors. The carapace dissolution is the most extensive in the coastal habitats under prolonged (1-month) long exposure, as demonstrated by the use of the model hindcast. Such dissolution has a potential to destabilize mechanoreceptors with important sensory and behavioral functions, a pathway of sensitivity to OA. Carapace dissolution is negatively related to crab larval width, demonstrating a basis for energetic trade-offs. Using a retrospective prediction from a regression models, we estimate an 8.3% increase in external carapace dissolution over the last two decades and identified a set of affected OA-related sublethal pathways to inform future risk assessment studies of Dungeness crabs. -- Keywords : Dungeness crab ; Larval sensitivity ; Global climate change ; Ocean acidification ; Exoskeleton structure ; Dissolution ; Mechanoreceptor damage

    Failure of artesunate-mefloquine combination therapy for uncomplicated Plasmodium falciparum malaria in southern Cambodia

    <p>Abstract</p> <p>Background</p> <p>Resistance to anti-malarial drugs hampers control efforts and increases the risk of morbidity and mortality from malaria. The efficacy of standard therapies for uncomplicated <it>Plasmodium falciparum </it>and <it>Plasmodium vivax </it>malaria was assessed in Chumkiri, Kampot Province, Cambodia.</p> <p>Methods</p> <p>One hundred fifty-one subjects with uncomplicated falciparum malaria received directly observed therapy with 12 mg/kg artesunate (over three days) and 25 mg/kg mefloquine, up to a maximum dose of 600 mg artesunate/1,000 mg mefloquine. One hundred nine subjects with uncomplicated vivax malaria received a total of 25 mg/kg chloroquine, up to a maximum dose of 1,500 mg, over three days. Subjects were followed for 42 days or until recurrent parasitaemia was observed. For <it>P. falciparum </it>infected subjects, PCR genotyping of <it>msp1</it>, <it>msp2</it>, and <it>glurp </it>was used to distinguish treatment failures from new infections. Treatment failure rates at days 28 and 42 were analyzed using both per protocol and Kaplan-Meier survival analysis. Real Time PCR was used to measure the copy number of the <it>pfmdr1 </it>gene and standard 48-hour isotopic hypoxanthine incorporation assays were used to measure IC<sub>50 </sub>for anti-malarial drugs.</p> <p>Results</p> <p>Among <it>P. falciparum </it>infected subjects, 47.0% were still parasitemic on day 2 and 11.3% on day 3. The PCR corrected treatment failure rates determined by survival analysis at 28 and 42 days were 13.1% and 18.8%, respectively. Treatment failure was associated with increased <it>pfmdr1 </it>copy number, higher initial parasitaemia, higher mefloquine IC<sub>50</sub>, and longer time to parasite clearance. One <it>P. falciparum </it>isolate, from a treatment failure, had markedly elevated IC<sub>50 </sub>for both mefloquine (130 nM) and artesunate (6.7 nM). Among <it>P. vivax </it>infected subjects, 42.1% suffered recurrent <it>P. vivax </it>parasitaemia. None acquired new <it>P. falciparum </it>infection.</p> <p>Conclusion</p> <p>The results suggest that artesunate-mefloquine combination therapy is beginning to fail in southern Cambodia and that resistance is not confined to the provinces at the Thai-Cambodian border. It is unclear whether the treatment failures are due solely to mefloquine resistance or to artesunate resistance as well. The findings of delayed clearance times and elevated artesunate IC<sub>50 </sub>suggest that artesunate resistance may be emerging on a background of mefloquine resistance.</p

    Basin-Scale Control on the Phytoplankton Biomass in Lake Victoria, Africa

    The relative bio-optical variability within Lake Victoria was analyzed through the spatio-temporal decomposition of a 1997–2004 dataset of remotely-sensed reflectance ratios in the visible spectral range. Results show a regular seasonal pattern with a phase shift (around 2 months) between the south and north parts of the lake. Interannual trends suggested a teleconnection between the lake dynamics and El-Niño phenomena. Both seasonal and interannual patterns were associated to conditions of light limitation for phytoplankton growth and basin-scale hydrodynamics on phytoplankton access to light. Phytoplankton blooms developed during the periods of lake surface warming and water column stability. The temporal shift apparent in the bio-optical seasonal cycles was related to the differential cooling of the lake surface by southeastern monsoon winds. North-south differences in the exposure to trade winds are supported by the orography of the Eastern Great Rift Valley. The result is that surface layer warming begins in the northern part of the lake while the formation of cool and dense water continues in the southern part. The resulting buoyancy field is sufficient to induce a lake-wide convective circulation and the tilting of the isotherms along the north-south axis. Once surface warming spreads over the whole lake, the phytoplankton bloom dynamics are subjected to the internal seiche derived from the relaxation of thermocline tilting. In 1997–98, El-Niño phenomenon weakened the monsoon wind flow which led to an increase in water column stability and a higher phytoplankton optical signal throughout the lake. This suggests that phytoplankton response to expected climate scenarios will be opposite to that proposed for nutrient-limited great lakes. The present analysis of remotely-sensed bio-optical properties in combination with environmental data provides a novel basin-scale framework for research and management strategies in Lake Victoria

    2011 SOSORT guidelines: Orthopaedic and Rehabilitation treatment of idiopathic scoliosis during growth

    <p>Abstract</p> <p>Background</p> <p>The International Scientific Society on Scoliosis Orthopaedic and Rehabilitation Treatment (SOSORT), that produced its first Guidelines in 2005, felt the need to revise them and increase their scientific quality. The aim is to offer to all professionals and their patients an evidence-based updated review of the actual evidence on conservative treatment of idiopathic scoliosis (CTIS).</p> <p>Methods</p> <p>All types of professionals (specialty physicians, and allied health professionals) engaged in CTIS have been involved together with a methodologist and a patient representative. A review of all the relevant literature and of the existing Guidelines have been performed. Documents, recommendations, and practical approach flow charts have been developed according to a Delphi procedure. A methodological and practical review has been made, and a final Consensus Session was held during the 2011 Barcelona SOSORT Meeting.</p> <p>Results</p> <p>The contents of the document are: methodology; generalities on idiopathic scoliosis; approach to CTIS in different patients, with practical flow-charts; literature review and recommendations on assessment, bracing, physiotherapy, Physiotherapeutic Specific Exercises (PSE) and other CTIS. Sixty-five recommendations have been given, divided in the following topics: Bracing (20 recommendations), PSE to prevent scoliosis progression during growth (8), PSE during brace treatment and surgical therapy (5), Other conservative treatments (3), Respiratory function and exercises (3), Sports activities (6), Assessment (20). No recommendations reached a Strength of Evidence level I; 2 were level II; 7 level III; and 20 level IV; through the Consensus procedure 26 reached level V and 10 level VI. The Strength of Recommendations was Grade A for 13, B for 49 and C for 3; none had grade D.</p> <p>Conclusion</p> <p>These Guidelines have been a big effort of SOSORT to paint the actual situation of CTIS, starting from the evidence, and filling all the gray areas using a scientific method. According to results, it is possible to understand the lack of research in general on CTIS. SOSORT invites researchers to join, and clinicians to develop good research strategies to allow in the future to support or refute these recommendations according to new and stronger evidence.</p

    Pengaruh Waktu Dan Suhu Pengeringan Terhadap Kandungan Pati Pada Pembuatan Bubuk Umbi Talas (Colocasia Esculenta L. Schott) Untuk Bioplastik

    Sektor pertanian merupakan salah satu sektor usaha di Indonesia yang banyak menghasilkan produk makanan untuk kebutuhan pangan masyarakat Indonesia. Hasil produksi pertanian tersebut seringkali dijual di pasaran sebagai bahan mentah, padahal dari variasi bahan-bahan tersebut dapat kita olah menjadi suatu produk baru yang pastinya dapat memiliki harga jual yang lebih tinggi dan menguntungkan. Salah satu bagian organ tanaman yang dapat dimanfaatkan adalah umbi. Kategori tanaman untuk jenis umbi-umbian ini sangat beragam. Salah satunya adalah umbi talas. Talas merupakan tanaman yang mengandung kadar pati tinggi yaitu 80%. Pati yang ada pada umbi talas ini sangat potensial untuk dijadikan bahan pembuatan bioplastik. Penelitian ini dilakukan untuk menguji kadar pati pada bubuk talas yang memiliki variasi suhu dan waktu pada pengeringannya. Metode penelitian ini menggunakan rancangan acak kelompok (RAK), yang terdiri dari dua faktor, yaitu: Faktor I : Waktu pengeringan (T) yang terdiri dari 4 taraf, ix yaitu T1 = 3 jam, T2 = 4 jam, T3 = 5 jam, dan T4 = 6 jam. Faktor II : Suhu Pengeringan (P) yang terdiri dari 3 taraf, yaitu P1 = 40°C, P2 = 50°C, dan P3 = 60°C. Banyaknya kombinasi perlakuan adalah 4 x 3 = 12, dan setiap perlakuan dibuat dalam 2 ulangan, sehingga jumlah keseluruhan sampel adalah 24 sampel. Berdasarkan hasil penelitian waktu dan suhu pengeringan memberikan pengaruh yang nyata terhadap kadar pati yang diperoleh. Semakin tinggi suhu dan waktu pengeringan maka kadar pati bubuk umbi talas akan semakin rendah. Perlakuan yang paling optimal dan terbaik dalam penelitian adalah perlakuan suhu 40oC waktu 4 jam. Pada perlakuan tersebut diperoleh kadar pati yang tinggi yaitu 76,89%, selain itu pada perlakuan tersebut dapat memperoleh rendemen bubuk umbi talas yang tinggi dalam waktu yang singkat
