Työperäinen haitta-ainealtistuminen kiertotaloudessa (HAKiTa)
Kiertotalous on kasvava toimiala, johon liittyvissä työtehtävissä voidaan altistua terveydelle haitallisille aineille. Tutkimuksen tavoitteena oli saada uutta tietoa altistumisesta kiertotalouden työpaikoilla, arvioida altistumiseen liittyviä terveysriskejä ja niin suojella työntekijöiden terveyttä. Tavoitteena oli lisäksi edistää terveydelle vaarallisten altisteiden riskinhallintaa työpaikoilla ja siten varmistaa kierrätettävien jätelajien kestävä käsittely sekä lisätä yritysten toimintaedellytyksiä ja tuottavuutta kiertotaloudessa.
Mittasimme työntekijöiden altistumista kemiallisille ja biologisille haitta-aineille rakennuspurkujätteen, sähköja elektroniikkaromun (SER) ja tekstiilien kierrätyksessä sekä rakennuspurkutyössä. Tutkimuksen kohteena olivat erityisesti sellaiset terveydelle tunnetusti vaaralliset aineet, joiden käyttö on nykyään kiellettyä tai rajoitettua, mutta joita voi esiintyä vanhemmissa kierrätettäväksi tulevissa materiaaleissa. Näitä olivat useat metallit, palonestoaineet, ftalaatit, PFAS-yhdisteet ja PCB-yhdisteet. Mittasimme myös mikrobeja, bakteerien endotoksiineja ja homeiden mykotoksiineja, joiden epäillään voivan aiheuttaa terveysriskejä. Työympäristöissä esiintyviä altisteita tutkimme työhygieenisillä mittauksilla ilmasta ja pinnoille laskeutuneesta pölystä. SER-kierrätykseen liittyen keräsimme myös pyyhintänäytteitä työntekijöiden käsistä. Lisäksi tutkimme työntekijöiden altistumista biomonitoroimalla eli mittaamalla altisteiden pitoisuuksia työntekijöiden virtsa- ja verinäytteistä. Terveysriskejä arvioimme toksikologisella riskinarvioinnilla.
Pääasiassa emme havainneet terveydelle haitallista altistumista. Poikkeuksena oli kuitenkin rakennuspurkujätteen kierrätystyö, jossa epäorgaanisen pölyn, mikrobien ja endotoksiinien pitoisuudet nousivat usein niin korkeiksi, että terveyshaittoja voi esiintyä, jos käytössä ei ole riittäviä riskinhallintakeinoja. Lisäksi havaitsimme SER-kierrätykseen liittyen lyijyaltistumista, jota on syytä pyrkiä minimoimaan työpaikoilla, sillä lyijy on elimistöön kertyvää ja liian suurina pitoisuuksina terveydelle haitallista. Lyijyaltistumista olisikin hyvä seurata SER-kierrätykseen liittyvillä työpaikoilla biomonitoroimalla. Tulosten perusteella työpaikoilla on syytä kiinnittää erityistä huomiota käsistä suun kautta tapahtuvaan lyijyaltistumiseen, joka saattaa olla merkittävä altistumisreitti. Tekstiilien kierrätystyössä ei havaittu terveysriskejä. Aineisto jäi kuitenkin pieneksi ja ainoassa tekstiilikierrätystä tehneessä tutkimuskohteessa oli jo käytössä useita riskinhallintatoimenpiteitä. Lisätutkimusta tarvitaan siis vielä, jotta tekstiilikierrätykseen liittyen voidaan tehdä johtopäätöksiä.
Tutkimus tuotti ajantasaista tietoa altistumisesta suomalaisilla työpaikoilla, joilla tehdään rakennuspurku-, SER-jätteen tai tekstiilien kierrätystä tai rakennuspurkutyötä. Tulosten perusteella laadimme avoimesti saatavilla olevan malliratkaisun Haitta-aineet SER- ja rakennuspurkujätteen kierrätystyössä – terveyshaittojen torjuminen. Sen ohjeilla työpaikat voivat tunnistaa mahdollisesti olennaisia altisteita sekä vähentää niille altistumista ja altistumisen aiheuttamia terveysriskejä

Genotoxicity biomarkers in workers exposed to hexavalent chromium: a study under the European Human Biomonitoring Initiative
No âmbito da Iniciativa Europeia em Biomonitorização Humana (HBM4EU)
realizou-se um estudo ocupacional, envolvendo trabalhadores com potencial
exposição a crómio hexavalente [Cr( VI)], um reconhecido agente
carcinogénico. No presente estudo são apresentados os resultados de
biomarcadores de genotoxicidade, incluindo a análise de lesão no DNA
e de alterações cromossómicas em células sanguíneas. O estudo foi realizado
em vários Países Europeus e abrangeu trabalhadores de diversos
setores industriais e atividades, bem como um grupo de controlo constituído
por trabalhadores administrativos das mesmas empresas (controlo
interno) e de outras não relacionadas com produção/aplicação de Cr( VI)
(controlo externo). Os resultados mostraram níveis de alterações cromossómicas
(ensaio do micronúcleo) e de lesão no DNA (ensaio do cometa)
significativamente aumentados nos trabalhadores expostos comparativamente
aos controlos externos ( p=0,03; p<0,001, respetivamente). Estes
resultados sugerem que mesmo um baixo nível de exposição ao Cr( VI)
representa um risco acrescido para a saúde dos trabalhadores e, principalmente,
para os que realizam cromagem em banho. O grupo controlo
interno apresentou níveis médios de lesões no DNA e nos cromossomas
comparáveis aos do grupo exposto, salientando a relevância de se considerarem
também em risco. O uso de biomarcadores de efeito demonstrou ser crucial para a deteção precoce de efeitos biológicos decorrentes de
baixos níveis de exposição ao Cr( VI), contribuindo para a identificação de
subgrupos em maior risco. O presente estudo vem apoiar a necessidade
de uma reavaliação do limite de exposição ocupacional a Cr( VI), bem
como da implementação de medidas de gestão de risco conducentes a
uma melhor proteção da saúde dos trabalhadores.An occupational study was carried out within the scope of the European
Initiative on Human Biomonitoring (HBM4EU), involving workers
with potential exposure to hexavalent chromium [Cr(VI)], a recognized
carcinogenic agent. In the present study, the results of biomarkers of
genotoxicity are presented, including the analysis of DNA damage and
chromosomal alterations in blood cells. The study was carried out in
several countries and included workers from different industrial sectors
and activities, as well as a control group including administrative
workers from the same companies (internal control) and from others
not related to the production/application of Cr(VI) (external control).
Results showed significantly increased levels of chromosomal alterations
(micronucleus assay) and DNA damage (comet assay) in exposed
workers compared to external controls (p=0.03; p<0.001, respectively). These results suggest that even a low level of exposure to Cr(VI) represents
an increased risk for the health of workers and, mainly, for
those who per form chromium bath plating. The internal control group
displayed DNA and chromosomal damage levels comparable to those
of the exposed group, highlighting the impor tance of also being considered
as potentially at risk. The use of ef fect biomarkers proved to
be crucial for the early detection of biological ef fects resulting from
low levels of exposure to Cr(VI), contributing to the identification of
subgroups at a higher risk. The present study suppor ts the need for a
reassessment of the occupational exposure limit to Cr(VI), as well as
the implementation of risk management measures leading to a better

Nanomateriaalit osana yhteiskuntaa : Kohti turvallista nanoteknologian tulevaisuutta
Katsaus sisältää tietoa nanomateriaalien käytöstä, turvallisuuteen liittyvistä kysymyksistä, sääntelystä, sekä tutkimuksesta Suomessa. Nanomateriaaleissa vähintään yksi niiden ulottuvuus on välillä 1–100 nanometriä. Aineella voi nanokoossa olla kemiallisia, fysikaalisia, sähköisiä ja mekaanisia erityisominaisuuksia. Nanoteknologiaa käytetään tuotteiden ominaisuuksien parantamiseen. Teollisesti tuotettuja nanomateriaaleja käytetään lähes kaikilla teollisuuden aloilla. Ihmistoiminnan seurauksena syntyy myös tahattomasti poltto- ja prosessiperäisiä nanohiukkasia. Nanomateriaalien terveydelle tai ympäristölle aiheuttamia vaikutuksia ei vielä täysin tunneta. Terveys- ja ympäristöriskien arviointi perustuu tietoihin nanomateriaalien vaaraominaisuuksista ja altistumistasoista. Teollisesti tuotetuille nanomateriaaleille on mahdollista altistua valmistuksessa ja käytössä. Altistuminen kuluttajatuotteista on pääsääntöisesti vähäistä. Nanomateriaalien sääntelyssä sovelletaan EU- ja kansallisia säädöksiä, jotka koskevat kemikaaleja, elintarvikkeita tai lääkkeitä. Lisäksi EU:ssa on sektorikohtaisia säädöksiä nanomateriaalien turvalliselle käytölle. Euroopan komissio rahoittaa yhä enemmän nanomateriaalien turvallisuuteen liittyvää tutkimista. Suomen yliopistoissa ja valtion tutkimuslaitoksissa tehdään ansiokasta nanomateriaaleja koskevaa materiaali- ja turvallisuustutkimusta
HBM4EU chromates study - Overall results and recommendations for the biomonitoring of occupational exposure to hexavalent chromium
Exposure to hexavalent chromium [Cr(VI)] may occur in several occupational activities, e.g., welding, Cr(VI) electroplating and other surface treatment processes. The aim of this study was to provide EU relevant data on occupational Cr(VI) exposure to support the regulatory risk assessment and decision-making. In addition, the capability and validity of different biomarkers for the assessment of Cr(VI) exposure were evaluated. The study involved nine European countries and involved 399 workers in different industry sectors with exposures to Cr(VI) such as welding, bath plating, applying or removing paint and other tasks. We also studied 203 controls to establish a background in workers with no direct exposure to Cr(VI). We applied a cross-sectional study design and used chromium in urine as the primary biomonitoring method for Cr(VI) exposure. Additionally, we studied the use of red blood cells (RBC) and exhaled breath condensate (EBC) for biomonitoring of exposure to Cr(VI). Personal measurements were used to study exposure to inhalable and respirable Cr(VI) by personal air sampling. Dermal exposure was studied by taking hand wipe samples. The highest internal exposures were observed in the use of Cr(VI) in electrolytic bath plating. In stainless steel welding the internal Cr exposure was clearly lower when compared to plating activities. We observed a high correlation between chromium urinary levels and air Cr(VI) or dermal total Cr exposure. Urinary chromium showed its value as a first approach for the assessment of total, internal exposure. Correlations between urinary chromium , Cr(VI) in EBC and Cr in RBC were low, probably due to differences in kinetics and indicating that these biomonitoring approaches may not be interchangeable but rather complementary. This study showed that occupational biomonitoring studies can be conducted successfully by multi-national collaboration and provide relevant information to support policy actions aiming to reduce occupational expo-sure to chemicals
Role of Surface Chemistry in the In Vitro Lung Response to Nanofibrillated Cellulose
Wood-derived nanofibrillated cellulose (NFC) has emerged as a sustainable material with a wide range of applications and increasing presence in the market. Surface charges are introduced during the preparation of NFC to facilitate the defibrillation process, which may also alter the toxicological properties of NFC. In the present study, we examined the in vitro toxicity of NFCs with five surface chemistries: nonfunctionalized, carboxymethylated, phosphorylated, sulfoethylated, and hydroxypropyltrimethylammonium-substituted. The NFC samples were characterized for surface functional group density, surface charge, and fiber morphology. Fibril aggregates predominated in the nonfunctionalized NFC, while individual nanofibrils were observed in the functionalized NFCs. Differences in surface group density among the functionalized NFCs were reflected in the fiber thickness of these samples. In human bronchial epithelial (BEAS-2B) cells, all NFCs showed low cytotoxicity (CellTiter-GloVR luminescent cell viability assay) which never exceeded 10% at any exposure time. None of the NFCs induced genotoxic effects, as evaluated by the alkaline comet assay and the cytokinesis-block micronucleus assay. The nonfunctionalized and carboxymethylated NFCs were able to increase intracellular reactive oxygen species (ROS) formation (chloromethyl derivative of 2 ',7 '-dichlorodihydrofluorescein diacetate assay). However, ROS induction did not result in increased DNA or chromosome damage
